Data from: The application of optical coherence tomography to image subsurface tissue structure of Antarctic krill Euphausia superba
Many small open ocean animals, such as Antarctic krill, are an important part of marine ecosystems. To discover what will happen to animals such as krill in a changing ocean, experiments are run in aquaria where conditions can be controlled to simulate water characteristics predicted to occur in the future. The response of individual animals to changing water conditions can be hard to observe, and with current observation techniques it is very difficult to follow the progress of an individual animal through its life. Optical coherence tomography (OCT) is an optical imaging technique that allows images at high resolution to be obtained from depths up to a few millimeters inside biological specimens. It is compatible with in vivo imaging and can be used repeatedly on the same specimens. In this work, we show how OCT may be applied to post mortem krill samples and how important physiological data such as shell thickness and estimates of organ volume can be obtained. Using OCT we find an average value for the thickness of krill exoskeleton to be (30±4) µm along a 1 cm length of the animal body. We also show that the technique may be used to provide detailed imagery of the internal structure of a pleopod joint and provide an estimate for the heart volume of (0.73±0.03) mm3.
Data from: Optical coherence tomography is highly sensitive in detecting prior optic neuritis
Objective: To explore sensitivity of optical coherence tomography (OCT) in detecting prior unilateral optic neuritis. Methods: Retrospective, observational clinical study of all patients who presented from 1/1/14 to 6/1/17 with unilateral optic neuritis and OCT available at least three months after the attack. We compared OCT retinal nerve fiber layer (RNFL) and ganglion cell inner plexiform layer (GCIPL) thicknesses between affected and unaffected contralateral eyes. We excluded patients with concomitant glaucoma or other optic neuropathies. Based on analysis of normal controls, thinning was considered significant if RNFL was at least 9-µm or GCIPL was at least 6-µm less in affected eye compared to unaffected eye. Results: Fifty-one patients (18 male and 33 female) were included in the study. RNFL and GCIPL thicknesses were significantly lower in eyes with optic neuritis compared to unaffected eyes (p<0.001). RNFL was thinner by ≥9-µm in 73% of optic neuritis eyes compared to the unaffected eye. GCIPL was thinner by ≥6-µm in 96% of optic neuritis eyes. The sensitivity of OCT in detecting prior optic neuritis was 96% when using both average RNFL and GCIPL thicknesses. When using a threshold ≤1st percentile of age-matched controls, sensitivities were 37% for RNFL, 76% for GCIPL, and 78% for either GCIPL or RNFL thicknesses, each of which was lower than those calculated using the inter-eye difference as the threshold (p<0.05). Conclusions: OCT, especially in GCIPL, is a highly sensitive modality in detecting prior optic neuritis, which is made more robust by using inter-eye differences to approximate change.

Photodynamic Ocular Drug Delivery System with Optical Coherence Tomography Oriented Microscale Robots
<p>Retinal diseases are the most common reason for irreversible blindness worldwide. Common retinal diseases, such as diabetic retinopathy, macular degeneration, retinal vein occlusion, and non-infectious uveitis, mostly require intravitreal steroid injections in clinical management. Unfortunately, intravitreal steroid injections lead to other important ocular problems, such as elevation of intraocular pressure, intravitreal infections, and the progression of cataracts. Because of these reasons, targeted drug delivery systems are necessary for the medical treatment of retinal diseases. PHOTodynamic Ocular Drug delivery system with Optical Coherence Tomography Oriented microscale Robots (PHOTODOCTOR) project combines three novel technologies to solve this important clinical problem. The first technological advancement for the project is dexamethasone-saturated hyaluronic acid (HA) hydrogels with organo-ruthenium complexes, which enables controllable degradation under visible light exposure. With the help of visible light-controlled degradation, dexamethasone will be released with the light pulses. Then the hydrogels will be supplemented with superparamagnetic iron oxide nanoparticles (SPIONs), and 3D printed as helical micro-swimmers. Insertion of the SPIONs will help the magnetic actuation of the hydrogel-based microrobots in the intraocular space. Lastly, SPION-supplemented HA hydrogels will be observed with high-resolution optical coherence tomography (OCT) system in the intraocular space to guide them in diseased areas of the retina. While HA-based structure increases penetration in the vitreous and helps the attachment on the retina, SPIONs will increase the visibility of the microrobots in OCT imaging. In this way, a magnetic-driven, visible light-triggered intraocular drug release system with OCT guidance will be produced, and it could be used not only for intravitreal steroid delivery but also for the treatment of retinal tumours and other retinal problems.</p>
