Measurements of dissolved inorganic concentrations of nutrient iron and of iron limitation at selected stations and depths from CalCOFI cruises in the California Current System, Nov. 2002 - July 2004 (completed)

Measurements were made of iron (Fe) and the potential for phytoplankton iron limitation on CalCOFI survey cruises (2002-2004) in coastal transition zones of the southern California Current System. This is a weak upwelling regime that is relatively low in nutrients and chlorophyll. The majority of seawater samples from the mixed layer were collected using a pole sampler, others were collected using a trace metal clean Teflon pump system. Profiles for dissolved Fe analysis were collected using GO Flo bottles attached to a synthetic line and the seawater samples were filtered under ultra-pure conditions. Dissolved Fe was measured using an FeLume flow injection analysis system. Changes in phytoplankton (Chla response to Fe+) and nutrient parameters upon iron addition were also investigated.

Chlorophyll and phaeopigments measured from discrete bottle samples from CCE LTER P0904 student cruise in the California Current System, determined by extraction and bench fluorometry, April 2009.

Discrete bottle samples taken from various depths in the California Current System aboard CCE LTER P0904 student cruise. Samples were frozen aboard the ship and taken back to the lab for extraction and analysis. Samples are filtered (known volumes) onto GF/F filters. The filters are placed into culture tubes containing 90% acetone, and the fluorescence of the samples is read on a fluorometer after 24 to 48 hours. The samples are then acidified to degrade the chlorophyll to phaeopigments (non-photosynthetic pigments) and a second reading is taken. The readings prior to and after acidification are used to calculate concentrations of both chlorophyll a and phaeopigments (i.e. phaeophytin).

Data from: Fractal measures of spatial pattern as a heuristic for return rate in vegetative systems

Measurement of population persistence is a long-standing problem in ecology; in particular, whether it is possible to gain insights into persistence without long time-series. Fractal measurements of spatial patterns, such as the Korcak exponent or boundary dimension, have been proposed as indicators of the persistence of underlying dynamics. Here we explore under what conditions a predictive relationship between fractal measures and persistence exists. We combine theoretical arguments with an aerial snapshot and time series from a long-term study of seagrass. For this form of vegetative growth, we find that the expected relationship between the Korcak exponent and persistence is evident at survey sites where the population return rate can be measured. This highlights a limitation of the use of power-law patch-size distributions and other indicators based on spatial snapshots. Moreover, our numeric simulations show that for a single species and a range of environmental conditions that the Korcak–persistence relationship provides a link between temporal dynamics and spatial pattern; however, this relationship is specific to demographic factors, so we cannot use this methodology to compare between species.

Oxygen measurements in closed system respirometry using blubber explants from suckling and fasting grey seal pups

<p>Understanding physiological responses of wildlife and domesticated animals to environmental challenges can be difficult or impossible to investigate at the whole animal level. Tissue culture approaches open up experimental possibilities, but are difficult to undertake in remote environments where facilities to assess tissue viability and minimise or monitor microbial contamination may not be available. Here we used planar optodes in closed system respirometry to measure oxygen use by blubber explants from grey seal (Halichoerus grypus) pups, obtained in a remote field environment in 2017 and processed in a field laboratory with minimal equipment. These data show oxygen measurements at timed intervals and demonstrate that the tissues were respiring aerobically for the 24 h measurement period, compared to empty control vials. We also measured glucose, lactate and glycerol levels in culture media after 24 hours to allow correlation of glucose uptake, lactate accumulation and lipolytic rate, which are also useful markers of metabolic activity, with oxygen use data. Nutritional state of the pups (either suckling or fasting) and tissue depth (closer to muscle (inner) or closer to skin (outer)) were recorded to allow effects of tissue characteristics on metabolic parameters to be examined. We were also able to use the oxygen use profiles in explant-containing and control vials to identify those contaminated by microbes well ahead of any visible colour change in the media stocks. The use of planar optodes for oxygen measurement thus allows simultaneous physiological measurements and contamination monitoring to be undertaken in a minimally equipped field laboratory. Robinson et al. (2021) published in Methods in Ecology and Evolution provides the details of tissue collection and tissue culture. These data will facilitate comparison of metabolic activity of adipose tissue within and between species and assessment of efficacy of monitoring microbial contamination in tissue culture experiments, particularly in field laboratories where other methods may not be logistically possible.</p>

Advanced Laser Fluorescence (ALF) measurements of chlorophyll-a (Chl), phycoerythrin (PE), chromophoric dissolved organic matter (CDOM), and variable fluorescence (Fv/Fm) from CCE process cruises in the California Current System, 2006 - 2012.

The Advanced Laser Fluorometry (ALF) provides spectral deconvolution (SDC) analysis of the laser-stimulated emission (LSE) excited at 405 or 532/510 nm for assessment of chlorophyll a, phycoerythrin, and chromophoric dissolved organic matter. Three spectral types of phycoerythrin are discriminated for characterization of cyanobacteria and cryptophytes in mixed phototrophic populations. The SDC analysis is integrated with measurements of variable fluorescence, Fv/Fm, corrected for the SDC-retrieved background fluorescence, BNC, for improved photophysiological assessments of phytoplankton photochemical efficiency. Fluorescence measurements are normalized to the water Raman scattering to improve the accuracy of measurements. The ALF measurements of discrete water samples from various depths were made using several ALF instrument modifications (ALF-1, ALF-T, Aquatic Laser Fluorescence Analyzer (ALFA) , or Custom Laser Analytical Spectroscopic System (CLASS)) during CCE process cruises.

ATom: Measurements from the Quantum Cascade Laser System (QCLS), Version 2

This dataset provides atmospheric concentrations of CO2, CH4, CO, and N2O measured by the Harvard Quantum Cascade Laser System (QCLS) instruments during airborne campaigns conducted by NASA's Atmospheric Tomography (ATom) mission. The QCLS (DUAL and CO2) instrument package contains two separate optical assemblies and calibration systems, and a common data system and power supply. The QCLS DUAL instrument simultaneously measures CO, CH4, and N2O concentrations, in situ, using two thermoelectrically cooled pulsed-quantum cascade lasers light sources, a multiple pass absorption cell, and two liquid nitrogen-cooled solid-state detectors. The QCLS CO2 instrument measures CO2 concentrations in situ using a thermoelectrically cooled pulsed-quantum cascade laser light source, gas cells, and liquid nitrogen cooled solid-state detectors. The CO2 mixing ratio of air flowing through the sample gas cell is determined by measuring absorption from a single infrared transition line at 4.32 microns relative to a reference gas of known concentration.
