WaveMAP analysis of extracellular waveforms from monkey premotor cortex during decision-making

<p>Cortical circuits are thought to contain a large number of cell types that coordinate to produce behavior. Current <em>in vivo</em> methods rely on clustering of specified features of extracellular waveforms to identify putative cell types, but these capture only a small amount of variation. Here, we develop a new method (<em>WaveMAP</em>) that combines non-linear dimensionality reduction with graph clustering to identify putative cell types. We apply <em>WaveMAP</em> to extracellular waveforms recorded from dorsal premotor cortex of macaque monkeys performing a decision-making task. Using <em>WaveMAP</em>, we robustly establish eight waveform clusters and show that these clusters recapitulate previously identified narrow- and broad-spiking types while revealing previously unknown diversity within these subtypes. The eight clusters exhibited distinct laminar distributions, characteristic firing rate patterns, and decision-related dynamics. Such insights were weaker when using feature-based approaches. <em>WaveMAP</em> therefore provides a more nuanced understanding of the dynamics of cell types in cortical circuits.</p>

Mitonuclear interactions and introgression genomics of macaque monkeys (Macaca) highlight the influence of behaviour on genome evolution

<p>In most macaques, females are philopatric and males migrate from their natal ranges, which results in pronounced divergence of mitochondrial genomes within and among species. We therefore predicted that some nuclear genes would have to acquire compensatory mutations to preserve compatibility with diverged interaction partners from the mitochondria. We additionally expected that these sex-differences would have distinctive effects on gene flow in the X and autosomes. Using new genomic data from 29 individuals from eight species of Southeast Asian macaque, we identified evidence of natural selection associated with mitonuclear interactions, including extreme outliers of interspecies differentiation and metrics of positive selection, low intraspecies polymorphism, and atypically long runs of homozygosity associated with nuclear-encoded genes that interact with mitochondria-encoded genes. In one individual with introgressed mitochondria, we detected a small but significant enrichment of autosomal introgression blocks from the source species of her mitochondria that contained genes that interact with mitochondria-encoded loci. Our analyses also demonstrate that sex-specific demography sculpts genetic exchange across multiple species boundaries. These findings show that behaviour can have profound but indirect effects on genome evolution by influencing how interacting components of different genomic compartments (mitochondria, the autosomes, the sex chromosomes) move through time and space.</p>

Titi monkey father-daughter bond-related behaviors explain stress response variability

<p>[Dataset]&nbsp;Social interactions regulate our behavior and physiology, and strong social bonds can buffer us from stress. Coppery titi monkeys (<em>Plecturocebus cupreus</em>) are socially monogamous South American monkeys that display strong social bonds. Infants form selective bonds with their fathers, making them ideal for studying father-daughter bonds. We established a novel method for quantifying variability in expression of bond-related behaviors in females (n =12). We also investigated how manipulations of oxytocin (OT) and vasopressin (AVP) influenced juvenile behavior and physiology. Subjects received acute intranasal treatments of saline, low/medium/high OT, low/high AVP, or OT antagonist (OTA) prior to an acute social separation. General linear mixed-effects model results revealed fathers were significant behavioral and physiological stress buffers for their daughters, as evidenced by fewer distress vocalizations (p &lt; .001), less locomotion (p &lt; .001), and lower plasma cortisol (p &lt; .001). Females vocalized less if they exhibited greater expression of bond-related behaviors with their fathers as infants (p = .01), and this stress buffering effect remained even when the daughter was separated from the father (p = .001). While treatments did not alter behaviors, OTA treatments caused the largest rise in plasma cortisol (p &lt; .001), suggesting blockade of OT receptors can inhibit fathers&rsquo; stress-buffering effects. Remarkably, females with greater expression of father-daughter bond-related behaviors exhibited an overall reduced physiological separation distress response (p = .04). Findings from the present study advance current knowledge of the neurobiological mechanisms foundational to female bonds and help inform how social disruptions may differently impact individuals based on expression of bond-related behaviors.</p>

Data from: Costs of social conflict: do injuries mirror patterns of aggression in blue monkeys?

<p class="MsoNormal">In wild animals, injuries often arise from intraspecific contests, and therefore may reveal challenges and selection pressures related to social conflict. We evaluated whether known aggressive patterns predicted injury patterns in wild blue monkeys, in which most injuries of known cause resulted from conspecifics. After describing injuries and their causes in 12 groups monitored over 13 years, we compared injury distributions across age-sex class and season. Adult males were much more likely, adult females about equally likely, and immatures much less likely to sustain injuries relative to random expectation. Among adult males, injuries were most common in the mating season. Using multivariate models, we examined how individual, social, and demographic factors predicted injury rates in adult females and immatures. Rates were higher for adult females versus all immatures, for individuals in smaller groups, and for those living in groups frequently exposed to novel adult males and with few immature members. Among older juvenile and adult females, these trends persisted; additionally, pregnant and lactating females were injured less often than others, but there was no evidence that dominance rank influenced injury rates. For juveniles, injury rates increased similarly with age in both sexes, providing only limited evidence that heightened aggression motivates natal dispersal by males. Most findings supported the hypothesis that aggression increases injury risk. Costs of within-group aggressive competition may manifest in ways other than injury, however, especially for females. We highlight the importance in comparative analyses of considering both within- and between-group aggression and demographic variables as influencing injury risk. <span> </span></p>
