Pinon-Juniper Dendrometer, Height, and Crown Area Measurements at Cerro Montosa, Sevilleta National Wildlife Refuge, New Mexico (2006-2009)
Allometry is a standard method of determining biomass and Net Primary Production of many trees. One of the standard variables used in such allometric regressions is bole diameter. On straight trunk trees measurements at breast height (DBH) taken using a DBH tape is adequate for quantifying changes in diameter over time. However, in scrub forests such as the Pinon Juniper PJ woodlands, common on the Sevilleta, both the pinon and particularly the junipers are relatively short and multi-trunked so that measurements must be taken near the ground and the consistency between measurements often lead to erroneous growth analysis. To reduce such discrepancy in readings, dendrometer bands were installed on 20 pinons and 20 junipers in the Cerro Montoso area where understory ANPP has been measured for some time. These dendrometers quantify the expansion (and contraction) of the tree bole through time. The beginning diameter at the time of installation was also measured and recorded. Heights of the 40 trees were also measured. The diameter of the foliage was measured across these trees at the widest point and then again on an axis perpendicular to this first reading. Readings of the dendrometers are repeated on about a monthly basis through the growing season. Heights of the trees are done on an annual basis. Measurements are collected to quantify the increase in bole diameter for a set of Pinons and Junipers in this area over time.
High-frequency observations of overwash water and bed elevations from five MeOw (Measuring Overwash) stations: Smith Island, VA, August-November 2019
Water and bed surface elevation measurements from five MeOw (Measuring Overwash) stations installed on Smith Island, VA, from 21 August 2019 to 22 November 2019. The five MeOw stations consist of one ultrasonic distance sensor inside a stilling well to measure water levels, and a second attached to an arm extending outwards from the well to measure the elevation of the sediment surface (when dry) or the water surface (when inundated). Note: Station MeOw2 was damaged and produced no data, and so is not included in this dataset.

The use of ancient DNA from sediment as a viable measure of Quaternary biodiversity within the Brecon Beacons National Park: a preliminary study

Contains 3 files.

One spreadsheet containing processed sequences -SM1_results_spreadsheets

The other two containing Read 1 and Read 2 of the raw sequences (fastq).

Abstract

A major challenge in the study of Quaternary environments is the variable preservation of paleoenvironmental evidence at many locations. Using ancient DNA from sediment cores (sedaDNA) allows the reconstruction of past biodiversity patterns at locations where traditional techniques often only reveal a partial picture. To test the feasibility of this approach, we analysed a sediment core from a peat bog, Traeth Mawr, in the Brecon Beacons National Park. Plant DNA was extracted and subjected to DNA metabarcoding, combining the use of universal PCR primers with Next-Generation Sequencing, to analyse a 7-143bp segment of the chloroplast trnL gene in five temporally separated sections of the core. Pollen analyses, from sections of the core, provide a comparison between the sedaDNA results and more traditional methods for studying the paleoenvironment. Partial reconstruction of past environmental variation was possible with the recovery of 113,682 DNA sequences representing six plant families, adding complementary results to traditional pollen analysis. This study is the first in the UK to successfully report the sequencing of ancient plant DNA from terrestrial sediments (currently a peat bog) using a DNA metabarcoding approach and exemplifies a promising area of research for addressing a range of questions about Quaternary environmental change.
