Data from: Protocols for the experimental release of kelp (Laminariales) zoospores

Kelps (order Laminariales) are foundation species in temperate and arctic seas globally, but they are in decline in many places. Laminarian kelp have an alternation of generations and this poses challenges for experimental studies due to the difficulties in achieving zoospore release and gametophyte growth. Here we review and synthesize the protocols that have been used to induce zoospore release in kelps to identify commonalities and provide guidance on best practices. We found 171 papers, where zoospore release was induced in four kelp families from 35 different ecoregions. The most commonly treated family was Laminariaceae, followed by Lessoniaceae and the most studied ecoregion was Central Chile, followed by the Southern California Bight. Zoospore release generally involved three steps: a pre-treatment which included cleaning of the reproductive tissue to eliminate epiphytic organisms, followed by desiccation of the tissue, and finally a post-desiccation immersion of the reproductive material in a seawater medium for zoospore release. Despite these commonalities, there was a high degree of variation in the detail within each of these steps, even among studies within genera and from the same ecoregions. This suggests that zoospore release may be relatively insensitive across the Laminariales. Nevertheless, greater attention to standardisation of protocols would facilitate comparisons of kelp zoospore release across species and locations. Based on practicality and most common practices, we suggest zoospore release should be achieved through 1) wiping clean the reproductive material, 2) desiccating the reproductive material by wrapping in moist paper towels for 12 hours at room temperature in darkness, and 3) submerging the tissue in seawater medium for one hour at room temperature in darkness. The choice of seawater medium should depend on the actual experiment conducted. Standardized protocols will facilitate broad scale comparative studies across this key taxa.

Data from: Soils from cold and snowy temperate deciduous forests release more nitrogen and phosphorus after soil freeze–thaw cycles than soils from warmer, snow-poor conditions

<p>Effects of global warming are most pronounced in winter. A reduction in snow cover due to warmer atmospheric temperature in formerly cold ecosystems, however, could counteract an increase in soil temperature by reduction of insulation. Thus, soil freeze-thaw cycles (FTC) might increase in frequency and magnitude with warming, potentially leading to a disturbance of the soil biota and release of nutrients.</p> <p>Here, we assessed how soil freeze-thaw magnitude and frequency affect short-term release of nutrients in temperate deciduous forest soils by conducting a three factorial gradient experiment with ex-situ soil samples in climate chambers. The fully-crossed experiment included soils from forests dominated by <i>Fagus sylvatica</i> (European beech) that originate from different winter climate (mean coldest month temperature range ΔT &gt; 4 K), a range of FTC magnitudes from no (T = 4.0 °C) to strong (T = -11.3 °C) soil frost, and a range of FTC frequencies (f = 0–7). We hypothesized that higher FTC magnitude and frequency, respectively, will increase the release of nutrients. Furthermore, soils from cold climates with historically stable winter soil temperatures due to deep snow cover will be more responsive to FTC than soils from warmer, more fluctuating winter soil climates.</p> <p>FTC magnitude and, to a lesser extent, also FTC frequency resulted in increased nitrate, ammonium, and phosphate release almost exclusively in soils from cold, snow-rich sites. The hierarchical regression analyses of our three-factorial gradient experiment revealed that the effects of climatic origin (mean minimum winter temperature) followed a sigmoidal curve for all studied nutrients and was modulated either by FTC magnitude (phosphate) or by FTC magnitude and frequency (nitrate, ammonium) in complex two- and, for all studied nutrients, in threefold interactions of the environmental drivers. Compared to initial concentrations, soluble nutrients were predicted to increase to 250 % for nitrate (up to 16 µg NO<sub>3</sub>-N kg<sup>-1</sup>DM), to 110 % for ammonium (up to 60 µg NH<sub>4</sub>-N kg<sup>-1</sup>DM), and to 400 % for phosphate (2.2 µg PO<sub>4</sub>-P kg<sup>-1</sup>DM) at the coldest site for strongest magnitude and highest frequency. Soils from warmer sites showed little nutrient release and were largely unaffected by the FTC treatments except for above-average nitrate release at the warmest sites in response to extremely cold FTC magnitude.</p> <p>We suggest that currently warmer forest soils have historically already passed the point of high responsiveness to winter climate change, displaying some form of adaptation either in the soil biotic composition or in labile nutrient sources. Our data suggests that previously cold sites, which will lose their protective snow cover during climate change, are most vulnerable to increasing FTC frequency and magnitude, resulting in strong shifts in nitrogen and phosphorus release. In nutrient poor European beech forests of the studied Pleistocene lowlands, nutrients released over winter may be leached out, inducing reduced plant growth rates in the following growing season.</p> <p>Here, we provide the raw data of a three-factorial regression experiment and the R-Code used in the hierarchical regression analysis of this raw data in Kreyling et al.: Soils from cold and snowy temperate deciduous forests release more nitrogen and phosphorus after soil freeze-thaw cycles than soils from warmer, snow-poor conditions.</p>

