Salt marsh ecosystems serve as critical nutrient filters by removing reactive nitrogen (N) through denitrification. We examined the influence of long-term fertilization on N transformation and removal in a salt marsh tidal creek ecosystem fringing the Plum Island Sound estuary in northern Massachusetts, USA. Sediment oxygen demand was within the range of other marsh systems (1271.9 to 7855.0 µmol m-2 h-1) and was not significantly different between the fertilized and reference creek. Net N2 fluxes ranged from net N fixation of -402.7 µmol N2-N m-2 h-1 in the reference creek to net denitrification of 524.9 µmol N2-N m-2 h-1 in the fertilized creek. Net N2 flux and nitrate uptake were significantly higher in the fertilized creek, and in both creeks, net denitrification appeared to be nitrate limited. We calculated rates of dissimilatory nitrate reduction to ammonium (DNRA) and found it to be significantly higher in the fertilized creek, representing 45 and 11% of the total nitrate uptake in the fertilized and reference creeks, respectively. Additionally, there was a strong relationship between ammonium and nitrite fluxes in both creeks. These results suggest that DNRA may outcompete denitrification at high nitrate concentrations. Increased anthropogenic nutrient loading may therefore have a detrimental effect on the N removal capacity of salt marsh ecosystems. Tidal flats are critical components of coastal estuarine ecosystems characterized by high rates of benthic primary productivity and biogeochemical cycling. In order to investigate the impact of anthropogenic nutrient loading on tidal flat biogeochemistry we carried out a two-week fertilization experiment. Throughout the course of the study we conducted two light-dark, whole-core incubations and took measurements of three indicators of microphytobenthos activity in addition to quantifying the resident eastern mud snail (Ilyanassa obsoleta) population.

<p>This project describes a drug discovery plan for Classic Galactosemia, a rare disorder of galactose metabolism where there is currently no disease-transforming therapy. This TEP provides the protein reagents, biophysical assays, and structural information to facilitate two aspects of galactosemia research: (i) to understand the molecular basis of the disease due to defective GALT enzyme, and (ii) to provide chemical starting points to explore metabolic intervention of the upstream enzyme GALK1 aimed at mitigating the GALT defect (substrate reduction).</p>
Improving the understanding of phosphate fertilization in soil P content, acid phosphatase and production of sugarcane
<p>The sugarcane crop is grown in large areas and has a productive chain of billions of dollars. In its chain, they are involved from sugar production to second generation hydrated alcohol. Phosphate nutrition is of great importance for culture, therefore requiring further studies. We sought to evaluate sources and doses of P in the absence/presence of filter cake. The first three models refer to P<sub>2</sub>O<sub>5</sub> sources and doses in the absence of the filter cake and the last three models refer to P<sub>2</sub>O<sub>5</sub> sources and doses in the presence of the filter cake (7.5 Mg ha<sup>-1</sup>, in dry weight). As a statistical basis, we used average, median, maximum, minimum and standard deviation data for the variables evaluated.</p>
Data from: Safety of single low-dose primaquine in glucose-6-phosphate dehydrogenase deficient falciparum-infected African males: two open-label, randomized, safety trials
Background: Primaquine (PQ) actively clears mature Plasmodium falciparum gametocytes but in glucose-6-phosphate dehydrogenase deficient (G6PDd) individuals can cause hemolysis. We assessed the safety of low-dose PQ in combination with artemether-lumefantrine (AL) or dihydroartemisinin-piperaquine (DP) in G6PDd African males with asymptomatic P. falciparum malaria. Methods and findings: In Burkina Faso, G6PDd adult males were randomized to treatment with AL alone (n = 10) or with PQ at 0.25 (n = 20) or 0.40 mg/kg (n = 20) dosage; G6PD-normal males received AL plus 0.25 (n = 10) or 0.40 mg/kg (n = 10) PQ. In The Gambia, G6PDd adult males and boys received DP alone (n = 10) or with 0.25 mg/kg PQ (n = 20); G6PD-normal males received DP plus 0.25 (n = 10) or 0.40 mg/kg (n = 10) PQ. The primary study endpoint was change in hemoglobin concentration during the 28-day follow-up. Cytochrome P-450 isoenzyme 2D6 (CYP2D6) metabolizer status, gametocyte carriage, haptoglobin, lactate dehydrogenase levels and reticulocyte counts were also determined. In Burkina Faso, the mean maximum absolute change in hemoglobin was -2.13 g/dL (95% confidence interval [CI], -2.78, -1.49) in G6PDd individuals randomized to 0.25 PQ mg/kg and -2.29 g/dL (95% CI, -2.79, -1.79) in those receiving 0.40 PQ mg/kg. In The Gambia, the mean maximum absolute change in hemoglobin concentration was -1.83 g/dL (95% CI, -2.19, -1.47) in G6PDd individuals receiving 0.25 PQ mg/kg. After adjustment for baseline concentrations, hemoglobin reductions in G6PDd individuals in Burkina Faso were more pronounced compared to those in G6PD-normal individuals receiving the same PQ doses (P = 0.062 and P = 0.022, respectively). Hemoglobin levels normalized during follow-up. Abnormal haptoglobin and lactate dehydrogenase levels provided additional evidence of mild transient hemolysis post-PQ. Conclusions: Single low-dose PQ in combination with AL and DP was associated with mild and transient reductions in hemoglobin. None of the study participants developed moderate or severe anemia; there were no severe adverse events. This indicates that single low-dose PQ is safe in G6PDd African males when used with artemisinin-based combination therapy.

