Data from: Analysing small insect glands with UV-LDI MS: high-resolution spatial analysis reveals the chemical composition and use of the osmeterium secretion in Themira superba (Sepsidae: Diptera)

For many insect species, pheromones are important communication tools, but chemical analysis and experimental study can be technically challenging because they require the detection and handling of complex chemicals in small quantities. One drawback of traditional mass spectrometry methods such as gas chromatography mass spectrometry is that whole-body extractions from one to several hundred individuals are required, with the consequence that intra- and interindividual differences cannot be detected. Here, we used the recently introduced UV-LDI MS (ultraviolet laser desorption/ionization mass spectrometry) to profile the 'osmeterium' of the sepsid fly Themira superba that is located on the edge of the hind tibia of males. Based on analyses of individual legs, we established that the gland produced a secretion that consisted of oxygenated hydrocarbons and putative isoprenoids. The secretion was first detected 24 h after eclosion, and its transfer to the wings of females during mating was demonstrated using UV-LDI MS. We then tested whether the secretion had an anti-aphrodisiac function, but experimental transfer of the secretion to virgin females did not affect mating success or copulation duration. Throughout the study, UV-LDI MS proved invaluable, because it allowed tracking the natural and experimental transfer of small quantities of pheromones to specific body parts of small flies.

Data from: Population genomic analysis suggests strong influence of river network on spatial distribution of genetic variation in invasive saltcedar across the southwestern US

Understanding the complex influences of landscape and anthropogenic elements that shape the population genetic structure of invasive species provides insight into patterns of colonization and spread. The application of landscape genomics techniques to these questions may offer detailed, previously undocumented insights into factors influencing species invasions. We investigated the spatial pattern of genetic variation and the influences of landscape factors on population similarity in the invasive riparian shrub saltcedar (Tamarix L.) by analyzing 1,997 genome-wide SNP markers for 259 individuals from 25 populations collected throughout the southwestern US. Our results revealed a broad-scale spatial genetic differentiation of saltcedar populations between the Colorado and Rio Grande river basins and identified potential barriers to population similarity along both river systems. River pathways most strongly contributed to population similarity. In contrast, low temperature and dams likely served as barriers to population similarity. We hypothesize that large-scale geographic patterns in genetic diversity resulted from a combination of early introductions from distinct populations, the subsequent influence of natural selection, dispersal barriers, and founder effects during range expansion.

Data from: Sex-determining 3D regulatory hubs revealed by genome spatial auto-correlation analysis

<p>Mammalian sex is determined by&nbsp;opposing networks of ovarian and testicular genes&nbsp;that are well characterized. <p>Supplementary data supporting the <em>SpatialOne: End-to-End Analysis of Spatial </em><em>Transcriptomics at Scale</em> publication</p> <p>&nbsp;</p> <blockquote> <p>To showcase the capabilities of SpatialOne, two human lung cancer formalin-fixed, paraffin-embedded (FFPE) samples are analyzed. These samples are prepared following the CG000495 protocol (Figure 1b), sequenced with the 10x Visium CytAssist, and processed using the 10x SpaceRanger version 2. We also present analysis of two adult mouse samples sequenced using 10x Visium samples (one fresh frozen brain tissue section processed using SpaceRanger v2 and one FFPE kidney sample processed using the SpaceRanger v1), and 75 internal samples.&nbsp;</p> <p>&nbsp;For the human lung cancer samples, single-cell data from the the Lung Cancer Atlas (Salcher et al., 2022) is used as reference. This dataset is filtered to include only Chromium-generated data. For the mice samples, the GSE107585 single-cell dataset serves as reference.
