Data from: Convergently evolved toxic secondary metabolites in plants drive the parallel molecular evolution of insect resistance
Natural selection imposed by natural toxins has led to striking levels of convergent evolution at the molecular level. Cardiac glycosides represent a group of plant toxins that block the Na,K-ATPase, a vital membrane protein in animals. Several herbivorous insects have convergently evolved resistant Na,K-ATPases, and in some species, convergent gene duplications have also arisen, likely to cope with pleiotropic costs of resistance. To understand the genetic basis and predictability of these adaptations, we studied five independent lineages of leaf-mining flies (Diptera: Agromyzidae). These flies have colonized host plants in four botanical families that convergently evolved cardiac glycosides of two structural types: cardenolides and bufadienolides. We compared each of six fly species feeding on such plants to a phylogenetically related but nonadapted species. Irrespective of the type of cardiac glycoside in the host plant, five out of six exposed species displayed substitutions in the cardiac glycoside–binding site of the Na,K-ATPase that were previously described in other insect orders; in only one species was the gene duplicated. In vitro assays of nervous tissue extractions confirmed that the substitutions lead to increased resistance of the Na,K-ATPase. Our results demonstrate that target site insensitivity of Na,K-ATPase is a common response to dietary cardiac glycosides leading to highly predictable amino acid changes; nonetheless, convergent evolution of gene duplication for this multifunctional enzyme appears more constrained.
Data from: Investigating the extent of parallelism in morphological and genomic divergence among lake trout ecotypes in Lake Superior
Understanding the emergence of species through the process of ecological speciation is a central question in evolutionary biology which also has implications for conservation and management. Lake Trout (Salvelinus namaycush) is renowned for the occurrence of different ecotypes linked to resource and habitat use throughout North America. We aimed to unravel the fine genetic structure of the four Lake Trout ecotypes in Lake Superior. A total of 486 individuals from four sites were genotyped at 6822 filtered SNPs using RADseq technology. Our results revealed different extent of morphological and genetic differentiation within the different sites. Overall, genetic differentiation was weak but significant and was on average three times higher between sites (Mean FST = 0.016) than between ecotypes within sites (Mean FST = 0.005) indicating higher level of gene flow or a more recent shared ancestor between ecotypes within each site than between populations of the same ecotype. Evidence of divergent selection was also found between ecotypes and/or in association with morphological variation. Outlier loci found in genes related to lipid metabolism and visual acuity were of particular interest in this context of ecotypic divergence. However, we did not find clear indication of parallelism at the genomic level, despite the presence of phenotypic parallelism among some ecotypes from different sampling sites. Overall, the occurrence of different levels of both genomic and phenotypic differentiation between ecotypes within each site with several differentiated loci linked to relevant biological functions support the presence of a continuum of divergence in Lake Trout.

Parallel striations
This is the base of a vessel created by experimental archaeologist Caroline Jeffra. It shows traces of parallel striations, caused by a wire to remove the vessel from the potter's wheel. The original pot was produced with the wheel-coiling method as defined by Roux-Courty (1998), shown below. The original file size is 664 MB, has a resolution of 0.0996 mm (vertex spacing) and contains 2931370 vertices. A 3UPS/S Spherical Parallel Manipulator Designed for Robot Assisted Hand Rehabilitation after Stroke
<p><span>Hand dysfunction is a common symptom in stroke patients. This paper presents a r<span>obot</span>ic device which assists rehabilitation process in order to reduce the need of physical therapy, i.e., a <span>3UPS/S parallel robot</span>ic device is employed for repetitive robot-assisted rehabilitation. Euler angle representation was used to solve the robot's inverse kinematics. The robot's joint space and rotational workspace are determined for two scenarios. In the first scenario, the workspace is obtained considering the actuator’s stroke limitations, while in the second scenario, the workspace is determined by adding a second condition, i.e. the range of motion of the spherical joints. Singularity analysis is performed using geometric algebra approach. The robot was manufactured using additive manufacturing technology. The solution of the inverse kinematic problem is employed to control the robot. The robot can perform full range of motion during wrist ulnar deviation and radial deviation motions, with the exception of limited wrist flexion and extension motions. The robot has singular configurations within its workspace. Although the spherical joints have roles in reducing the workspace, the primary causes are actuator selection, radiuses of base and moving platforms and the length of the central leg. These factors can be considered to improve the workspace. Singularity can be avoided by carefully selecting the rotation of the moving platform about Z-axis and avoiding same leg lengths.</span></p>
