Egnazia, Fasano (BR). Il complesso residenziale di età messapica, ricostruzione virtuale e backend scientifico

<p>La ricostruzione virtuale (Altair4 Multimedia in collaborazione con il team dell&#39;Universit&agrave; di Bari guidato da Gianluca Mastrocinque), di questo complesso residenziale indigeno, il primo documentato ad Egnazia con scavo stratigrafico, ha proceduto di pari passo con la prima pubblicazione scientifica, in un proficuo condizionamento reciproco per i diversi obiettivi.<br> Sono stati individuati 5 punti di vista corrispondenti a quelli della domus ad atrio romana, per visualizzare efficacemente la trasformazione, con relativo back-end scientifico: vista dall&rsquo;esterno per il rapporto con il tessuto urbano della pi&ugrave; antica citt&agrave; (V01), cortile centrale (V02), recinto sacro in corrispondenza del sacrario della casa romana (V04), due viste del grande vano artigianale (V05, V06), di cui la prima in corrispondenza della fornace in situ.<br> Particolarmente accurato &egrave; stato il lavoro per la resa di questo vano: in quanto ambiente unico, &egrave; stato necessario inserire due pilastri per ridurre la luce della trave principale, sono state studiate aperture per l&rsquo;areazione e il deflusso dei fumi di combustione, di cui &egrave; stata realizzata una simulazione dinamica per verificare &nbsp;i parametri dimensionali. Sono stati ricostruiti gli arredi per l&rsquo;essiccatura e lo stoccaggio delle ceramiche, tra cui la trozzella, produzione tipica della Messapia conservata nel Museo, di cui &egrave; stato creato un modello fotogrammetrico. Il modello tridimensionale &egrave; stato realizzato con Autodesk 3DSTUDIO MAX, e renderizzato con V-RAY. Sono incluse nel dataset le immagini panoramiche a 360&deg; dello stato attuale dei 5 punti prescelti realizzate con camera INSTA360 PRO2 per le riprese a terra e i rendering a 360&deg; realizzati dai medesimi punti di vista.</p>

Egnazia, Fasano (BR). La piazza messapica e il mercato in erà romana, ricostruzione virtuale e backend scientifico.

<p>La ricostruzione della piazza in et&agrave; messapica &egrave; stata realizzata da Altair4 Multimedia sulla base degli elementi planimetrici (piante di fase) che individuano la superficie dell&rsquo;area, dei resti frammentari in situ dei bordi lastricati, che sono stati estesi a tutto il perimetro per similitudine e dei dati di scavo sistematizzati dal gruppo dell&rsquo;Universit&agrave; di Bari guidato da Gianluca Mastrocinque. Per l&rsquo;et&agrave; romana Altair4 Multimedia si &egrave; basata sul modello ricostruttivo realizzato nel 2015 da Massimo Caggese per Altair srl, sul quale sono state operate alcune modifiche e integrazioni per aggiornarlo allo stato attuale delle conoscenze e delle condizioni di utilizzo, con l&rsquo;inserimento dei banchi di vendita e delle relative merci e suppellettili sulla base dei ritrovamenti, secondo le indicazioni fornite dal team della Universit&agrave; di Bari. Per il periodo messapico &egrave; presente un solo punto di vista, al centro della piazza (V02), per l&rsquo;et&agrave; romana sono stati individuati 6 punti di vista privilegiati, ciascuno pertinente a un particolare aspetto della piazza del mercato e della attivit&agrave; che vi si svolgevano. Sono incluse nel dataset le immagini panoramiche a 360&deg; dello stato attuale realizzate con camera INSTA360 PRO2 per le riprese a terra, e i rendering a 360&deg; realizzati dai medesimi punti di vista realizzati con VRAY su Autodesk 3DS MAX. A ciascuno dei 6 punti di vista panoramici sono associate le immagini di backend con le informazioni relative alle fonti e ai processi interpretativi che hanno supportato l&#39;ipotesi ricostruttiva.</p>

Effects of ACTH4-10Pro8-Gly9-Pro10 on anti-inflammatory cytokine (IL-4, IL-10, IL-13) expression in acute spinal cord injury model (Sprague Dawley rats)

<p class="MsoNormal"><span><strong>Background:</strong> Spinal cord injury (SCI) is a destructive neurological and pathological state that causes major motor, sensory and autonomic dysfunctions. It's final neurological outcome determined from both primary and secondary injury process. Neuroinflammation is a key component of the secondary injury mechanisms with local and systemic consequences. </span>A neuroprotective compound, ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10 </sup>also known as Semax has <span>shown neuroprotective and anti-inflammatory properties. </span>ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10</sup> <span>also has actively used in the treatment of brain ischemia without serious complication reported. Here we analyzed the effects of </span>ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10</sup> in regulating inflammatory cascade in SCI by looking at the expression of anti-inflammatory cytokine IL-4, IL-10, IL-13 in acute compression SCI.</p> <p><span><strong>Method: </strong>We do laminectomy in Sprague Dawley rats at the second thoracic vertebrae. After laminectomy we expose the myelum and create mild SCI model with 20gr and severe SCI with 35gr aneurysm clips. </span>ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10 </sup><span>was administered intranasally to the treatment group and 0,9% NaCl to the control group (placebo). Both group was remain alive and terminated at 3 and 6 hour. </span>The preparations tissue sample were fixed in formalin and examined for immunohistochemistry<span>. Quantitative measurement of anti-inflammatory cytokine (</span>IL-4, IL-10, IL-13) was done in posterior horn with associated anti-monoclonal antibodies.</p> <p> </p> <p><strong>Result:</strong> Rats with mild SCI that were given ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10</sup> shown greater expression of IL-4, IL-10 and IL-13 at three hour post compression but only IL-10 and IL-13 elevated significantly at six hour. Rats with severe compression in ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10</sup> group shown greater expression of IL-10, IL-13 at three hour and IL-4, IL-10 at six hour compared with the placebo group.</p> <p> </p> <p><strong>Conclusion: </strong>Administration of ACTH<sub>4-10</sub>Pro<sup>8</sup>-Gly<sup>9</sup>-Pro<sup>10</sup> intranasal can increase anti inflammatory cytokine expression at Sprague Dawley rat model with mild and severe SCI. Expression of anti inflammatory cytokine was greater in mild compression and early hour (3 hour). Further research needs to be done to determine optimal dose and the actual clinical outcome in vivo.</p>

The target, IL-9R&alpha;, is the receptor involved in the signaling pathway of IL-9, a pro-inflammatory cytokine medically important for its involvement in respiratory diseases. The successive use of modified protocols of ribosome and yeast displays allowed us to combine their strengths - the virtually infinite selection power of ribosome display and the production of (mostly) properly folded and soluble proteins in yeast display. The described experimental protocol is optimized to produce binders highly specific to the target, including selectivity to common proteins such as BSA, and proteins potentially competing for the binder such as receptors of other cytokines. The binders were trained from DNA libraries of two protein scaffolds called 57aBi and 57bBi developed in our laboratory. We show that the described unconventional combination of ribosome and yeast displays is effective in developing selective small protein binders to the medically relevant molecular target.</p>
