Data from: Harsh conditions during early development influence telomere length in an altricial passerine: links with oxidative stress and corticosteroids

Stress during early development can induce substantial long-term effects in organisms. In the case of birds, despite growth compensations, nestlings reared under harsh conditions typically show reduced survival chances in adulthood. It has been proposed that environmental early-life stressors could affect longevity via effects on telomere length, possibly mediated through oxidative stress. However, the link between these processes is not clear. In this study, we experimentally manipulated brood size in spotless starlings (Sturnus unicolor) to test the causal relationship between early stress, oxidative and corticosterone-mediated stress and telomere shortening. Our results show that experimentally enlarged brood sizes led to a reduction in morphometric development on nestlings, the effect being stronger for females than males. Additionally, basal corticosterone levels increased with increasing brood size in female nestlings. Neither plasma antioxidant status nor malondialdehyde levels (a marker of lipid peroxidation) were affected by experimental brood size, although the levels of a key intracellular antioxidant (glutathione) decreased with increasing brood size. We found that the treatment showed a quadratic effect on nestling telomere lengths, that were shortened either by increases or decreases of the original brood size. Our study provides experimental evidence for a link between developmental stress and telomere length, but does not support a direct causal link of this reduction with corticosterone or oxidative stress. We suggest that future studies should focus on how telomere length responds to additional markers of allostatic load.

Short-term responses to high-dose inhaled corticosteroid treatment in patients with chronic obstructive pulmonary disease having a fractional nitric oxide concentration over 35 parts per billion: a single-center pre-post study

<p>Background: There is currently no strategy to identify chronic obstructive pulmonary disease (COPD) patients whose pulmonary function could benefit from inhaled corticosteroids. This study investigates whether a 28-day regime of inhaled corticosteroids improves pulmonary function test results among chronic obstructive pulmonary disease patients having a fractional exhaled nitric oxide concentration &gt; 35 parts per billion.<br> Methods: This was a single-center one-arm pre–post trial. Chronic obstructive pulmonary disease patients with a fractional exhaled nitric oxide concentration &gt; 35 parts per billion treated at our institution from September 2018 to August 2019 were included. Each patient was administered budesonide (200 μg, 8 puffs daily) for 28 days. The primary outcome measure was the difference between the values of forced expiratory volume in 1 s (FEV1) at baseline and 28 days after inhaled corticosteroid initiation. Secondary outcomes included differences of Chronic Obstructive Pulmonary Disease Assessment Test scores, %FEV1, and that between the percent forced vital capacity at baseline and 28 days after inhaled corticosteroid initiation.<br> Results: Twenty patients were included; all patients completed the 28-day inhaled corticosteroid regime. The mean difference in FEV1 between day 1 and 28 days after inhaled corticosteroid initiation was 340 mL (95% confidence interval: −100 to 770 mL; p = 0.122). The mean differences in secondary outcomes were: % forced vital capacity , −0.16% (95% confidence interval: −2.84 to 2.53%; p = 0.905); %FEV1, 1.63% (95% confidence interval: −4.56 to 7.81%; p = 0.589); Chronic Obstructive Pulmonary Disease Assessment Test score, −2.50 (95% confidence interval: −5.72 to 0.72; p = 0.121).<br> Conclusion: The 28-day course of inhaled corticosteroids showed no significant difference in FEV1 for chronic obstructive pulmonary disease patients having fractional exhaled nitric oxide concentration &gt; 35 parts per billion.<br>  </p>

Sex-specific maternal programming of corticosteroid binding globulin by predator odour

<p>Predation is a key organizing force in ecosystems. The threat of predation may act to program the endocrine hypothalamic-pituitary-adrenal axis during development to prepare offspring for the environment they are likely to encounter. Such effects are typically investigated through measurement of corticosteroids (Cort). Corticosteroid-binding globulin (CBG) plays a key role in regulating the bioavailability of Cort, with only free unbound Cort being biologically active. We investigated the effects of prenatal predator odour exposure (POE) in mice on offspring CBG and its impact on Cort dynamics before, during, and after restraint stress in adulthood. POE males, but not females, had significantly higher serum CBG at baseline and during restraint and lower circulating levels of Free Cort. Restraint stress was associated with reduced liver transcript abundance of <i>SerpinA6 </i>(CBG-encoding gene) only in control males. POE did not affect <i>SerpinA6</i> promoter DNA methylation. Our results indicate that prenatal exposure to a natural stressor led to increased CBG levels, decreased percent of Free Cort relative to total, and inhibited restraint stress-induced downregulation of CBG transcription.<i> </i>These changes suggest an adaptive response to a high predator risk environment in males but not females, that could buffer male offspring from chronic Cort exposure.</p>
