Rapid and specific degradation of endogenous proteins in mouse models using auxin-inducible degrons

<p>Auxin-inducible degrons are a chemical genetic tool for targeted protein degradation and are widely used to study protein function in cultured mammalian cells. Here we develop CRISPR-engineered mouse lines that enable rapid and highly specific degradation of tagged endogenous proteins <em>in vivo</em>. Most but not all cell types are competent for degradation. By combining ligand titrations with genetic crosses to generate animals with different allelic combinations, we show that degradation kinetics depend upon the dose of the tagged protein, ligand, and the E3 ligase substrate receptor TIR1. Rapid degradation of condensin I and condensin II – two essential regulators of mitotic chromosome structure - revealed that both complexes are individually required for cell division in precursor lymphocytes, but not in their differentiated peripheral lymphocyte derivatives. This generalisable approach provides unprecedented temporal control over the dose of endogenous proteins in mouse models, with implications for studying essential biological pathways and modeling drug activity in mammalian tissues.</p>

The gut microbiome variability of a butterflyfish increases on severely degraded Caribbean reefs

<p>Environmental degradation has the potential to alter key mutualisms that underlie the structure and function of ecological communities. How microbial communities associated with fishes vary across populations and in relation to habitat characteristics remains largely unknown despite their fundamental roles in host nutrition and immunity. We find significant differences in the gut microbiome composition of a facultative coral-feeding butterflyfish (Chaetodon capistratus) across Caribbean reefs that differ markedly in live coral cover (∼0–30%). Fish gut microbiomes were significantly more variable at degraded reefs, a pattern driven by changes in the relative abundance of the most common taxa potentially associated with stress. We also demonstrate that fish gut microbiomes on severely degraded reefs have a lower abundance of Endozoicomonas and a higher diversity of anaerobic fermentative bacteria, which may suggest a less coral dominated diet. The observed shifts in fish gut bacterial communities across the habitat gradient extend to a small set of potentially beneficial host associated bacteria (i.e., the core microbiome) suggesting essential fish-microbiome interactions may be vulnerable to severe coral degradation.</p>

Monitoring of degradation of 5 organic micropollutant under a multitude of scenarios

<p>The spreadsheet contains HPLC integration peaks monitoring the photolytic and photocatalytic degradation of 5 organic micropollutants (ciprofloxacin, sulfamethoxazole, trimethoprim, venlafaxine and o-desmethyl-venlafaxine) under different conditions. The first sheets show the degradation by UV-A and UV-C photolysis and photocatalysis of each compound in individual solution in milliQ water (initial concentration of each, 2 mg/L). Later, all compounds are present in an initial mixture (concentration of each 2 mg/L, resulting in final mixture solution of 10 mg/L). The impact on degradation of adding 0.1 mM of peroxide in the solution for all 4 processes is&nbsp;studied. Additionally, the impact of: 1)&nbsp;of using simultaneous LED wavelengths (UV-A and UV-C combined); 2) using tap water as matrix; and 3) varying the initial pH for the 1st order kinetic rates of each compound in the mixture is investigated. Plots of the kinetic rate and calculations of EEO values (electrical energy per order consumption) are made. The last spreadsheets contain kinetic monitoring of several experiments performed by altering the composition of the matrix with the addition of nitrates, humic acids and bicarbonates. The data was used to obtain&nbsp;a surface-response box-behnken design of experiments.</p>

Monitoring of ciprofloxacin degradation by UV-A LED photolysis and photocatalysis under different scenarios

<p>The spreadsheet contains the HPLC integration peaks monitoring the photolytic and photocatalytic degradation of the antibiotic ciprofloxacin (CIP) in milliQ water. The initial concentration of CIP is 10 mg/L. In the spreadsheets is shown the obtainment of 1st order kinetic constant rates for different photoreactor designs using 2, 3, 4 or 6 UVA-LED strips around the reactor, distanced 10 or 15 mm from the reactor&#39;s walls. The influence of different controlled periodic illumination&#39;s duty cycle (0.50 and 0.75) is also shown. Calculations of EEO (electrical energy per order consumption) are also made for each reactor set up.</p> <p>The irradiance values for each photoreactor design calculated by an optical software is presented as planar projections of the cylindrical reactor. Images showing the radiant flux in the photoreactor&#39;s middle cross section obtained by the software are also shown</p> <p>Finally, a full factorial design of experiments is made to obtain a model of prediction of kinetic constant rates and EEO values. Predicted vs experimental values are plotted.</p>

