Data and R code used in: Plant geographic distribution influences chemical defenses in native and introduced Plantago lanceolata populations

<p>Plants growing outside their native range may be confronted by new regimes of herbivory, but how this affects plant chemical defense profiles has rarely been studied. Using <em>Plantago lanceolata</em> as a model species, we investigated whether introduced populations show significant differences from native populations in several growth and chemical defense traits. <em>Plantago lanceolata </em>(ribwort plantain) is an herbaceous plant species native to Europe and Western Asia that has been introduced to numerous countries worldwide. We sampled seeds from nine native and ten introduced populations that covered a broad geographic and environmental range and performed a common garden experiment in a greenhouse, in which we infested half of the plants in each population with caterpillars of the generalist herbivore <em>Spodoptera littoralis</em>. We then measured size-related and resource-allocation traits as well as the levels of constitutive and induced chemical defense compounds in roots and shoots of <em>P. lanceolata</em>. When we considered the environmental characteristics of the site of origin, our results revealed that populations from introduced ranges were characterized by an increase of chemical defense compounds without compromising plant biomass. The concentrations of iridoid glycosides and verbascoside, the major anti-herbivore defense compounds of <em>P. lanceolata</em>,<em> </em>were higher in introduced populations than in native populations. In addition, introduced populations exhibited greater rates of herbivore-induced volatile organic compound emission and diversity, and similar chemical diversity based on untargeted analyses of leaf methanol extracts. In general, the geographic origin of the populations had a significant influence on morphological and chemical plant traits, suggesting that <em>P. lanceolata</em> populations are not only adapted to different environments in their native range but also in their introduced range.</p>

The more microplastic types pollute the soil, the stronger the growth suppression of invasive alien and native plants

<p>The ecological consequences of microplastic pollution for plants remain largely unknown, and the few studies that tested the effects usually focused on a single type of microplastic and a single plant species. However, most plants will be exposed to multiple microplastic types simultaneously, and the effects may vary among species.</p> <p>To test the effects of microplastic diversity on plants, we grew single plants of eight invasive and eight native species in pots with substrate polluted with 0, 1, 3 and 6 types of microplastics.</p> <p>We found that the growth suppression by microplastic pollution became stronger with the number of microplastic types the plants were exposed to. This tended to be particularly the case for invasive species, as their biomass advantage over natives diminished with the number of microplastic types. The biomass responses coincided with a positive effect of the number of microplastic types on root allocation and thickness, which was also stronger for invasive than for native species. In addition, the results of hierarchical diversity-interaction models suggest that the negative impact of microplastic diversity on the total biomass of invasive plant species was influenced by both the identities of the microplastic and certain types of microplastic with strong pairwise interactions. In contrast, the effect on native species was determined solely by the microplastic identities.</p> <p><em>Synthesis: </em>Our multi-species study thus shows for the first time that the negative effects of microplastic pollution on plant growth increase with the number of microplastic types. We also found tentative evidence that the negative impacts of microplastic diversity were more pronounced for invasive plants compared to native plants, and that this might be due to differences in the responses of root allocation and thickness.</p>

In press,&nbsp;Ecological Solutions and Evidence</p> <p>&nbsp;</p> <p>Paper abstract:&nbsp;</p> <p><span>Managers tasked with controlling invasive species require effective methods that are quick and easy to use without inflicting extensive nontarget damage, while also being compatible with other scheduled management responsibilities.&nbsp;<em>Lonicera maackii</em> (Amur honeysuckle) is a non-native shrub that has invaded eastern and midwestern North American deciduous forests, altering ecosystem function and reducing biodiversity. </span><span>This study explores prescribed fire and seasonal basal applications of triclopyr ester as control methods and examines the extent of nontarget damage. We used paired-split plots to implement basal bark treatments in different seasons within burned and unburned units, and we tracked individual<em> L. maackii</em> to determine mortality and hyperlocal impacts of management.&nbsp;</span><span>Basal bark treatments killed 98.4% of&nbsp;<em>L. maackii</em> across seasonal timings. Nontarget plant cover immediately declined similarly for all herbicide application seasons, but the early- and late-spring treatments showing signs of recovery within four years. Meanwhile, species richness showed biologically small but statistically different declines across all treatment times. Prescribed fire did not impact <em>L. maackii </em>mortality or interact with herbicide efficacy. </span><span>Basal bark applications of triclopyr are an effective means of&nbsp;<em>L. maackii</em> control regardless of application timing, which allows managers to implement it at their convenience to avoid interfering with other management tasks that have time constraints.</span></p>

