Patterns of Testosterone in Male White-tailed Deer (Odocoileus virginianus): Seasonal and Lifetime Variation

<p>Testosterone is strongly associated with the annual development of antlers in cervids, but. endocrine research on wild, freely breeding ungulates is often done without repeated capture of known-aged individuals. As a result, our knowledge on how testosterone fluctuates over the course of a lifetime and variation in lifetime patterns among individuals is limited. We investigated patterns of testosterone in a freely breeding population of white-tailed deer (<i>Odocoileus virginianus</i>) in Alabama, USA that breeds in January. Testosterone peaked during the height of the breeding season, despite this period occurring approximately two months later than in most temperate-region, white-tailed deer populations. Age-related differences in testosterone were only prevalent during the breeding season, with bucks ≥3.5 years old having greater testosterone (853 ng/dl ±96 SE; <i>P</i> = 0.012) than bucks 1.5–2.5 years old (364 ng/dl ±100 SE). Additionally, an individual's testosterone level as a yearling was not positively associated with their lifetime maximum testosterone level (<i>P</i> = 0.583), and an individual's mean testosterone level was positively associated with lifetime testosterone variation (<i>P</i> &lt; 0.001). To our knowledge our study is one of the first to assess how testosterone early in life might relate to individual testosterone later in life. We believe these data provide insight into lifetime hormonal patterns in cervids, and that these patterns may indicate intraspecific variation of lifetime reproductive strategies.</p>

Wind turbines without curtailment produce large numbers of bat fatalities throughout their lifetime: A call against ignorance and neglect

<p>Bats are protected by national and international legislation in European countries, yet many species, particularly migratory aerial insectivores, collide with wind turbines which counteracts conservation efforts. Within the European Union it is legally required<br> to curtail the operation of wind turbines at periods of high bat activity, yet this is not practiced at old wind turbines. Based on data from the national carcass repository in Germany and from our own carcass searches at a wind park with three turbines west<br> of Berlin, we evaluated the magnitude of bat casualties at old, potentially mal-sited wind turbines operating without curtailment. We report 88 documented bat carcasses collected by various searchers over the 20-year operation period of this wind park from<br> 2001 to 2021. Common noctule bats ( Nyctalus noctula ) and common pipistrelles (Pipistrellus pipistrellus ) were most often found dead at these turbines. Our search campaign in August and September 2021 yielded a total of 18 carcasses. We estimated that at least 209 bats were likely killed during our field survey, yielding more than 70 casualties/wind turbine or 39 casualties/MW in two months. Since our campaign covered only part of the migration season, we consider this value as an underestimate. The 20-year period of the wind park emphasises the substantial impact old turbines may have on bat individuals and populations when operating without curtailments. We call for reconsidering the operation procedures of old wind turbines to stop the continuous loss of bats in Germany and other countries where turbine curtailments are even less practiced than in Germany.</p>

Lifetime reproductive output and life-history traits of lizards

<p><span><span><span><span><span><span><span><span><span><span><span><span><b>Aim: </b>Latitudinal gradients in life-history traits are apparent in many taxa and are expected to be strong for ectotherms that have temperature-driven constraints on performance and fitness. The strength of these gradients, however, should also be affected by diet. Diet type, however, influences latitudinal patterns of individual life-history traits. Carnivores exhibit a shift towards "slower-paced" life-histories at higher latitudes for most traits (increased longevity, age at maturity, reproductive lifespan, and decreased clutch frequency). By contrast, herbivores either display "faster-paced" life-histories (reduction in reproductive lifespan, hatchling mass, female mass) or no change (clutch frequency, clutch size, age at maturity) at higher latitudes. Omnivores exhibit intermediate and muted latitudinal patterns.</span></span></span></span></span></span></span></span></span></span></span></span></p> <p><span><span><span><span><span><span><span><span><span><span><span><span><b>Main conclusions: </b>We suggest that the nutritional challenges of herbivory, compounded by thermal constraints at higher latitudes, may explain differences in life-history characteristics of herbivorous ectotherms. Intermediate patterns exhibited by omnivores highlight how flexibility in diet can buffer environmental challenges at higher latitudes. Our results indicate that lizards with different diet types display various trends in their life-histories across latitudes, which eventually balance out to result in similar reproductive outputs throughout their lifetime, with little benefits to carnivory.</span></span></span></span></span></span></span></span></span></span></span></span></p>

Ageing as early-life inertia: disentangling life-history trade-offs along a lifetime of an individual

<p>The theory that ageing evolves because of competitive resource allocation between the soma and the germline has been challenged by studies showing that somatic maintenance can be improved without impairing reproduction. However, it has been suggested that cost-free improvement in somatic maintenance is possible only under a narrow range of benign conditions. Here we show that experimental downregulation of insulin/IGF-1 signalling (IIS) in C. elegans nematodes, a robustly reproducible lifespan and healthspan-extending treatment, reduces fitness in a complex variable environment when initiated during development but does not reduce fitness when initiated in adulthood. Thus, our results show that the costs and benefits of reduced IIS can be uncoupled when organisms inhabit variable environments, and, therefore, do not provide support for the resource allocation theory. Our findings support the theory that the force of natural selection on gene expression in evolutionarily conserved signalling pathways that shape life-history traits declines after the onset of reproduction resulting in organismal senescence.</p>

Simultaneous NAD(P)H and FAD fluorescence lifetime microscopy of long UVA–induced metabolic stress in reconstructed human skin

<p>Solar ultraviolet longwave UVA1 exposure of human skin has short-term consequences at cellular and molecular level, leading at long-term to photoaging. Following exposure, reactive oxygen species (ROS) are generated, inducing oxidative stress that might impair cellular metabolic activity. However, the dynamic of UVA1 impact on cellular metabolism remains unknown because of lacking adequate live imaging techniques. Here we assess the UVA1-induced metabolic stress response in reconstructed human skin with multicolor two-photon fluorescence lifetime microscopy (FLIM). Simultaneous imaging of nicotinamide adenine dinucleotide (NAD(P)H) and flavin adenine dinucleotide (FAD) by wavelength mixing allows quantifying cellular metabolism in function of NAD(P)+/NAD(P)H and FAD/FADH2&nbsp;redox ratios. After UVA1 exposure, we observe an increase of fraction of bound NAD(P)H and decrease of fraction of bound FAD indicating a metabolic switch from glycolysis to oxidative phosphorylation or oxidative stress possibly correlated to ROS generation. NAD(P)H and FAD biomarkers have unique temporal dynamic and sensitivity to skin cell types and UVA1 dose. While the FAD biomarker is UVA1 dose-dependent in keratinocytes, the NAD(P)H biomarker shows no dose dependence in keratinocytes, but is directly affected after exposure in fibroblasts, thus reflecting different skin cells sensitivities to oxidative stress. Finally, we show that a sunscreen including a UVA1 filter prevents UVA1 metabolic stress response from occurring.</p>
