Research data of Calcium Imaging after electrical stimulation
<p>For intracellular calcium ions (Ca<sup>2+</sup>) mobilization analysis by Calcium Imaging, a human osteoblast-like cell line MG-63 (ATCC ®, CRL1427™) were used [1-3]. The MG-63 cell line has similar characteristics in terms of morphological behavior, adhesion and signaling properties as primary human osteoblasts [3]. Cells were cultured at 37 °C in a humidified atmosphere (5% CO<sub>2</sub>) in Dulbecco's modified eagle medium (DMEM; Gibco) containing 10% fetal calf serum (FCS; PAA Laboratories) and 1% antibiotic (gentamicin, Ratiopharm GmbH) [1-3]. Cells in the near-confluent state (70-80%) were used for the corresponding <em>in vitro</em> experiments. Therefore, cells were washed with PBS, trypsinized with 0.05% trypsin/0.02% EDTA (PAA) for 5 min and then treated with medium to stop the reaction. After centrifugation, 2x10<sup>6</sup> cells / 2ml were incubated shaking in complete medium at 37°C for a complete independent experiment. For the method, 2.5x10<sup>5</sup> cells were stained per electrical stimulation setup (non-stimulated, 1 V 7.9 Hz, 1 V 20 Hz, 5 V 7.9 Hz or 5 V 20 Hz). First, a wash step was performed with PBS, and after centrifugation, sedimented cells were stained with a membrane-permeable calcium indicator fluo-3/AM (Life Technologies Corporation; 5 μM) in slightly hypotonic 4-(2-hydroxyethyl)- 1-piperazineethanesulfonic acid (HEPES) buffer [1-3] shaking at 37°C. After incubation for 40 min, cells were centrifuged and then resuspended in 250 µl of isotonic HEPES. In a 12-well plate, 750 µl of isotonic HEPES was placed. Then the stained suspended cells were added, and immediately electrical stimulation by using IonOptix chamber was started under the LSM780 (Carl Zeiss). After 5 min of adhesion phase, the global calcium signal of the attaching cells was visualized using the inverted LSM 780 with a C Apochromat 40× water immersion objective. Fluo-3/AM dye was excited with the argon ion laser at 488 nm (emission at 515 nm). A full frame (512 x 512 pixels) at maximum pinhole aperture was evaluated using Zen2011 (black edition) software (Carl Zeiss) and "time series" mode. The first time series with electrical stimulation included 150 cycles each 2 s. After this time series (total electrical stimulation of 10 min), the electrical stimulation was stopped and a second time series without electrical stimulation was started. This second time series comprised 240 cycles every 2 s. After the 90th cycle of these series, fluo-3/AM stained cells were treated with 10 mM adenosine-triphosphate (ATP; SERVA). The Atomic-Level Structure of Zinc-Modified Cementitious Calcium Silicate Hydrate
<p>It has recently been demonstrated that the addition of zinc can enhance the mechanical strength of tri-calcium silicates (C<sub>3</sub>S) upon hydration, but the structure of the main hydration product of cement, calcium silicate hydrate (C-S-H), in zinc-modified formulations remains unresolved. Here we combine <sup>29</sup>Si DNP enhanced solid-state nuclear magnetic resonance (NMR), density functional theory (DFT) based chemical shift computations and molecular dynamics (MD) modelling to determine the atomic-level structure of zinc-modified C-S-H. The structure contains two main new silicon species (Q<sup>(1,Zn)</sup> and Q<sup>(2p,Zn)</sup>) where zinc substitutes Q<sup>(1)</sup> silicon species in dimers and bridging Q<sup>(2b)</sup> silicon sites, respectively. Structures determined as a function of zinc content show that zinc promotes an increase in the dreierketten mean chain lengths.</p> <p>The uploaded folder contains the pertinent CIF files, DFT files, raw NMR data files, raw XRD data files, and LAMMPS input files.</p>
Multisite imaging of neural activity using a genetically encoded calcium sensor in the honey bee
<p><span>Understanding of the neural bases for complex behaviors in Hymenoptera insect species has been limited by a lack of tools that allow measuring neuronal activity simultaneously in different brain regions. </span><span>Here, we developed the first pan-neuronal genetic driver in a Hymenopteran model organism, the honey bee, and expressed the calcium indicator GCaMP6f under the control of the honey bee <em>synapsin</em> promoter. We show that GCaMP6f is widely expressed in the honey bee brain, allowing it to record neural activity from multiple brain regions. </span><span>To assess the power of this tool, we focused on the olfactory system, recording simultaneous responses from the antennal lobe, and from the more poorly investigated lateral horn and mushroom body calyces. Neural