Low-dose biliatresone treatment of pregnant mice causes subclinical biliary disease in their offspring: Evidence for a spectrum of neonatal injury

<p>Biliary atresia is a neonatal disease characterized by damage, inflammation, and fibrosis of the liver and bile ducts and by abnormal bile metabolism. It likely results from a prenatal environmental exposure that spares the mother and affects the fetus. Our aim was to develop a model of fetal injury by exposing pregnant mice to low-dose biliatresone, a plant toxin implicated in biliary atresia in livestock, and then to determine whether there was a hepatobiliary phenotype in their pups.<strong> </strong>Pregnant mice were treated orally with 15 mg/kg/d biliatresone for 2 days. Histology of the liver and bile ducts, serum bile acids, and liver immune cells of pups from treated mothers were analyzed at P5 and P21. Pups had no evidence of histological liver or bile duct injury or fibrosis at either timepoint. In addition, growth was normal. However, serum levels of glycocholic acid were elevated at  P5, suggesting altered bile metabolism, and the serum bile acid profile became increasingly abnormal through P21, with enhanced glycine conjugation of bile acids. There was also immune cell activation observed in the liver at P21. These results suggest that prenatal exposure to low doses of an environmental toxin can cause subclinical disease including liver inflammation and aberrant bile metabolism even in the absence of histological changes. This finding suggests a wide potential spectrum of disease after fetal biliary injury.</p>

Contours of the caregiver experience: Social resources and health behaviors in caregiving partners of persons with a spinal cord injury

<p class="MsoCommentText"><b>Background and rationale:</b><b> </b>Informal caregiving is highly prevalent in Switzerland and in the long-term associated with chronic stress as evidenced by its negative effects on caregivers health and well-being. To date, most of research on informal caregiving has been focused on the elderly. Yet, the evidence from these studies may not be applicable and misinform policy in context of physical disabilities, which may further concerns younger-aged caregivers. Spinal cord injury (SCI) is an exemplary condition for the enforced need of long-term informal care that oftentimes occurs in middle-age. To better understand the mechanisms behind the health-adverse effects of caregiving, a improved understanding of the interplay between social and behavioral factors and the caregiver experience is needed. This project was a follow-up of the longitudinal pro-WELL study (pro-WELL 1), which investigated associations of productive activities and social relationships with health and well-being in persons with SCI and their caregiving partners (SNSF Project 100017_153256/1). Using the follow-up funding by SNF (pro-WELL 2) we further explored the pro-WELL data to foster a  comprehensive understanding of the caregiver experience, also to promote the discussion and transfer of the gained knowledge to stakeholders.</p> <p><b>Objectives:</b> The overall objective of this pro-WELL follow-up study ("pro-WELL 2") was to explore the relationship of social and behavioral factors with the caregiver experience to better understand the causal pathway linking the caregiver experience with health and well-being of the caregiving partners of persons with SCI. The specific aims were to investigate 1) whether socioeconomic conditions shape the caregiver experience, 2) whether social relationships moderate the association between the caregiver experience and health, and 3) whether the caregiver experience has an effect on health behaviors in the caregiving partners of persons with SCI. Using the evidence gained by pro-WELL 1 and 2, we aimed to conduct stakeholder dialogues in order to develop strategies to support burdened caregivers within the network of the Swiss Paraplegic Group.</p> <p><span><b>Methods: </b>Pro-WELL is an observational longitudinal survey with three measurement waves. Data collection was completed in January 2017 and n=133, n=123, and n=119 couples participated at baseline, month 6 and month 12, respectively. The analyses were based on longitudinal dyadic data from the caregiving partners of persons with SCI that completed at least two waves (n=123). We applied multivariable regression modelling and path analysis to explore the research questions.</span></p> <p><span><b>Results: </b>We demonstrated that caregivers with lower socioeconomic status reported a higher burden of care (Tough, Brinkhof, Siegrist, Fekete 2020, Int J Equity Health), provided evidence that social support moderates the association between the caregiver burden and health (Tough, Brinkhof, Fekete. submitted), and showed that the caregiver burden negatively affects specific health behaviors (Tough, Brinkhof, Fekete 2020, Health Psychology Behav Med)</span><span>.  </span></p> <p><span><b>Discussion:</b> This study was among the first to investigate the social and behavioral factors related to the caregiver experience, and to evaluate possible pathways through which the caregiver experience may impact upon health and well-being in middle-aged caregivers of persons with a physical disability. This study identified targets for interventions aimed at improving the caregiver experience with the ultimate goal to contribute to caregivers' health and well-being. The results of the pro-WELL 2 study were used to inform stakeholder dialogues on the current needs and potential actions to improve the situation of informal caregivers in Switzerland. </span></p>

