<p><strong>Abstract</strong></p> <p>Managing biofilms on the surface of materials is one of the most important public health challenges. One of the possible approaches could be the treatment of biofilms with cold plasma as a sustainable and effective method to destroy biofilms. The aim of this study was to analyse the efficacy of a new open-source cold plasma device especially adapted for treatment with low power consumption of 35 W. We tested the reduction of bacterial cells in biofilms of <em>E. coli, S. aureus</em> and <em>P. aeruginosa</em> after treatment with different exposure times and distance regimes. The results show great antibacterial potential in the form of distinct zones of inhibition on solid growth media. In addition, analysis of biofilm efficacy shows significant differences between bacterial cells, suggesting that Gramme-positive <em>S. aureus</em> is less susceptible to plasma treatment than Gramme-negative <em>E. coli</em> and <em>P. aeruginosa</em>. The results show that increasing the exposure time and decreasing the distance between the nozzle and the biofilm decreases the number of cells in the biofilm (p < 0.05). However, the combination of close distance and long exposure time resulted in synergistic effects in bacterial reduction. We demonstrated a reduction of up to 6.6 CFU cm<sup>2</sup> for <em>E. coli</em> at a distance of 1 mm and an exposure time of 60 seconds. All these indicate that the new cold plasma jet device can be an important key to ensure surface hygiene and numerous applications in medicine and engineering are possible.</p>

Periodically disturbing biofilms reduces expression of quorum sensing regulated virulence factors in Pseudomonas aeruginosa
<p>The opportunistic pathogen <em>Pseudomonas aeruginosa</em> uses quorum sensing to control the expression of multiple virulence factors. In this dataset, we provide raw data demonstrating that periodically disturbing biofilms composed of <em>P. aeruginosa</em> using a physical force reduces the expression of multiple quorum sensing regulated virulence factors from the three major regulons. This dataset also contains cell density measurements of bacteria both the in biofilm and planktonic state under a variety of conditions that use physical force to manipulate the spatial positioning of bacteria. Finally, the data set contains raw outputs from a network logic model that described the core quorum sensing network. This data is affiliated with the manuscript entitled "Periodically disturbing biofilms reduces expression of quorum sensing regulated virulence factors in <em>Pseudomonas aeruginosa</em>."</p>
Assessment of four in vitro phenotypic biofilm detection methods in relation to antimicrobial resistance in aerobic clinical bacterial isolates
<p><strong>Introduction</strong>: The lack of standardized methods for detecting biofilms continues to pose a challenge to microbiological diagnostics since biofilm-mediated infections induce persistent and recurrent infections in humans that often defy treatment with common antibiotics. This study aimed to evaluate diagnostic parameters of four in vitro phenotypic biofilm detection assays in relation to antimicrobial resistance in aerobic clinical bacterial isolates.</p> <p><strong>Methods</strong>: In this cross-sectional study, bacterial strains from clinical samples were isolated and identified following the standard microbiological guidelines. The antibiotic resistance profile was assessed through the Kirby-Bauer disc diffusion method. Biofilm formation was detected by gold standard tissue culture plate method (TCPM), tube method (TM), Congo red agar (CRA), and modified Congo red agar (MCRA). Statistical analyses were performed using SPSS version 17.0, with a significant association considered at p<0.05.</p> <p><strong>Result</strong>: Among the total isolates (n=226), TCPM detected 140 (61.95%) biofilm producers, with CoNS (9/9) (p<0.001) as the predominant biofilm former. When compared to TCPM, TM (n=119) (p<0.001) showed 90.8% sensitivity and 70.1% specificity, CRA (n=88) (p=0.123) showed 68.2% sensitivity and 42% specificity, and MCRA (n=86) (p=0.442) showed 65.1% sensitivity and 40% specificity. Juxtaposed to CRA, colonies formed on MCRA developed more intense black pigmentation from 24 to 96 hours. There were 77 multi-drug-resistant (MDR)-biofilm formers and 39 extensively drug-resistant (XDR)-biofilm formers, with 100% resistance to ampicillin and ceftazidime, respectively.</p> <p><strong>Conclusion</strong>: It is suggested that TM be used for biofilm detection. Unlike MCRA, black pigmentation in colonies formed on CRA declined with time. MDR- and XDR-biofilm formers were frequent among the clinical isolates.</p>
