Alzheimer's disease: Ablating single master site abolishes tau hyperphosphorylation
<p>Hyperphosphorylation of the neuronal tau protein is a hallmark of neurodegenerative tauopathies such as Alzheimer's disease. A central unanswered question is why tau becomes progressively hyperphosphorylated. Here, we show that tau phosphorylation is governed by interdependence— a mechanistic link between initial site-specific and subsequent multi-site phosphorylation. Systematic assessment of site interdependence identified distinct residues (threonine-50, threonine-69, and threonine-181) as master sites that determine propagation of phosphorylation at multiple epitopes. CRISPR point mutation and expression of human tau in Alzheimer's mice showed that site interdependence governs physiologic and amyloid-associated multi-site phosphorylation and cognitive deficits, respectively. Combined targeting of master sites and p38α, the most central tau kinase linked to interdependence, synergistically ablated hyperphosphorylation. In summary, our work delineates how complex tau phosphorylation arises to inform therapeutic and biomarker design for tauopathies.</p>

Mapping and assembly of the Midas cichlid male-specific region supports molecular parallelism in the evolution of a master sex-determining role for amhr2
<p>The evolution of sex chromosomes and their differentiation from autosomes is a major event during genome evolution that happened many times in several lineages. The repeated evolution and lability of sex-determination mechanisms in fishes makes this a well-suited system to test for general and predictable patterns in evolution. According to current theory, differentiation is triggered by the suppression of recombination following the evolution of a new master-sex determining gene. However, the molecular mechanisms that establish recombination suppression are known from few examples, owing to the intrinsic difficulties of assembling sex determining regions (SDRs). Forward-genetics data and the development of long-read sequencing have generated a wealth of data questioning central aspects of the current theory. Here, we demonstrate that sex in Midas cichlids is determined by an XY system, identify and assemble the SDR by combining forward-genetics, long-read sequencing and optical mapping. We show how long-reads aid in the detection of artifacts in genotype-phenotype mapping that arise from incomplete genome assemblies. The male-specific region is restricted to a 100 kb segment on chromosome 4 that harbors transposable elements and a Y-specific duplicate of the anti-Mullerian receptor 2 locus, a known sex-determining gene. Our data suggests that <em>amhr2Y</em> originated by an interchromosomal translocation from chromosome 20 to 4 predating the split of Midas and Flier cichlids. In the later, it is pseudogenized and translocated to another chromosome. Duplication of anti-Mullerian genes is a common route to establishing new sex determiners, highlighting the role of molecular parallelism in the evolution of sex determination.</p>
EVALUATION OF RADAR-BASED PRECIPITATION DATASETS FOR APPLICATIONS IN THE LOUISIANA COASTAL MASTER PLAN
<p>A suite of hydrological and ecological models is used in the Louisiana Coastal Master Plan (CMP) Integrated Compartment Model (ICM) to assess potential benefits of restoration and protection projects. A main driving input for these models is precipitation. Since precipitation is considered a major source of freshwater in coastal Louisiana (50-60 inches/year), accurate information about its magnitudes and spatial and temporal distributions is critical for successful implementation of the ICM and other CMP-related modeling studies. Accurate information on precipitation is also critical for establishing reliable and representative water budget analyses for coastal Louisiana. In contrast to the sparse availability of rain gauges, weather radars can provide high-resolution rainfall estimates with excellent spatial resolutions and coverage over coastal Louisiana. The potential value of radar-based precipitation products has not been capitalized on yet by the CMP studies. Louisiana is covered by several National Weather Service (NWS) NEXRAD radar stations, with the two radar stations located near Slidell and Lake Charles being the most relevant to coastal Louisiana. The main objective of the graduate research project is to perform a regional-scale assessment of radar-rainfall datasets available over coastal Louisiana and evaluate whether they can be directly used by the CMP studies. Unlike past assessment studies, which were geared towards short-term applications (e.g., flood prediction and forecasting), the focus of this study will be on assessing accuracy aspects that are of direct relevance to the CMP applications. The study will examine (a) the reproducibility of inter-annual and intra-annual variability by the radar dataset, (b) the representation of extreme rainfall events, and (c) the representation of spatial distributions of annual and seasonal rainfall across various regions of the coast with distinctly different climatic regimes. The study will provide the CMP with quantitative assessment on the advantages and limitations of radar-based precipitation datasets and whether they can be readily used by the ICM models. This research will also provide insight on the levels of uncertainties in the radar-rainfall datasets and the implications for these uncertainties for the ICM models.</p> <p> </p> <p>Please make sure you work with the latest version of this data set only (check the panel on the right for versioning).</p>

Data from: The non-coding RNA CcnA modulates the master cell cycle regulators CtrA and GcrA in Caulobacter crescentus
<p>Bacteria are powerful models for understanding how cells divide and accomplish global regulatory programs. In <em>Caulobacter crescentus</em>, a cascade of essential master regulators supervises the correct and sequential activation of DNA replication, cell division and development of different cell types. Among them, the response regulator CtrA plays a crucial role coordinating all those functions. Here, for the first time we describe the role of a novel factor named CcnA, a cell cycle regulated ncRNA located at the origin of replication, presumably activated by CtrA and responsible for the accumulation of CtrA itself. In addition, CcnA may be also involved in the inhibition of translation of the S-phase regulator, GcrA, by interacting with its 5' untranslated region (5'-UTR). Performing <em>in vitro</em> experiments and mutagenesis, we propose a mechanism of action of CcnA based on liberation (<em>ctrA</em>) or sequestration (<em>gcrA</em>) of their ribosome-binding site (RBS). Finally, its role may be conserved in other alphaproteobacterial species, such as <em>Sinorhizobium</em> <em>meliloti</em>, representing indeed a potentially conserved process modulating cell cycle in <em>Caulobacterales </em>and<em> Rhizobiales</em>. </p>