Data mining tool to discover DevOps trends from public repositories: Predicting Release Candidates with gthbmining.rc

<p>Public repositories have been performing an essential role in bring- ing software and services to technical communities and general users. Most of the cases, public repositories have a DevOps tool, with a live and historical database behind it, to support delivering and all steps this software or service should adopt before going to production. This paper introduces gthbmining, a data mining set of tools to discover DevOps trends from public repositories, and presents the module gthbmining.rc. Considering the premise of a GitHub public repository, the main contribution here is pre- dicting release candidates, an important label a software release has. The methodology, architecture, components and interfaces are explained, as well as potential users. The results show a reliable and flexible tool, as classifiers metrics and graphics are provided, along with the possibility to add new data mining algorithms in the open source module presented. Related works are also supplied, and a conclusion shows the outcomes gthbmining.rc can provide.</p>

<p>This collection of data sets was obtained during a study of the effect of acute ethanol intoxication on vigilance-dependent astroglia network activation in mice. The main findings have been that ethanol inhibits astroglia activation by inhibiting vigilance-dependent norepinephrine release. This leads to a failure of activation of alpha<sub>1A</sub>-adrenergic receptors on astroglia. Further, this study has revealed that ethanol inhibition of cerebellar Bergmann glia Ca<sup>2+</sup> activation does not account for ataxic motor behavior, but may rather contribute to cognitive deficits.</p>

Data From: Evaluating the correlation between genome-wide diversity and the release of plastic phenotypic variation in experimental translocations to novel natural environments

<p>Phenotypic reaction norms are often shaped and constrained by selection and are important for allowing organisms to respond to environmental change. However, selection cannot constrain reaction norms for environmental conditions that populations have not experienced. This may allow cryptic neutral genetic variation for the reaction norm to accumulate such that a release of phenotypic variation occurs when it is exposed to novel conditions. Most genomic diversity behaves as if functionally neutral. Genome-wide diversity metrics may therefore correlate with levels of cryptic genetic variation and, as a result, could exhibit a positive relationship with a release of phenotypic variation in novel environments. To test this hypothesis, we conducted translocations of juvenile brook trout (Salvelinus fontinalis) from 12 populations to novel uninhabited ponds that represented a gradient of environmental conditions. We assessed reaction norms for morphological traits (body size and four morphometric relative warps) across pond environmental gradients and evaluated the effect of genome-wide heterozygosity on phenotypic variability. All traits displayed plastic reaction norms. Overall, we found some evidence that a release of phenotypic variation consistent with cryptic genetic variation can occur in novel environmental conditions. However, the extent to which this release was correlated with average genome-wide diversity was limited to only one of five morphological traits examined. Our results suggest that the link between genomic diversity and the accumulation of cryptic genetic variation in reaction norms may be limited. Similarly, reaction norms were constrained for many of the morphological traits examined. Past conditions may have constrained reaction norms in the putatively novel environments despite significant deviations from contemporary source population habitat. Additionally, as a generalist colonizing species brook trout may exhibit plastic phenotypes across a wide range of environmental conditions.</p>

Data from: Ecological release of the Anna's Hummingbird during a northern range expansion