responses to 16 distinct odorants demonstrate that odorant quality (chemical structure) and quantity are faithfully encoded in the honey bee antennal lobe. In contrast, odor coding in the lateral horn departs from this simple physico-chemical coding, supporting the role of this structure in coding the biological value of odorants. We further demonstrate robust neural responses to several bee pheromone odorants, key drivers of social behavior, in the lateral horn. Combined, these brain recordings represent the first use of a neurogenetic tool for recording large-scale neural activity in a eusocial insect, and will be of utility in assessing the neural underpinnings of olfactory and other sensory modalities and of social behaviors and cognitive abilities.</span></p>
Hybrid AgNPs/eggshell calcium carbonate particles and their antimicrobial efficiency
<p>In this study, hybrid particles of AgNPs<span>-</span>loaded eggshell calcium carbonate <span>(</span>AgNPs<span>/</span>eCaCO<sub>3</sub><span>)</span> were prepared<em> in situ</em> by precipitating the eggshell in the presence of freshly prepared AgNPs with a particle size of 10<span>-</span>30 nm<span>. </span>The hybrid particles were comparatively precipitated<em> in situ</em> at 25 °C and 35 °C using poly <span>(</span>sodium 4<span>-</span>styrenesulphonate<span>) </span>as a polyelectrolyte<span>. </span>The AgNPs<span>/</span>eCaCO<sub>3</sub> particles prepared at 25 °C had a spherical morphology with a mean diameter of 3<span>.</span>56 mm, and BET surface area of 85.08 m<sup>2</sup>/g. On the other hand, the particles prepared at 35 °C had a broader size distribution with a mean diameter of 3<span>.</span>19 mm, and a BET surface area of 79.25 m<sup>2</sup>/g<span>.</span> AgNPs<span>-</span>loaded commercial calcium carbonate particles<span> (</span>AgNPs<span>/</span>e=CaCO<sub>3</sub><span>) </span>comparatively prepared at 35 °C were perfectly spherical with a mean diameter of 5<span>.</span>61 mm<span>. </span>At preparing temperature of 25 °C, the hybrid particles contain AgNPs of 0<span>.</span>78 wt<span>% </span>for AgNPs<span>/</span>eCaCO<sub>3</sub> and 3<span>.</span>20 wt<span>% </span>for AgNPs<span>/</span>CaCO<sub>3</sub><span>. </span>The AgNPs<span>/</span>eCaCO<sub>3</sub> and AgNPs<span>/</span>CaCO<sub>3</sub> particles exhibited the same efficiency against bacteria extracted from beef with an average inhibition zone diameter of 7–10 mm according to the modified Kirby<span>–</span>Bauer disc diffusion assay depending on their concentration and beef source<span>. </span>Freshly prepared silver colloids showed comparatively poorer antimicrobial efficiency with an inhibition zone diameter around 7 mm<span>. </span></p>
Calcium fortification of water during pregnancy for the prevention of preeclampsia in a low-income setting: a cost-effectiveness analysis
<p>This database contains the model and the parameters of the study. It's is important to highlight that this is the first version of the model, and thus modifications are forthcoming. With the current database, the abstract is the following:</p> <p>Fortification of water with calcium during pregnancy is proposed as a suitable and promising intervention to increase calcium intake and prevent preeclampsia and eclampsia (PE/E). However, the evidence of the cost-effectiveness of this type of intervention is scarce. We conducted a model-based cost-effectiveness analysis to estimate the incremental cost-effectiveness ratio (ICER) of calcium pills and bottled water fortified with calcium, compared to the standard of care (defined as the use of MgSO4 as the standard treatment for PE/E), in the context of Nepal. Outcome measures were years of life gained (YLG) by mothers and newborns and healthcare costs. We considered a lifetime time horizon and health benefits were discounted at 3%. The cost-effectiveness threshold was set at the 2019 Nepal gross domestic product per capita (USD 1071). For calcium pills versus standard of care, ICER was USD 63 per YLG. By switching from calcium pills to bottled water fortified with calcium, the resulting ICER was USD 1702 per YLG. In conclusion, the bottle of water fortified with calcium was found not cost-effective at the defined threshold for Nepal. Complementary economic evidence adapted to other low-and middle income contexts is required to guide novel preventive interventions to improve maternal and child health based on the efficiency principle</p>

Reproductive output and telomere dynamics following calcium supplementation in wild Tree Swallows (Tachycineta bicolor)
