Meta-Analysis of the transcriptomic profile of human placenta
<p>The establishment of a complex multi-scale model of biological tissue is of great significance for the study of related diseases, and the integration of relevant quantitative data is the premise to achieve this goal. Whereas, the systematic collation of data sets related to placental tissue is relatively lacking. In this study, 18 published transcriptomes (a total of 425 samples) datasets of human pregnancy-related tissues (including chorionic villus and decidua, term placenta, endometrium, in vitro cell lines, etc.) from public databases were collected and analyzed. We compared the most widely used dimensionality reduction (DR) methods to generate a 2D-map for visualization of these data. We also compared the effects of different parameter settings and commonly used manifold learning methods on the results. The result indicates that the nonlinear method can better preserve the small differences between different subtypes of placental tissue than the linear method. It led the foundation for the study on accurate computational modeling of placental tissue development in the future. The datasets and analysis provide a useful source for the researchers in the field of the maternal-fetal interface and the establishment of pregnancy.</p>
Data from: The effect of funding sources on donepezil randomised controlled trial outcome: a meta-analysis
Objective: To investigate whether there is a difference in the treatment effect of donepezil on cognition in Alzheimer disease between industry-funded and independent randomised controlled trials. Design: Fixed effects meta-analysis of standardised effects of donepezil on cognition as measured by the Mini Mental State Examination and the Alzheimer's Disease Assessment Scale-cognitive subscale. Data sources: Studies included in the meta-analyses reported in the National Institute for Health and Care Excellence (NICE) technical appraisal 217 updated with new studies through a PubMed search. Eligibility criteria: Inclusion criteria were double-blind, placebo-controlled trials of any length comparing patients diagnosed with probable Alzheimer disease (according to the NINCDS-ADRDA/DSM-III/IV criteria) taking any dosage of donepezil. Studies of combination therapies (eg, donepezil and memantine) were excluded, as were studies that enrolled patients with a diagnosis of Alzheimer disease associated with other disorders (eg, Parkinson's disease and Down's syndrome). Results: Our search strategy identified 14 relevant trials (4 independent) with suitable data. Trials sponsored by pharmaceutical companies reported a larger effect of donepezil on standardised cognitive tests than trials published by independent research groups (standardised mean difference (SMD)=0.46, 95% CI 0.37 to 0.55 vs SMD=0.33, 95% CI 0.18 to 0.48, respectively). This difference remained when only data representing change up to 12 weeks from baseline were analysed (industry SMD=0.44, 95% CI 0.34 to 0.53 vs independent SMD=0.35, 95% CI 0.18 to 0.52). Analysis revealed that the effect of funding as a moderator variable of study heterogeneity was not statistically significant at either time point. Conclusions: The effect size of donepezil on cognition is larger in industry-funded than independent trials and this is not explained by the longer duration of industry-funded trials. The lack of a statistically significant moderator effect may indicate that the differences are due to chance, but may also result from lack of power.
Data from: Genome-wide meta-analysis of sciatica in Finnish population
Sciatica or the sciatic syndrome is a common and often disabling low back disorder in the working-age population. It has a relatively high heritability but poorly understood molecular mechanisms. The Finnish population is a genetic isolate where small founder population and bottleneck events have led to enrichment of certain rare and low frequency variants. We performed here the first genome-wide association (GWAS) and meta-analysis of sciatica. The meta-analysis was conducted across two GWAS covering 291 Finnish sciatica cases and 3671 controls genotyped and imputed at 7.7 million autosomal variants. The most promising loci (p<1x10-6) were replicated in 776 Finnish sciatica patients and 18,489 controls. We identified five intragenic variants, with relatively low frequencies, at two novel loci associated with sciatica at genome-wide significance. These included chr9:14344410:I (rs71321981) at 9p22.3 (NFIB gene; p=1.30x10-8, MAF=0.08) and four variants at 15q21.2: rs145901849, rs80035109, rs190200374 and rs117458827 (MYO5A; p=1.34x10-8, MAF=0.06; p=2.32x10-8, MAF=0.07; p=3.85x10-8, MAF=0.06; p=4.78x10-8, MAF=0.07, respectively). The most significant association in the meta-analysis, a single base insertion rs71321981 within the regulatory region of the transcription factor NFIB, replicated in an independent Finnish population sample (p=0.04). Despite identifying 15q21.2 as a promising locus, we were not able to replicate it. It was differentiated; the lead variants within 15q21.2 were more frequent in Finland (6-7%) than in other European populations (1-2%). Imputation accuracies of the three significantly associated variants (chr9:14344410:I, rs190200374, and rs80035109) were validated by genotyping. In summary, our results suggest a novel locus, 9p22.3 (NFIB), which may be involved in susceptibility to sciatica. In addition, another locus, 15q21.2, emerged as a promising one, but failed to replicate.