Comparing natural hydrogels to self-assembling peptides in spinal cord injury treatment: a systematic review

<p><strong>Abstract</strong></p> <p><em><strong>Background:</strong></em><em>&nbsp;</em>In many cases, central nervous system (CNS) injury is unchanging due to the absence of neuronal regeneration and repair capabilities.<strong>&nbsp;</strong>In recent years, regenerative medicine, and especially hydrogels, have reached a significant amount of attention for their promising results for the treatment of spinal cord injury (SCI) currently considered permanent. Hydrogels are categorized based on their foundation: synthetic, natural, and combination.&nbsp;The objective of this study was to compare the properties and efficacy of commonly used hydrogels, like collagen, and other natural peptides with synthetic self-assembling peptide hydrogels in the treatment of SCI.&nbsp;</p> <p><em><strong>Methods</strong></em><em>:</em><em>&nbsp;</em>Articles were searched in PubMed, Scopus, Web of Science, and Embase. All studies from 1985 until January 2020 were included in the primary search. Eligible articles were included based on the following criteria: administering hydrogels (both natural and synthetic) for SCI treatment,&nbsp; soley foucsing on spinal cord injury treatment, and published in a peer-reviewd journal. Data surronding xonal regeneration, revascularization, elasticity, drug delivery efficacy, and porosity were extracted.</p> <p><em><strong>Results:</strong></em>&nbsp;A total of 24 articles were included for full-text review and data extraction. There were only one experimental study directly comparing Collagen I (as natural hydorgel) and PEG (as synthetic hydrogels) in an <em>in vitro </em>setting. The included study suggested PEG&rsquo;s cell behavior is more expectable in the injury site, which makes it a more reliable scaffold.</p> <p><em><strong>Conclusions:</strong></em>&nbsp;There is limited research comparing and evaluating both types of natural and self-assembling peptides (SAPs) in the same animal or <em>in vitro</em> study, despite its importance. Although we assume that the remodeling of natural scaffolds may lead to a stable hydrogel, there was not a definitive conclusion that synthetic hydrogels are more beneficial than natural hydrogels in neuronal regeneration.</p>

Experimental comparison of fish mortality and injuries at innovative and conventional small hydropower plants - raw data

<p><span>Resolving the controversy about hydropower is only possible based on reliable data on its ecological effects, particularly fish welfare.</span></p> <p><span>Herein, we propose a comprehensive assessment of conventional and innovative hydropower using a data set of 52 250 fish.</span></p> <p><span>The </span><span>effects of hydropower on fish were most harmful at sites with Kaplan turbines, showing </span><span>£</span> <span>83% </span><span>mortality.</span> <span>Innovative hydropower, often termed 'fish-friendly', caused </span><span>£</span> <span>64% mortality.</span></p> <p><span>Our findings suggested that the runner peripheral speed, number of turbine blades, turbulence and gas oversaturation at turbine outlets were the most important factors.</span></p> <p><span>Synthesis and Applications:</span><span> To reduce the impact of hydropower on fish, site-specific characteristics such as head drop, bypass options and river-specific species composition need to be more intensively considered in optimal turbine technologies and operation modes.</span></p>

