Aggregated variant data from whole-genome sequenced tinnitus patients (TIGER)
<p>Aggregated variant data obtained from tinnitus patients from Sweden.</p> <p>Uploaded datasets are storage in annotated csv files. Annotation was performed using VEP (v106), including population frequencies for each variant from gnomAD, non-finnish Europeans from gnomAD, and swedish population from SweGen project. Pathogenicity scores from CADD are also annotated for each variant. C5a and C5aR1 are key drivers of microvascular platelet aggregation in clinical entities spanning from aHUS to COVID-19
<p>Unrestrained activation of the complement system till the terminal products, C5a and C5b-9, plays a pathogenetic role in acute and chronic inflammatory diseases. In endothelial cells, complement hyperactivation may translate into cell dysfunction, favoring thrombus formation. The aim of this study was to investigate the role of the C5a/C5aR1 axis as opposed to C5b-9 in inducing endothelial dysfunction and loss of antithrombogenic properties. In vitro and ex vivo assays with serum from patients with atypical hemolytic uremic syndrome (aHUS), a prototype rare disease of complement-mediated microvascular thrombosis due to genetically determined alternative pathway dysregulation, and cultured microvascular endothelial cells, demonstrated that the C5a/C5aR1 axis is a key player in endothelial thromboresistance loss. C5a added to normal human serum fully recapitulated the prothrombotic effects of aHUS serum. Mechanistic studies showed that C5a caused RalA-mediated exocytosis of von Willebrand factor (vWF) and P-selectin from Weibel-Palade bodies, which favored further vWF binding on the endothelium and platelet adhesion and aggregation. In patients with severe COVID-19 who suffered from acute activation of complement triggered by severe acute respiratory syndrome coronavirus 2 infection, we found the same C5a-dependent pathogenic mechanisms. These results highlight C5a/C5aR1 as a common prothrombogenic effector spanning from genetic rare diseases to viral infections, and it may have clinical implications. Selective C5a/C5aR1 blockade could have advantages over C5 inhibition because the former preserves the formation of C5b-9, which is critical for controlling bacterial infections that often develop as comorbidities in severely ill patients. </p>

PMID: 35737839; PMCID: PMC9245614.</p> <p>Abstract</p> <p>Protein aggregation into amyloid fibrils is the archetype of aberrant biomolecular self-assembly processes, with more than 50 associated diseases that are mostly uncurable. Understanding aggregation mechanisms is thus of fundamental importance and goes in parallel with the structural characterization of the transient oligomers formed during the process. Oligomers have been proven elusive to high-resolution structural techniques, while the large sizes and long time scales, typical of aggregation processes, have limited the use of computational methods to date. To surmount these limitations, we here present multi-<em>e</em>GO, an atomistic, hybrid structure-based model which, leveraging the knowledge of monomers conformational dynamics and of fibril structures, efficiently captures the essential structural and kinetics aspects of protein aggregation. Multi-<em>e</em>GO molecular dynamics simulations can describe the aggregation kinetics of thousands of monomers. The concentration dependence of the simulated kinetics, as well as the structural features of the resulting fibrils, are in qualitative agreement with in vitro experiments carried out on an amyloidogenic peptide from Transthyretin, a protein responsible for one of the most common cardiac amyloidoses. Multi-<em>e</em>GO simulations allow the formation of primary nuclei in a sea of transient lower-order oligomers to be observed over time and at atomic resolution, following their growth and the subsequent secondary nucleation events, until the maturation of multiple fibrils is achieved. Multi-<em>e</em>GO, combined with the many experimental techniques deployed to study protein aggregation, can provide the structural basis needed to advance the design of molecules targeting amyloidogenic diseases.</p>

MOLA AGGREGATED EXPERIMENT DATA RECORD
The MOLA AEDR data set contains the raw altimetry measurements acquired by the Mars Orbiter Laser Altimeter on the Mars Global Surveyor spacecraft. The AEDR data are used as input to the MOLA Precision Experiment Data Record (PEDR) data set, in which the altimetry data have been corrected according to precise measurements of the orbit. The PEDR data set is preferred for science analysis.