Meta-Memetic Optimization: A Supporting Analysis of the GOD Framework
<p>This paper presents a supplementary proof to the GOD (Gravitational Omnipotent Dimensionality) framework, introducing the concept of Meta-Memetic Optimization—a recursive, self-correcting process through which cognitive and knowledge systems refine themselves. By extending the principles of memetic coherence, dissemination entropy, and recursive self-similarity, this analysis offers a new layer of interaction that facilitates the continuous improvement of knowledge propagation and cognitive evolution.</p>
The role of hemostatic agents following dental extractions: a systematic review and meta-analysis
<p class="MsoNormal"><span><strong>Objective: </strong>This study critically evaluated the role of hemostatic agents following dental extractions, and the benefits they add over conventional hemostatic measures.</span></p> <p class="MsoNormal"><span><strong>Materials and methods: </strong>a literature search was conducted, including publications that fulfilled the criteria <span>developed based on the PICOS guidelines; </span><span>Population: <span>patients who receive simple or surgical tooth extractions</span>, </span><span>Intervention: the use of hemostatic agents following dental extraction, </span><span>Control: </span><span>any type of conventional hemostatic measures (i.e. suture, gauze pressure)</span><span>, </span><span>Outcome: </span><span>time to achieve hemostasis following tooth removal, postoperative pain, swelling, or bleeding events, </span><span>and Study design: randomized/controlled clinical trials.</span></span></p> <p class="MsoNormal"><span><strong>Results:</strong> Twenty-eight articles met the inclusion criteria, and 24 were meta-analyzed. Several types of hemostatic agents were used, namely <span>tissue adhesives made from cyanoacrylate or fibrin, tranexamic acid, hemocoagulase</span>, <span>collagen membrane</span>, <span>HemCon dental dressing, epsilon aminocaproic acid</span>, <span>Surgicel</span>, <span>sponges or plugs composed of gelatin</span>, <span>oxidized cellulose</span>, <span>and fibrin</span>. <span>Newer hemostatic agents were also applied, such as Ankaferd blood stopper,</span> <span>and feracrylum</span>. Hemostatic agents demonstrated significantly shorter time to achieve hemostasis and fewer bleeding events in patients on antithrombotic drugs or with bleeding disorders<span> (WMD= -2.12, 95% CI -2.63 to -1.61, P < 0.00001, and RR </span><span>0.61, 95% CI 0.43 to 0.86; P= 0.005, respectively). Furthermore, the use of these agents resulted in significantly less postoperative pain following 3<sup>rd</sup> molar surgery in healthy patients (</span><span>WMD= -</span>0.33<span>, 95% CI </span>-0.65 to -0.02<span>, P = 0.04). Conversely, they did not have any effect on postoperative swelling (WMD= </span>-0.11<span>, 95% CI </span>-0.31 to 0.08<span>, </span>P = 0.26<span>). The time needed to finish the surgical procedure when hemostatic agents were used was shorter but with no significant difference </span>(MD= -78.93, 95% CI -187.35 to 29.49, P = 0.15).</span></p> <p class="MsoNormal"><span><strong>Conclusion: </strong>Included studies differ in their methods of reporting bleeding, pain, and swelling. Thus, the results of this article should be confirmed in future research. Within these limitations, it is concluded that hemostatic agents lead to faster hemostasis and fewer postoperative bleeding incidences, compared to conventional hemostatic measures. Their use is highly recommended following dental extractions in patients on antithrombotic drugs or with bleeding disorders, who are likely to develop postoperative hemorrhage events.</span></p>