<p>This data was the result of an experiment investigating whether biologically relevant differences in reproductive investment influence fitness in a wild bird species. We employed calcium supplementation to experimentally modify reproductive output in Tree Swallow mothers and offspring and we used telomere dynamics during the breeding season as a proxy for fitness. <span>To confirm that calcium supplementation positively impacts reproductive parameters at our study site, we initiated a 4-year trial of calcium supplementation with our Tree Swallow study population. In the following two breeding seasons, we used calcium supplementation to experimentally alter reproductive output and measured telomere length in mothers immediately before egg-laying and shortly before fledging to determine the direct relationship between reproductive investment and telomere shortening in this species. </span>Our study includes one hundred eighty-eight nests initiated and monitored over six breeding seasons. </p>
Data from: Freshwater fishes maintain multi-trait phenotypic stability across an environmental gradient in aqueous calcium
<p>Reductions in a limiting nutrient might be expected to necessitate compromises in the functional traits that depend on that nutrient; yet populations existing in locations with low levels of such nutrients often do not show the expected degradation of functional traits. Indeed, we previously found that fish of three species, Logperch (<em>Percina caprodes</em>),Pumpkinseed Sunfish (<em>Lepomis gibbosus</em>), and Yellow Perch (<em>Perca flavescens</em>) residing in low-calcium water in the Upper St. Lawrence River nevertheless maintained levels of scale calcium comparable to those of conspecific populations in high-calcium water. However, it remains possible that the maintenance of one functional trait under nutrient-limited conditions could come at the expense of maintaining other functional traits that depend on that same nutrient. Hence, we here extend prior work by examining other calcium-dependent traits: skeletal element sizes and bone densities in the same fish species in the same area. Using radiographs of 101 fish from the three species across four locations (two in high-calcium water and two in low-calcium water), we document multi-trait "homeostasis" along the gradient of water calcium. That is, we did not detect any effect of calcium regime (low-calcium versus high-calcium) on any of the measured variables. Further, effect sizes for the skeletal traits were very low – lower even than effect sizes previously documented for scale calcium. Our results thus show that native fishes maintain phenotype stability across a suite of functional traits linked to calcium regulation, perhaps pointing to an "organismal-level homeostasis" scenario rather than a "trait-level homeostasis" scenario. </p>

Calcium imaging data from: Functional organization of visual responses in the octopus optic lobe
<p>Cephalopods are highly visual animals with camera-type eyes, large brains, and a rich repertoire of visually guided behaviors. However, the cephalopod brain evolved independently from that of other highly visual species, such as vertebrates, and therefore the neural circuits that process sensory information are profoundly different. It is largely unknown how their powerful but unique visual system functions, since there have been no direct neural measurements of visual responses in the cephalopod brain. In this study, we used two-photon calcium imaging to record visually evoked responses in the primary visual processing center of the octopus central brain, the optic lobe, to determine how basic features of the visual scene are represented and organized. We found spatially localized receptive fields for light (ON) and dark (OFF) stimuli, which were retinotopically organized across the optic lobe, demonstrating a hallmark of visual system organization shared across many species. Examination of these responses revealed transformations of the visual representation across the layers of the optic lobe, including the emergence of the OFF pathway and increased size selectivity. We also identified asymmetries in the spatial processing of ON and OFF stimuli, which suggest unique circuit mechanisms for form processing that may have evolved to suit the specific demands of processing an underwater visual scene. This study provides insight into the neural processing and functional organization of the octopus visual system, highlighting both shared and unique aspects, and lays a foundation for future studies of the neural circuits that mediate visual processing and behavior in cephalopods.</p>