<p>Under depleted external phosphate (Pi), many plant species adapt to this stress by initiating downstream signalling cascades. In plants, the vascular system delivers nutrients and signalling agents to control physiological and developmental processes. Currently, limited information is available regarding the direct role of phloem-borne long-distance signals in plant growth and development under Pi-stress conditions. Here, we report on the identification and characterization of a cucumber protein, <em>Cucumis sativus</em> Phloem Phosphate-Stress-Repressed 1 (CsPPSR1), whose level in the phloem translocation stream rapidly responds to imposed Pi-limiting conditions. CsPPSR1 degradation is mediated by the 26S proteasome; under Pi-sufficient conditions, CsPPSR1 is stabilized by its phosphorylation, within the sieve tube system, through the action of CsPPSR1 Kinase. Further, we discovered that CsPPSR1 Kinase was susceptible to Pi-starvation-induced degradation, in the sieve tube system. Our findings offer insight into a molecular mechanism underlying the response of phloem-borne proteins to Pi-limited stress conditions.</p>

Data from: Experimental analysis of organ decay and pH gradients within a carcass and the implications for phosphatization of soft tissues
<p>Replacement of soft-tissues by calcium phosphate yields spectacular fossils. Decay experiments have shown that pH is a major control on the precipitation of calcium phosphate and tissue replication: for this to occur pH must fall below the carbonic acid dissociation constant (pH 6.38). However, in the fossil record, phosphatisation is highly selective - some internal organs, such as muscles, stomachs, and intestines, appear to preferentially phosphatise while other organs seldomly phosphatise. The reasons for this are unclear but one hypothesis is that, during decay, organs create distinct chemical microenvironments and only some fall below the critical pH threshold for mineralization to occur. Here, we present a novel investigation using microelectrodes that records fluctuating dynamic spatial and temporal pH gradients inside of organs within a carcass in real time. Our experiments demonstrate that within a decaying carcass, organ-specific microenvironments are not generated. Rather, a pervasive pH environment forms within the body cavity (i.e. the coelom) which persists until integumentary failure. With no evidence to support the development of organ-specific microenvironments during decay other factors must control organ phosphatisation. We propose it is tissue histology that plays an important role in selective phosphatisation. Tissues with high phosphate content (and those rich in collagen) are most likely to phosphatise. Internal organs that have low tissue-bound phosphate, including the integuments of the stomach and intestine, only phosphatise when associated with ingested phosphate-rich organic matter. Identifying the driver behind selective phosphatisation may provide insights into other highly selective modes of soft-tissue preservation i.e. pyritization.</p>
The oxygen isotope of phosphate in the East China Sea in 2020
<p>For the analysis of the main sources of phosphate in the ESC during summer, we choose the area between 120.93 ˚ E-125.9 ˚ E and 26.08˚ N-32.35 ˚ N as research sites . The samples were all from the voyage of the research ship (Xiangyanghong 18). Surface seawater was collected at 49 stations for δ<sup>18</sup>O<sub>p</sub> analysis by a conductivity-temperature-depth profiler. Then we utilized δ<sup>18</sup>O<sub>p</sub> to trace the source of phosphate in the ESC. </p> <p>The δ<sup>18</sup>O<sub>p</sub> values in seawater are affected by water masses with different phosphate concentrations.We need the two-component mixing model for the analysis of water masses that control phosphate.In this research, atmospheric deposition and adjacent sea transportation were used as terrestrial and seawater end-members respectively to construct the δ<sup>18</sup>O<sub>p</sub> end-member mixing model.</p> <p>In order to quantify the source of phosphate in the sea, a Bayesian isotope mixing model run in the R software package (Stable Isotope Analysis in R, SIAR) was selected for analysis.</p>
The time course of acclimation to the stress of triose phosphate use limitation
<p>Triose-phosphate utilization (TPU) limits the maximum rate at which plants can photosynthesize. However, TPU is almost never found to be limiting photosynthesis under ambient conditions for plants. This, along with previous results showing adaptability of TPU at low temperature, suggest that TPU capacity is regulated to be just above the photosynthetic rate achievable under the prevailing conditions. A set of experiments were performed to study the adaptability of TPU capacity when plants are acclimated to elevated CO<sub>2</sub> concentrations. Plants held at 1500 ppm CO<sub>2</sub> were initially TPU-limited. After 30 hours, they no longer exhibited TPU limitations, but they did not elevate their TPU capacity. Instead, the maximum rates of carboxylation and electron transport declined. A time course of regulatory responses was established. A step increase of CO<sub>2</sub> first caused PSI to be oxidized, but after 40 s, both PSI and PSII had excess electrons as a result of acceptor-side limitations. Electron flow to PSI slowed, and the proton motive force increased. Eventually, non-photochemical quenching reduced electron flow sufficiently to balance the TPU limitation. Over several minutes rubisco deactivated contributing to regulation of metabolism to overcome the TPU limitation.</p>
