Data from: Expression and function of ATP-dependent potassium channels in zebrafish islet β-cells

ATP-sensitive potassium channels (KATP channels) are critical nutrient sensors in many mammalian tissues. In the pancreas, KATP channels are essential for coupling glucose metabolism to insulin secretion. While orthologous genes for many components of metabolism–secretion coupling in mammals are present in lower vertebrates, their expression, functionality and ultimate impact on body glucose homeostasis are unclear. In this paper, we demonstrate that zebrafish islet β-cells express functional KATP channels of similar subunit composition, structure and metabolic sensitivity to their mammalian counterparts. We further show that pharmacological activation of native zebrafish KATP using diazoxide, a specific KATP channel opener, is sufficient to disturb glucose tolerance in adult zebrafish. That β-cell KATP channel expression and function are conserved between zebrafish and mammals illustrates the evolutionary conservation of islet metabolic sensing from fish to humans, and lends relevance to the use of zebrafish to model islet glucose sensing and diseases of membrane excitability such as neonatal diabetes.

Data from: Age-dependent modulation of songbird summer feather molt by temporal and functional constraints

Time constraints influence various ecological, life-history, and demographic properties of individuals and populations of many species throughout the annual cycle. Feather molt is a timely undertaking that is considered among the three most energy-demanding processes in the life cycle of birds. To deal with time pressure, passerines may shorten their molt duration, using three non–mutually exclusive mechanisms: (1) replacing only part of the plumage, (2) increasing the speed of molt, and (3) postponing the renewal of some or all the plumage to a later season (i.e., from the summer to the overwintering period). We used a comparative approach by measuring 12,349 individuals from 134 passerine species to explore how feather molt of juvenile and adult passerines is evolutionarily modulated under time constraints. The results indicate that breeding at northern latitudes and long-distance migration limit the time available for molt and that the consequences of time constraints were age dependent. While the duration of adult summer molt decreased, the extent, rather than the duration, of juvenile molt declined under time constraints. This study highlights the importance of considering time constraints in order to enhance the understanding of selective forces that shape life-history processes and their consequences throughout the annual routine.

Data from: Correlated variability in primate superior colliculus depends on functional class

<p><span>Information read-out from populations of neurons depends strongly on the correlated variability within a population, termed r<sub>SC </sub>(spike count correlations). Traditionally, r<sub>SC</sub> is reported as a single value for a population of neurons comprising a brain area. However, single values, like summary statistics, can obscure underlying features of the constituent elements. We predict that in brain areas consisting of distinct subpopulations of neurons, different subpopulations will exhibit distinct levels of r<sub>SC</sub> not captured by the population r<sub>SC</sub>. We tested this idea by recording from neurons in macaque superior colliculus (SC), a structure that contains several subpopulations (or classes) of neurons based on function. We found that during guided saccade tasks, different classes of neurons exhibited differing degrees of r<sub>SC</sub>, which could not be attributed to variations in behavior. Neurons belonging to the "Delay" class displayed a particularly high r<sub>SC</sub>, especially during the delay epoch of tasks that relied on visual working memory. Our finding that r<sub>SC</sub> depends on functional class, as well as the type of task, demonstrate the importance of taking functional subpopulations into account when attempting to model or infer population coding principles in primate SC, or other areas of the brain that contain subpopulations supporting specific functions.</span></p>

Data for: Effects of biotic interactions on plant fecundity depend on spatial and functional structure of communities and time since disturbance

<p><span>Biotic interactions in plant communities affect individual fitness and community dynamics. Interactions between plants vary in space, over time and with organisational complexity. Yet it is challenging to quantify temporal, spatial and functional determinants of different types of interactions between long-lived perennial plant species and their effect on lifetime fecundity. We studied how plant-plant, pollinator- and seed predator-mediated interactions affect year-to-year variation in three fecundity components (cone production, seed set and seed survival) during post-fire recovery. Age-stratified data on the three fecundity components were collected in 19 even-aged communities comprising 20 serotinous <em>Protea </em>shrub species in the South African Fynbos. We analyse data on these fecundity components with neighbourhood models to infer the sign and strength of interactions throughout post-disturbance recovery, the neighbour plant traits that shape them and the spatial scale at which interactions take place. For each fecundity component, these models describe how neighbourhood effects change over time and with spatial distance between plants. For each focal plant, we then predicted neighbourhood effects on individual fecundity components and cumulative reproductive output at different post-fire stand ages. Competitive effects on cone production and seed set increased with post-fire stand age as biomass and floral resources for pollinators build up. In contrast, neighbourhood effects on seed survival were weak throughout post-disturbance recovery. Plant-plant interactions were shaped by neighbour traits related to resource acquisition, whereas animal-mediated interactions depended on neighbour traits related to resource availability for pollinators and seed predators. The spatial scale of the interactions increased from plant-plant over predator-mediated to pollinator-mediated interactions. The joint effect of these interactions on cumulative reproductive output caused the proportion of focal plants experiencing competition to increase with time since fire. We show that temporal changes in biotic interactions throughout post-disturbance recovery of perennial plant communities depend on functional traits and can be integrated to neighbourhood effects on lifetime fecundity. Studying the temporal, spatial and functional determinants of neighbourhood effects on lifetime fecundity is important for predicting not only individual plant fitness, but also population and community dynamics in changing environments. </span></p>

Silicon enrichment alters functional traits in legumes depending on plant genotype and symbiosis with nitrogen-fixing bacteria

<p>1. Silicon (Si) uptake and deposition (silicification) in tissues is known to alleviate stresses and generally improve plant health. This is mostly studied in Si-high accumulators, such as grasses, with comparatively less known about its effects on other plant functional groups, such as legumes. There is speculation that Si may positively impact the symbiosis between legumes and the nitrogen-fixing bacteria (rhizobia) they associate with, but this is poorly understood. This study examined the effects of Si enrichment on legume species associated with rhizobia and the potential underlying mechanism of Si impacts.</p> <p>2. We conducted a glasshouse experiment with lucerne (<i>Medicago sativa</i>) and barrel medic (<i>M. truncatula</i>) associated with a model rhizobial strain. Six genotypes (three per species) were either supplemented with Si (+Si) or untreated (-Si). We quantified 16 functional traits which could be classified as plant growth, physiology, elemental chemistry, nodule activity and nitrogen fixation.</p> <p>3. The two legume species responded to Si distinctively. For example, Si supplementation increased shoot biomass by more than 10% in lucerne but growth was unaffected in barrel medic. Conversely, nitrogen-fixing enzyme (nitrogenase) activity was promoted by more than 85% in +Si barrel medic plants but not in lucerne. Moreover, Si supplementation of lucerne increased the concentrations of Si in leaves by more than 36% but not in root nodules. Increased foliar concentrations of Si in lucerne was positively associated with increased shoot and root biomass in Sequel and Trifecta genotypes, respectively. Conversely, Si supplementation of barrel medic increased the concentration of Si in root nodules by 29% but not that in foliar tissues. Nitrogenase activity and where silicification occurred, differed between genotypes in barrel medic; nitrogenase activity was correlated with concentrations of Si in root nodules rather than that in foliar tissues in one genotype (Sephi) but the reverse was true in another (Hannaford).</p> <p>4.This study demonstrates that two closely related legume species can respond to Si in distinct ways, depending on plant genotype and symbiosis. These results present the overlooked function of Si in legume-rhizobia interactions, which could potentially enhance productivity of this important group of plants.</p>