During range expansions species can experience rapid population growth if changes in climate or interspecific interactions remove limits on growth rates in novel habitats. Here I document a century of range expansion in the Anna's Hummingbird (Calypte anna) and investigate the causes of its recent abundance through a combination of demographic, climatic, and phenological analyses. Christmas Bird Count records show that populations have been growing within the native range since the early twentieth century. Sites across the Pacific Northwest show striking fits to simple models of exponential growth following colonization in the 1960's and 70's, and nest records indicate that the species now delays the start of the nesting season by at least 16 days in the north. Although the species now occurs in a much wider range of climates than it did in its native range, the fastest growing sites in the northwest are in regions with minimum breeding season temperatures similar to those occupied by the species prior to the expansion. Range expansions in the Anna's Hummingbird thus reflect an ecological release likely caused by a mix of human facilitation, introduced plants, and phenological acclimation that allowed a California native to expand across western North America.

Data from: No release for the wicked: enemy release is dynamic and not associated with invasiveness

The enemy release hypothesis predicts that invasive species will receive less damage from enemies, compared to co-occurring native and noninvasive exotic species in their introduced range. However, release operating early in invasion could be lost over time and with increased range size as introduced species acquire new enemies. We used three years of data, from 61 plant species planted into common gardens, to determine whether (1) invasive, noninvasive exotic, and native species experience differential damage from insect herbivores and mammalian browsers, and (2) enemy release is lost with increased residence time and geographic spread in the introduced range. We find no evidence suggesting enemy release is a general mechanism contributing to invasiveness in this region. Invasive species received the most insect herbivory, and damage increased with longer residence times and larger range sizes at three spatial scales. Our results show that invasive and exotic species fail to escape enemies, particularly over longer temporal and larger spatial scales.

Data from: Controlled release of basic fibroblast growth factor from a peptide biomaterial for bone regeneration

<p>Self-assembled peptide scaffolds based on D-RADA16 (D16) are an important matrix for controlled drug release and 3D cell culture. In this work, D16 peptide hydrogels were coated on artificial bone composed of nano-hydroxyapatite/polyamide 66 (NHA/PA66) to obtain a porous drug-releasing structure for treating bone defects. The developed materials were characterized via transmission electron microscopy (TEM), scanning electron microscopy (SEM). The proliferation and adhesion of bone mesenchymal stem cells (BMSCs) were examined by Confocal laser microscopy (CLS) and CCK-8 experiments. The osteogenic ability of the porous materials towards bone BMSCs was examined in vitro by staining with Alizarin Red S and alkaline phosphatase (ALP) and bioactivity were evaluated in vivo. The results revealed that NHA/PA66/D-RADA16/bFGF reduce the degradation rate of D16 hydrogels and prolong sustained release of bFGF, which would promote BMSCs proliferation, adhesion and osteogenesis in vitro and bone repair in vivo. Thus, it deserves more attention and is worthy of further research.</p>

Data from: Enemy release from the effects of generalist granivores can facilitate Bromus tectorum invasion in the Great Basin Desert

The enemy release hypothesis (ERH) of plant invasion asserts that natural enemies limit populations of invasive plants more strongly in native ranges than in non-native ranges. Despite considerable empirical attention, few studies have directly tested this idea, especially with respect to generalist herbivores. This knowledge gap is important because escaping the effects of generalists is a critical aspect of the ERH that may help explain successful plant invasions. Here, we used consumer exclosures and seed addition experiments to contrast the effects of granivorous rodents (an important guild of generalists) on the establishment of cheatgrass (Bromus tectorum) in western Asia, where cheatgrass is native, vs. the Great Basin Desert, USA, where cheatgrass is exotic and highly invasive. Consistent with the ERH, rodent foraging reduced cheatgrass establishment by nearly 60% in western Asia but had no effect in the Great Basin. This main result corresponded with a region-specific foraging pattern: rodents in the Great Basin but not western Asia generally avoided seeds from cheatgrass relative to seeds from native competitors. Our results suggest that enemy release from the effects of an important guild of generalists may contribute to the explosive success of cheatgrass in the Great Basin. These findings corroborate classic theory on enemy release and expand our understanding of how generalists can influence the trajectory of exotic plant invasions.