Degradation of pharmaceuticals using the hybrid ozonation-filtration system

<p>The hybrid ozonation-membrane filtration unit allows the simultaneous treatment of the contaminated water with ozone and micro- and nano-filtration (MF/NF) under controlled conditions using commercial single tubular ceramic membranes with an inside-out operation. In total, four commercial ceramic (CM) membranes with different compositions and pore sizes were selected. The composition of the selected membranes is given below</p> <ul> <li>TiO<sub>2</sub>/ZrO<sub>2</sub> composition of three different pore sizes: 50-, 150-, and 300 kDa MWCO) and</li> <li>TiO<sub>2</sub>/ TiO<sub>2</sub>+ZrO<sub>2</sub> composition of one pore size (0.200 &mu;m MWCO).</li> </ul> <p>The files contain information on the degradation of the pharmaceuticals using the different membranes under the three surface modifications; CeO2, CeTiOx and CeO2+CeTiOx.</p> <p>Permeability tests have been performed on all four membranes. The filtration unit can operate from 0.3 -1.5 bar pressure with a permeability flow ranging from 200 - 1900 L.m<sup>-2</sup>.h<sup>-1</sup> at 1 bar, starting from the lowest molecular weight cut-off (MWCO) membrane (50 kDa) and moving to the higher (0,200 &mu;m) one.</p>

Degradation of pharmaceuticals in batch experiments with ozone and nanoparticles

<p>The catalytic activity of five different nanoparticles (NPs), &alpha;-Al<sub>2</sub>O<sub>3</sub>, &gamma;-Al<sub>2</sub>O<sub>3</sub>, Mn<sub>2</sub>O<sub>3</sub>, CeO<sub>2</sub>, and TiO<sub>2</sub> with different particle sizes during ozonation was examined, and the transferred ozone dose (TOD) required for the degradation of the emerging compounds was defined.</p> <p>Data show the degradation of the micropollutants in batch experiments. We studied the degradation of pharmaceuticals in milliQ in the presence or not of NP (C=0.5 g/L). The initial concentration of the pollutants was 10 &mu;M and the added ozone doses ranged from 0, 10, 20, 50, 75, 100, 150, and 200 &mu;M. Bottles of 240 mL working volume were left to react at ambient temperature for four hours after ozone addition, and samples were collected, filtrated through a 0.2 &mu;M nylon filter, and finally, analyzed in the HPLC. Residual ozone, if any, was determined by the indigo method.</p>

Data for: Activation of bisulfite by LaFeO3 loaded on red mud for degradation of organic dye

<p>In this study, red mud was used as a support for LaFeO3 to prepare a synergistic catalyst (LaFeO3-RM) via ultrasonic-assisted sol-gel method for the enhanced removal of methylene blue (MB) assisted with bisulfite (BS) in an aqueous solution. The Characterization of SEM and BET indicated that comparing with the non-porous structure of bulk LaFeO3, LaFeO3-RM exhibited high surface area and obvious porous structure with a higher pore volume (i.e. 10 times). XRD, XPS and FTIR revealed that in addition to dispersing LaFeO3 particles, the support of porous RM also increased Fe oxidation activity, oxygen-containing functional groups and chemically adsorbed oxygen (from 44.3% to 90.3%) of LaFeO3-RM which enhanced the catalytic performance in structure and chemical composition. MB was removed through the synergistic effect of adsorption and catalysis with MB molecule was first absorbed on the surface and then degraded. The pseudo-first-order kinetic constant of LaFeO3-RM was 6 times than that of LaFeO3. Fe(III) in LaFeO3-RM played a key role in the activation of BS to produce SO4·- by the redox cycle of Fe(III)/Fe(II). With the application of LaFeO3-RM as a promising low-cost synergistic catalyst for the activation of BS under neutral condition, this work provides a new approach of using porous industrial waste to improve the catalytic performance of LaFeO3.</p>

