Dissolved organic carbon dynamics in an urban desert stream ecosystem in central Arizona-Phoenix: site locations

Variation in stream chemistry is a function of the strength of terrestrial-aquatic linkages, the extent to which surface and groundwater exchanges, and the rate of instream biotic processes. The importance of these variables may fluctuate as a function of climate regime, catchment geomorphology, or level of human impact. A mechanistic understanding of the influence of each of these variables on ecosystem functioning will increase understanding of the role of streams in global nutrient and carbon cycles. Of particular interest is dissolved organic carbon (DOC), an important source of carbon and energy for microbial processes. Respiration by heterotrophic bacterial communities has recently been linked to the quality (ability of microbes to utilize C source) of the DOC pool in streams. DOC not only influences stream nutrient supply, but also the transport of contaminants and the attenuation of UV radiation. This dissertation focused on DOC delivery to two arid-land stream ecosystems, one native desert (Sycamore Creek, AZ), and one urban (Phoenix, AZ). The overall objectives of this work were to (1) document patterns in DOC quantity and chemical composition in response to flooding and groundwater exchange, (2) generate and test hypotheses explaining variation in DOC quantity and quality and (3) relate this variation to microbial activity. In the native desert stream ecosystem, the climate regime influenced seasonal variation in the quantity and quality of DOC inputs, with higher complexity and higher concentrations of DOC in summer monsoonal runoff. In contrast, human alteration of geomorphology and hydrologic flowpaths in the Phoenix metropolitan area significantly influenced streamwater chemistry in comparison to low-impact streams in the Sonoran Desert. In the city, mechanisms of nutrient retention and transformation were often shifted from dominance by biotic to abiotic ones, severely dampening the influence of climate regime and substituting instead the maintenance

Data from: Growth mode and carbon source impact the surfaceome dynamics of Lactobacillus rhamnosus GG

Bacterial biofilms have clear implications in disease and in food applications involving probiotics. Here, we show that switching the carbohydrate source from glucose to fructose increased the biofilm formation and the total surface-antigenicity of a well-known probiotic, Lactobacillus rhamnosus GG. Surfaceomes (all cell surface-associated proteins) of GG cells grown with glucose and fructose in planktonic and biofilm cultures were identified and compared, which indicated carbohydrate source-dependent variations, especially during biofilm growth. The most distinctive differences under these conditions were detected with several surface adhesins (e.g., MBF, SpaC pilus protein and penicillin-binding proteins), enzymes (glycoside hydrolases, PrsA, PrtP, PrtR and HtrA) and moonlighting proteins (glycolytic, transcription/translation and stress-associated proteins, r-proteins, tRNA synthetases, Clp family proteins, PepC, PepN and PepA). The abundance of several known adhesins and candidate moonlighters, including enzymes acting on casein-derived peptides (ClpP, PepC and PepN), increased in the biofilm cells grown on fructose, from which the surface-associated aminopeptidase activity mediated by PepC and PepN was further confirmed by an enzymatic assay. The mucus binding factor (MBF) was found most abundant in fructose grown biofilm cells whereas SpaC adhesin was identified specifically from planktonic cells growing on fructose. An additional indirect ELISA indicated both growth mode- and carbohydrate-dependent differences in abundance of SpaC, whereas the overall adherence of GG assessed with porcine mucus indicated that the carbon source and the growth mode affected mucus adhesion. The adherence of GG cells to mucus was almost completely inhibited by anti-SpaC antibodies regardless of growth mode and/or carbohydrate source, indicating the key role of the SpaCBA pilus in adherence under the tested conditions. Altogether, our results suggest that carbon source and growth mode coordinate mechanisms shaping the proteinaceous composition of GG cell surface, which potentially contributes to resistance, nutrient acquisition and cell-cell interactions under different conditions. In conclusion, the present study shows that different growth regimes and conditions can have a profound impact on the adherent and antigenic features of GG, thereby providing new information on how to gain additional benefits from this probiotic.

Data from: Strong interactive effects of warming and insect herbivory on soil carbon and nitrogen dynamics at Subarctic tree line

<p>Warming will likely stimulate Arctic primary production, but also soil C and N mineralization, and it remains uncertain whether the Arctic will become a sink or a source for CO<sub>2</sub>. Increasing insect herbivory may also dampen the positive response of plant production and soil C input to warming. We conducted an open-air warming experiment with Subarctic field layer vegetation in North Finland to explore the effects of warming (+3°C) and reduced insect herbivory (67% reduction in leaf damage using an insecticide) on soil C and N dynamics. We found that plant root growth, soil C and N concentrations, microbial biomass C, microbial activity, and soil NH<sub>4</sub><sup>+</sup> availability were increased by both warming and reduced herbivory when applied alone, but not when combined. Soil NO<sub>3</sub><sup>-</sup> availability increased by warming only and in-situ soil respiration by reduced herbivory only. Our results suggest that increasing C input from vegetation under climate warming increases soil C concentration, but also stimulates soil C turnover. On the other hand, it appears that insect herbivores can significantly reduce plant growth. If their abundance increases with warming as predicted, they may curtail the positive effect of warming on soil C concentration. Moreover, our results suggest that temperature and herbivory effects on root growth and soil variables interact strongly, which probably arises from a combination of N demand increasing under lower herbivory and soil mineral N supply increasing under higher temperature. This may further complicate the effects of rising temperatures on Subarctic soil C dynamics.</p>
