Data for: Comparing the effect of landscape context on vascular plant and bryophyte communities in a human-dominated landscape
<p><b>Aims:</b> It is important to understand the effect of landscape context on biological communities to predict how biodiversity will be affected on human-dominated landscapes. While many studies have tested the effects of landscape context on the species richness and composition of vascular plants, few have compared the responses of vascular plants and bryophytes on the same landscape. We sampled non-epiphytic bryophytes and vascular plants in 184 plots to test whether three landscape context factors measured four years or four decades previously could predict bryophyte or vascular plant species richness and composition after accounting for local factors.</p> <p><b>Location:</b> Temperate forests and oak savannahs, Vancouver Island, British Columbia, Canada.</p> <p><b>Methods: </b>We used model selection and comparisons to test the effects of<b> </b>surrounding road density, total amount of forest, and distance to the nearest forest edge on species richness, species richness of non-disturbance-associated species, and community composition after controlling for important local predictors including substrate availability and topography.</p> <p><b>Results:</b> The species richness of non-disturbance-associated vascular plants was lower in plots with greater surrounding historical road density, and perennial stayer bryophyte richness declined with increasing historical road density and lower historical forest amount, suggesting a potential extinction debt. Landscape context significantly affected total species richness and community composition of vascular plants, but not bryophytes.</p> <p><b>Conclusion:</b> While bryophytes appear to be less sensitive overall to landscape context than vascular plants, disturbance-intolerant perennial stayer bryophytes may decline in the future in response to the increased road density and loss of forest cover that has occurred over the past four decades.</p>
Data from: Comparing the effectiveness of metagenomics and metabarcoding for diet analysis of a leaf-feeding monkey (Pygathrix nemaeus)
Fecal samples are of great value as a non-invasive means to gather information on the genetics, distribution, demography, diet, and parasite infestation of endangered species. Direct shotgun sequencing of fecal DNA could give information on these simultaneously, but this approach is largely untested. Here we use two fecal samples to characterize the diet of two Red-Shanked Doucs Langurs (Pygathrix nemaeus) that were fed a known combination of foliage, fruits, vegetables and cereals. Illumina HiSeq sequencing produced ~70 million paired reads per sample, of which ~10000 (0.014%) and ~44000 (0.066%) respectively corresponded to chloroplast genomes. Sequences were matched against a database of available chloroplast 'barcodes' for angiosperms. The results were compared with 'metabarcoding' using PCR amplification of the P6 loop of trnL. Shotgun sequencing identified 7 and 9 of the likely 16 diet plants, against 6 and 5 plant species identified by metabarcoding. Metabarcoding produced thousands of reads that were consistent with the known diet, but the barcodes were too short to identify several diet plants to genus. Metagenomics could utilize multiple, longer barcodes that combined had greater power of identification, but rare diet items were not recovered. Read numbers for diet species in metagenomic and metabarcoding data were correlated, indicating that both approaches are useful for determining relative sequence abundance. Metagenomic reads were uniformly distributed across the chloroplast genomes; thus if chloroplast genomes were to be used as reference, the precision of identifications and species recovery would improve further. Metagenomics also recovered the host mitochondrial genome and numerous intestinal parasite sequences in addition to generating data useful for characterizing the microbiome.
Data from: A randomised trial comparing the clinical effectiveness of different emergency department healthcare professionals in soft tissue injury management
OBJECTIVE: To evaluate the clinical effectiveness of soft tissue injury management by emergency nurse practitioners (ENPs) and extended scope physiotherapists (ESPs) compared to the routine care provided by doctors in a UK emergency department (ED). DESIGN: Randomised, pragmatic trial of equivalence. SETTING: One adult ED in England. PARTICIPANTS: 372 patients were randomised; 126 to the ESP group, 123 to the ENP group and 123 to the doctor group. Participants were adults (older than 16 years) presenting to the ED with a peripheral soft tissue injury eligible for management by any of the three professional groups. Patients were excluded if they had any of the following: injury greater than 72 hours old; systemic disease; dislocated joints; recent surgery; unable to give informed consent (eg, dementia), open wounds; major deformities; opiate analgesia required; concurrent chest/rib injury; neurovascular deficits and associated fracture. INTERVENTIONS: Patients were randomised to treatment by ESPs, ENPs or routine care provided by doctors (of all grades). MAIN OUTCOME MEASURES: Upper-limb and lower-limb functional scores, quality of life, physical well-being, preference-based health measures and the number of days off work. RESULTS: The clinical outcomes of soft tissue injury treated by ESPs and ENPs in the ED were equivalent to routine care provided by doctors. CONCLUSIONS: As all groups were clinically equivalent it is other factors such as cost, workforce sustainability, service provision and skill mix that become important. This result validates the role of the ENP, which is becoming established as an integral part of minor injuries care, and demonstrates that the ESP should be considered as part of the clinical skill mix without detriment to outcomes.
