Data and Code for Yeager et al., 2022: Enhanced Skill and Signal-to-noise in an Eddy-Resolving Decadal Prediction System

The sensitivity of decadal prediction system performance to model resolution is examined by comparing results from low- and high-resolution (LR and HR) predictions conducted with the Community Earth System Model (CESM). The primary difference between the two systems is the horizontal grid spacing of the ocean and atmosphere models (1° for both in LR; 0.1° and 0.25°, respectively, in HR), permitting a first direct comparison of how skill and signal-to-noise characteristics change when moving to the ocean eddy-resolved modeling regime. HR exhibits significantly increased skill and enhanced signal-to-noise for atmospheric fields compared to LR. This result suggests that mesoscale atmosphere-ocean interaction, which is present in HR but absent in LR, is a key mechanism involved in the transmission of predictable signals from the ocean to the atmosphere. Climate predictions can potentially be improved (and the signal-to-noise paradox alleviated) through explicit representation of ocean eddies and their interactions with the atmosphere.

<p>This data publication includes raw ensemble forecast output (forecasts.zip), as well as summary score files (scores.zip) for process-based forecasts produced with the Forecasting Lake and Reservoir Ecosystems (FLARE) framework. In addition, it includes scores for climatology (climatology_scores.csv) and random walk (RW_scores.csv) null forecasts, formatted observational data of target variables (sunp-targets-insitu.csv), and meteorological driver files required for analysis to accompany the manuscript "Skill of process-based forecasts relative to multiple null models varies across time and depth for water temperature and dissolved oxygen". Forecasts were made of water temperature and dissolved oxygen at Lake Sunapee, NH in 2021 and 2022.</p>

Cholecystokinin facilitates motor skill learning by modulating neuroplasticity in the motor cortex

<p>Cholecystokinin (CCK) is an essential modulator for neuroplasticity in sensory and emotional domains. Here, we investigated the role of CCK in motor learning using a single pellet reaching task in mice. Mice with a knockout of <em>cck</em> gene (CCK<sup>-/-</sup>) or blockade of CCK-B receptor (CCKBR) showed defective motor learning ability; the success rate of retrieving reward remained at the baseline level compared to the wildtype mice with significantly increased success rate. We observed no long-term potentiation (LTP) upon high-frequency stimulation (HFS) in the motor cortex of CCK<sup>-/-</sup> mice, indicating a possible association between motor learning deficiency and neuronal plasticity in the motor cortex. In vivo calcium imaging demonstrated that the deficiency of CCK signalling disrupted the refinement of population neuronal activity in the motor cortex during motor skill training. Anatomical tracing revealed direct projections from CCK-expressing neurons in the rhinal cortex to the motor cortex. Inactivating the CCK neurons in the rhinal cortex using chemogenetic methods significantly suppressed motor learning, and intraperitoneal application of CCK4, a tetrapeptide CCK agonist, rescued the motor learning deficits of CCK<sup>-/-</sup> mice. In summary, our results suggest that CCK, which could be provided from the rhinal cortex, enables neuroplasticity in the motor cortex leading to motor skill learning.</p>

Teachers' knowledge of soft skills and flipped classrooms: nursing and health technologies

<p><strong><span>Background</span></strong><span>: Soft skills and flipped classrooms are important in the fields of education, nursing, and health techniques. </span></p> <p><strong><span>Aim</span></strong><span>: This study determined nursing teachers&rsquo; knowledge of soft skills and flipped classrooms.</span></p> <p><strong><span>Design</span></strong><span>: This is a descriptive cross-sectional study with an analytical focus involving 100 teachers.</span></p> <p><strong><span>Settings</span></strong><span>: The study was conducted at </span><span>a higher institute of nursing professions and health techniques&mdash;ISPITS at Agadir and its annex in Tiznit.</span></p> <p><strong><span>Participants</span></strong><span>:</span><span> One hundred permanent and part-time teachers (with an equal distribution of male and female teachers) at ISPITS in Agadir and its annex in Tiznit participated in the survey.</span></p> <p><strong><span>Methods:</span></strong><span> A five-part</span><span> questionnaire was used to collect data. The independent variable was the use of a flipped classroom and the dependent variable was the development of soft skills. SPSS version 25 software was used to process the data. A Chi-square test, Ficher&rsquo;s test, a binary logistic regression test, and a multivariate logistic regression model were employed for data analysis.</span></p> <p><strong><span>Results</span></strong><span>: In total, 80% of the participants had knowledge of soft skills, and 75% knew about flipped classrooms. The most important soft skills were considered to be communication, stress management, teamwork, conflict management, problem-solving, time management, critical thinking, autonomy, adaptability, and decision-making. The five soft skills considered most likely to be developed by flipped classrooms were autonomy, analysis, teamwork, communication, and time management. Furthermore, the development of soft skills through the flipped classrooms was independently associated with soft skills training (p=0.008) and the use of the flipped classrooms (p=0.042).</span></p> <p><strong><span>Conclusions</span></strong><span>: Teachers have knowledge of soft skills and flipped classrooms despite a lack of training, and flipped classrooms contribute to the development of soft skills</span></p>

Metagenomics education in a modular CURE format positively affects students' scientific discovery perception and data analytical skills

<p>The targeted metagenomics study performed by Pollock et al.&nbsp;<em>(Pollock 2018)</em> was part of the Global Coral Microbiome Project <em>(Vega Thurber Lab 2014)</em>.&nbsp; Since the original publication of Pollock et al. <em>(Pollock 2018) </em>was written for a highly specialized academic public, we provided Bsc. students with a short introduction of the paper and guided them through the metadata table stored in the <em>gcmp16S_map_r25.txt </em>file under the <em>GCMP Australia sequence data, OTU tables, and metadata</em> folder (<a href="https://doi.org/10.6084/m9.figshare.c.3855466.v2">https://doi.org/10.6084/m9.figshare.c.3855466.v2</a>).</p> <p>In order to process the fastq-files and obtain OTU tables, a custom-made pipeline on a Galaxy instance <em>(Galaxy Community)</em> was used. This pipeline contained data analytical tools obtained from the Naturalis Biodiversity Center GitHub environment (<a href="https://github.com/naturalis">https://github.com/naturalis</a>). In order to align with the newest Galaxy best practices, we updated the tools and links to the original as well as the updated GitHub and toolshed versions are included in the references list.</p> <p>The names of the original raw fastq-files contain many dots which can hamper file-type recognition during downstream analyses in Galaxy. Therefore, prior to distributing the fastq-files to the students, these names were renamed using the ManageZIP tool <em>(The BLFS Development Team 1999-2023)</em>. Dots were replaced with underscores, with an exception made for the last 2 file type extension dots. For example, the file name <em>E1.2.Tur.pelt.1.20140814.M_S71_L001_R1_001.fastq.gz</em> was changed into <em>E1_2_Tur_pelt_1_20140814_M_S71_L001_R1_001_1.fastq.gz</em>. Similarly, the filenames in the metadata table were adjusted accordingly. The total collection of renamed file names is available through this zenodo repository as wel as the accompanying metadata file.</p>