Tibetan pastoralists' attitudes toward grassland degradation and restoration

<p>Ecosystem restoration practices are gaining prominence in coping with the crisis of ecosystem degradation. To improve the long-term effectiveness of restoration activities, community engagement in restoration projects needs to be strengthened. In communities where traditional and local values drive adaptation to environmental changes, a community-engaged restoration approach requires the integration of restoration techniques and local value systems.</p> <p>In two pastoral communities on the Qinghai-Tibetan Plateau, we used the Q method to explore pastoralists' attitudes towards grassland degradation and restoration and to understand different levels of community engagement in grassland restoration activities. Through this study, we aim to promote an understanding of the value-based approach of local communities to enable better community engagement for the long-term effectiveness of ecosystem restoration projects, and contribute to the achievement and synergy of global Sustainable Development Goals (SDGs).</p> <p>The study revealed eight types of pastoralists with different attitudes in two communities, with the type of <em>Active Agents</em> leveraging transformative restoration action through creating value-inclusive narratives of grassland restoration in the community with a higher level of engagement in restoration practices. Alongside this, we explored the pastoralists' perspectives of biodiversity in restoration and the importance of local elites in facilitating multiple-actor cooperation in restoration projects.</p> <p>We recommend that decision-makers recognize the significance of local worldviews and values in facilitating environmental adaptations, as well as the irreplaceable role played by local people in developing value-inclusive narratives that align with the local cultural context. Open dialogues and sufficient communication between multiple stakeholders are needed to build mutual trust and accumulate social capital for driving transformative actions in traditional communities. Such approaches are essential to foster community engagement and gain better momentum in ecosystem management initiatives.</p>

Lenze Gearmotor Degradation Dataset (Lenze-GD)

<p>A run-to-failure experiment&nbsp;for geared motors is introduced. A geared motor is installed&nbsp;in healthy condition and operated until it fails. Throughout&nbsp;the experiment, a data acquisition system is active to monitor&nbsp;the signals of all degradation states. In order to complete the&nbsp;experiment in limited time, the geared motors nominal torque&nbsp;is exceeded. Monthly production and open-circuit string voltage measurements after 10-year operation of three photovoltaic plants in Southern Spain affected by severe potential-induced degradation

<p>Data are formated in a spreadsheet file.&nbsp; Data are presented from three photovoltaic plants in Southern Spain (C&oacute;rdoba province - Northern Andaluc&iacute;a) designed and installed by the same person, with the same photovoltaic module and the same model of inverter, deployed at the same time (end 2009). The plants are severely affected by potential-induced degradation (PID), so that secondary effects produce some by-pass diodes to activate, producing three families of open-circuit voltage (Voc) in the modules (~40V), (~26V) and (~12V) of a total of nominal Voc of 42,6V.&nbsp;</p> <p>Monthly production is shown along 11 years (2010-2021), and the measurements of the open-circuit voltage of the strings after 10 years of operation along with the voltage range of the modules of each string.</p> <p>Sheet 1: configuration of the architecture of the three photovoltaic plants and the features of the photovoltaic module installed.</p> <p>Sheet 2: energy production for 11 years of opetation. Electromagnetic Wave Dataset for Strength Degradation Detection in Reinforced Concrete Structures Using RFID Measurements and CNN Model

<p>This dataset comprises 1,800 electromagnetic wave (EM-wave) images collected from three different reinforced concrete beams subjected to varying levels of corrosion. Each image is classified into 'normal' or 'reduced strength' categories based on the beam's structural integrity. Generated through a non-destructive RFID-based monitoring technique, this dataset integrates advanced analyses like 2-D Fourier transforms and fractal dimensions. It is specifically designed to train and validate Convolutional Neural Networks (CNNs) for detecting strength degradation in reinforced concrete structures.</p>
