Data from: Expressed mutational load increases towards the edge of a species' geographic range

There is no general explanation for why species have restricted geographic distributions. One hypothesis posits that range expansion or increasing scarcity of suitable habitat result in accumulation of mutational load due to enhanced genetic drift, which constrains population performance towards range limits and further expansion. We tested this hypothesis in the North American plant, Arabidopsis lyrata. We experimentally assessed mutational load by crossing plants of 20 populations from across the entire species range and by raising the offspring of within- and between-population crosses at five common garden sites within and beyond the range. Offspring performance was tracked over three growing seasons. The heterosis effect, depicting expressed mutational load, was increased in populations with heightened genomic estimates of load, longer expansion distance or long-term isolation, and a selfing mating system. The decline in performance of within-population crosses amounted to 80%. Mutation accumulation due to past range expansion and long-term isolation of populations in the area of range margins is therefore a strong determinant of population-mean performance, and the magnitude of effect may be sufficient to cause range limits.

Data from: Evolutionary trade-offs of insecticide resistance – the fitness costs associated with target-site mutations in the nAChR of Drosophila melanogaster

<p>The evolution of resistance to drugs and pesticides poses a major threat to human health and food security. Neonicotinoids are highly effective insecticides used to control agricultural pests. They target the insect nicotinic acetylcholine receptor and mutations of the receptor that confer resistance have been slow to develop, with only one field-evolved mutation being reported to date. This is an arginine to threonine substitution at position 81 of the nAChR_β1 subunit in neonicotinoid resistant aphids. To validate the role of R81T in neonicotinoid resistance and to test whether it may confer any significant fitness costs to insects, CRISPR/Cas9 was used to introduce an analogous mutation in the genome of <i>Drosophila melanogaster</i>. Flies carrying R81T showed an increased tolerance (resistance) to neonicotinoid insecticides, accompanied by a significant reduction in fitness. In comparison, flies carrying a deletion of the whole nAChR_α<em>6</em> subunit, the target-site of spinosyns, showed an increased tolerance to this class of insecticides but presented almost no fitness deficits.</p>

Data from: Large-scale mutation in the evolution of a gene complex for cryptic coloration

<p class="western">The types of mutations affecting adaptation in the wild are only beginning to be understood. In particular, whether structural changes shape adaptation by suppressing recombination or by creating new mutations is unresolved. Here we show that multiple, linked but recombining loci underlie cryptic color morphs of <i>Timema chumash </i>stick insects. In a related species, these loci are found in a region of suppressed recombination, forming a supergene. However, in seven species of <i>Timema </i>we find that a mega-base size 'supermutation' has deleted color loci in green morphs. Moreover, we find that balancing selection likely contributes more to maintaining this mutation than does introgression. Our results show how suppressed recombination and large-scale mutation can help package gene complexes into discrete units of diversity, such as morphs, ecotypes, or species.</p>

Short-term heritable variation overwhelms two hundred generations of mutational variance for metabolic traits in Caenorhabditis elegans

<p>Metabolic disorders have a large heritable component, and have increased markedly in human populations over the past few generations.  Genome-wide association studies of metabolic traits typically find a substantial unexplained fraction of total heritability, suggesting an important role of spontaneous mutation.  An alternative explanation is that epigenetic effects contribute significantly to the heritable variation. Here we report a study designed to quantify the cumulative effects of spontaneous mutation on adenosine metabolism in the nematode <i>Caenorhabditis elegans</i>, including both the activity and concentration of two metabolic enzymes and the standing pools of their associated metabolites.  The only prior studies on the effects of mutation on metabolic enzyme activity, in <i>Drosophila melanogaster</i>, found that total enzyme activity presents a mutational target similar to that of morphological and life-history traits.  However, those studies were not designed to account for short-term heritable effects.  We find that the short-term heritable variance for most traits is of similar magnitude as the variance among MA lines.  This result suggests that the potential heritable effects of epigenetic variation in metabolic disease warrant additional scrutiny.</p>