Spatial aggregation and seedling survival of the Borneo Ironwood (Eusideroxylon zwageri)
<p>This dataset was collected by the Imperial College London MRes Tropical Forest Ecology Field Course as a group project in Maliau Basin. Fieldwork was carried out on 29 Jan - 1 Feb 2018. A 4-ha plot was set up to assess the spatial aggregation of the Borneo Ironwood (Eusideroxylon zwageri, local name Belian) and seedling survival. All trees with DBH >1cm were mapped with XY coordinate rounded to nearest 1m. DBH class was rounded down to the nearest 10cm, e.g. size 10, 20, 30 etc. Size class 1 refers to trees with DBH between 1 and 10cm. A swamp area on the north and northeast of ~1-ha was not assessible hence data defficient, but as Belian is not known to growth in water logged conditions the species was presumed to be absent in this swamp area. Belian seedlings were surveyed in 540 5x5m subplots in the centre zone of the 4-ha plot. In each seedling plot, healthy and damaged (primarily herbivory damage by mammals, defined as damaged when top shoot, i.e. apical meristem, was missing). Other variables recorded in seedling plots include slope angle and ground vegetation cover.</p>

<p>This data set includes supporting data for the manuscript 'Modelling the size distribution of aggregated volcanic ash and implications for operational atmospheric dispersion modelling' submitted to <em>Atmospheric Chemistry and Physics</em>.</p> <p>The data set consists of the following folders:</p> <p><br> 1. 'SensitivityStudy_Uniform' includes input files and data output from the sensitivity study presented in Section 3 of the paper.</p> <p>2. Folders with the prefix 'Scaling' include the data generated from the scaling analysis presented in Section 4.</p> <p>3. Aerial drones reveal the dynamic structuring of sea turtle breeding aggregations and minimum survey effort required to capture climatic and sex-specific effects
<p class="MsoNormal">Quantifying how animals use key habitats and resources for their survival allows managers to optimise conservation planning; however, obtaining representative sample sizes of wildlife distributions in both time and space is challenging, particularly in the marine environment. Here, we used unoccupied aircraft systems (UASs) to evaluate temporal and spatial variation in the distribution of loggerhead sea turtles (<em>Caretta caretta</em>) at two high-density breeding aggregations in the Mediterranean, and the effect of varying sample size and survey frequency. In May–June of 2017 to 2019, we conducted 69 surveys, assimilating 10,075 inwater turtle records at the two sites. Optimal survey frequency to capture the dynamics of aggregations over the breeding period was <2-week intervals and >500 turtles (from the combined surveys). This minimum threshold was attributed to the core-area use of female turtles shifting across surveys in relation to wind direction to access warmer nearshore waters and male presence. Males were more widely distributed within aggregations than females, particularly in May when mating encounters were high. Most males were recorded swimming and oriented parallel to shore, likely to enhance encounter rates with females. In contrast, most females were generally stationary (resting on the seabed or basking), likely to conserve energy for reproduction, with orientation appearing to shift in relation to male numbers at the breeding area. Thus, by identifying the main factors regulating the movement and distribution of animals, appropriate survey intervals can be selected for appropriate home range analyses. Our study demonstrates the versatility of UAS to capture the fine-scale dynamics of wildlife aggregations and associated factors, which is important for implementing effective conservation.</p>
ACOS GOSAT/TANSO-FTS Level 2 bias-corrected XCO2 and other select fields from the full-physics retrieval aggregated as daily files V9r (ACOS_L2_Lite_FP) at GES DISC
Version 9r is the current version of the data set. Older versions will no longer be available and are superseded by Version 9r.The ACOS Lite files contain bias-corrected XCO2 along with other select fields aggregated as daily files. Orbital granules of the ACOS Level 2 standard product (ACOS_L2S) are used as input. The "ACOS" data set contains Carbon Dioxide (CO2) column averaged dry air mole fraction for all soundings for which retrieval was attempted. These are the highest-level products made available by the OCO Project, using TANSO-FTS spectral radiances.The GOSAT team at JAXA produces GOSAT TANSO-FTS Level 1B (L1B) data products for internal use and for distribution to collaborative partners, such as ESA and NASA. These calibrated products are augmented by the OCO Project with additional geolocation information and further corrections. Thus produced Level 1B products (with calibrated radiances and geolocation) are the input to the "ACOS" Level 2 production process.