Managing multiple threats: Evaluating the efficacy of broad-scale introduced predator management in improving native mammal resilience to fire

<p>Preventing further biodiversity loss requires understanding which processes threaten biodiversity and the effectiveness of management actions in mitigating them. Threatening processes can interact in complex and unexpected ways, but different threats are often managed independently. Here, we develop a conceptual model to identify the conditions needed for management of a single threat to achieve a net conservation benefit in systems with multiple interacting threats, and demonstrate its relevance in a replicated case-study experiment. In Australia, introduced red foxes (<em>Vulpes vulpes</em>) and feral cats (<em>Felis catus</em>) may hunt vulnerable native mammals more effectively after fire, due to loss of understory vegetation. However, the efficacy of broad-scale control of introduced predators in improving native mammal resilience to fire has not been quantified. Moreover, many studies assessing the impacts of prescribed fire on species rely on a much smaller number of independent replicates. Using a natural before-after control-impact experiment with 14 prescribed fires, each &gt; 200 ha, we tested whether existing landscape-scale fox baiting programs influenced the immediate effects of prescribed fire on these two introduced predators and five medium-sized native mammals, including the threatened long-nosed potoroo (<em>Potorous tridactylus</em>) and southern brown bandicoot (<em>Isoodon obesulus</em>). Fox occupancy increased across both treatments post-fire, but baiting reduced the magnitude of increase. In contrast, mean feral cat occupancy remained constant in unbaited areas post-fire, but nearly doubled in fox-controlled areas, possibly due to a mesopredator release. Existing landscape-scale fox control programs did not clearly improve the short-term resilience of native mammals to prescribed fire (at least under the current fire and fox management regimes in our study landscapes). These results likely emphasise the need to integrate fire and predator management strategies for threatened faunal conservation iIn the presence of acute disturbances such as fire, threatened native mammals may require more intensive and integrated management of fire and introduced predators, such as &nbsp;(e.g., through more intensive targeted predator controlbaiting around fire events, or intensive protection usingthrough natural or artificial refuges).</p>

CaM was twice passed through a Bio-Spin P-6 gel filtration spin column (6 kDa cut-off, <i>Bio-Rad</i>), CaD and the myoVa peptide were buffer exchanged through five cycles of tenfold dilution and re-concentration using centrifugal concentrators Vivaspin 500 (10 kDa cut-off, <i>Sartorius</i>) or Amicon Ultra 0.5mL (3 kDa cut-off, <i>Merck/Millipore</i>), respectively. Desalted proteins were introduced into an Orbitrap Q Exactive UHMR mass spectrometer (<i>Thermo Scientific</i>) via static nanoelectrospray ionization from in-house prepared gold-coated borosilicate glass capillaries Kwik-Fil 1B120F-4 (<i>World Precision Instruments</i>). Proteins were sprayed and analysed at 8.5 µM concentration in ammonium acetate alone or supplemented with 200 µM calcium acetate and 100 µM magnesium acetate (both for trace metal analysis, <i>Sigma-Aldrich</i>). For interaction analysis, CaM and/or caldendrin were mixed with myoVa peptide which had final concentration of 8.5 µM (low concentration) or 34 µM (high concentration). The mass spectrometer was tuned for best signal quality and intensity, keeping ion activation and unfolding minimal. Namely, electrospray voltage was kept at 1.3 kV, source desolvation temperature 250°C, in-source desolvation -50 V, ion transfer profile "high m/z", analyzer profile "low m/z", analyzer target resolution 12500 acquiring in mass range 500 – 9000 m/z. Nitrogen was used as collision gas in HCD cell at relative gas pressure setting 7.0 with gentle collisional activation by 10 V HCD voltage gradient.</p><p><strong>Data processing:</strong></p><p>Raw spectra were averaged over at least 50 scans for mass deconvolution and peak assignment in UniDec 4.4.1 package (<i>Marty et al., 2015</i>). The averaged spectra were exported for ZENODO deposition using <i>Thermo Scientific</i> FreeStyle 1.5.93.34 as single-scan Thermo .raw files (including instrumental parameters metadata) as well as in plain m/z vs intensity .txt files.</p>