Weightlifting injuries in master athletes

<p class="MsoNormal"><strong>Data collection:</strong> A total of 976 Masters weightlifters from Australia, Canada, Europe, and the USA, ages 35 to 88 (51.1% female), completed an online survey in June 2021 that included questions on weightlifting injuries, chronic diseases, sport history, and training practices. </p> <p class="MsoNormal"> </p> <p class="MsoNormal"><strong>Definition of injuries:</strong> <span>Injuries were defined in relation to participation to weightlifting, namely whether training restriction occurred due to acute injuries sustained during weightlifting.</span><span> The correspoding srurvey question was </span><span>"</span><em>Have you ever had training restrictions due to acute injuries sustained during or while performing weightlifting?" </em></p> <p class="MsoNormal"> </p> <p class="MsoNormal"><strong>Purpose of the data collection:</strong></p> <p class="MsoNormal">To quantify acute injuries sustained during weightlifting that result in training restrictions and identify potential risk factors or preventative factors in Master athletes.  To evaluate potentially complex interactions of age, sex, health-related, and training-related predictors of injuries with machine learning algorithms. Ensembles of machine learning algorithms (<span>support vector machines, generalized boosted methods,  random forests, one-layer neural network, and naïve Bayesian methods) were used for classification and prediction and compared to logistic regression models.</span></p>

Colonic contractility recordings following spinal cord injury in mice with dietary fiber interventions

<p>Here, we performed colonic contractility measurements in mice following spinal cord injury, with or without a dietary fiber intervention at 2 weeks post-injury. Provided here are is a .zip file containing the raw ABF measurement files necessary for analysis of the frequency and amplitude of contractions. <span>Breifly, data were acquired using AxoClamp 900A (Axon Instruments). Each tissue segment was recorded for at least 30min in 5min gap-free files. 15min of stable recordings were selected from the middle of each recording session for analysis. Using Clampfit software (Molecular Devices, RRID:SCR_011323), data files were filtered at 300Hz (Bessel 8-pole) and reduced by a factor of 100. Files were concatenated and the baseline was adjusted based on overall slope. Amplitude was calculated by subtracting the minimum value of the whole trace from the value of the peak being assessed.&nbsp;</span></p>

Spatial transcriptomics defines injury specific microenvironments and cellular interactions in kidney regeneration and disease

<p>This dataset contains raw and processed seqFISH data quantifying 1300 genes within single cells from three Acute Kidney Injury (AKI) and three control mice kidneys. The dataset also contains the codebook and probe sequences used to create probe libraries for the seqFISH experiments.</p> <p><strong>Supplementary_data_tables</strong> folder contains the supplementary data tables for the manuscript: Data_1 contains DE gene expression, Data_2 and Data_3 contain the codebook and probe sequence information needed to generate the probe libraries used for the seqFISH experiments. Data_4 contains the probe sequence information needed to generate the serial probes against <em>Vcam1</em> and <em>Havcr1</em></p> <p><strong>AKI_Ctrl_object.rds</strong> - Seurat object generated from seqFISH data for the AKI and healthy control mice as detailed in the manuscript.</p> <p><strong>Counts_raw.csv </strong>contains raw gene counts for all cells in the dataset.</p> <p><strong>coordinates.csv</strong> contains the xy coordinates (in um) for every cell in the dataset.</p> <p><strong>metadata.csv</strong> contains the metadata for each cell in the dataset including sample and cell type allocation as well as Microenvironment (ME) assignment. This file also contains expression data of <em>Vcam1</em> and <em>Havcr1,&nbsp;</em>which were detected using serial probes as detailed in the manuscript.</p>

Spatial transcriptomics defines injury specific microenvironments and cellular interactions in kidney regeneration and disease

<p>Kidney injury disrupts the intricate renal architecture and triggers limited regeneration, and injury-invoked inflammation and fibrosis. Deciphering molecular pathways and cellular interactions driving these processes is challenging due to the complex renal architecture. Here, we apply single cell spatial transcriptomics to examine ischemia-reperfusion injury in the mouse kidney. Spatial transcriptomics reveals injury-specific and spatially-dependent gene expression patterns in distinct cellular microenvironments within the kidney and predicts <em>Clcf1-Crfl1</em> in a molecular interplay between persistently injured proximal tubule cells and neighboring fibroblasts. Immune cell types play a critical role in organ repair. Spatial analysis reveals cellular microenvironments resembling early tertiary lymphoid structures and identifies associated molecular pathways. Collectively, this study supports a focus on molecular interactions in cellular microenvironments to enhance understanding of injury, repair and disease.</p>
