Data from: Non-invasive stratification of non-alcoholic fatty liver disease by whole-transcriptome cell-free mRNA characterization
<p>Hepatic fibrosis stage is the most important determinant of outcomes in patients with non-alcoholic fatty liver disease (NAFLD). There is an urgent need for non-invasive tests that can accurately stage fibrosis and determine efficacy of interventions. Here we describe a novel cf-mRNA-Sequencing approach that can accurately and reproducibly profile low levels of circulating mRNAs and evaluate the feasibility of developing a cf-mRNA-based NAFLD fibrosis classifier. Using separate discovery and validation cohorts with biopsy-confirmed NAFLD (n=176 and 59, respectively) and healthy subjects (n=23), we performed serum cf-mRNA RNA-Seq profiling. Differential expression analysis identified 2498 dysregulated genes between NAFLD and healthy subjects and 134 fibrosis-associated genes in NAFLD patients. Comparison between cf-mRNA and liver tissues transcripts revealed significant overlap of fibrosis associated genes and pathways indicating that the circulating cf-mRNA transcriptome reflects molecular changes in the livers of NAFLD patients. In particular, metabolic and immune pathways reflective of known underlying steatosis and inflammation were highly dysregulated in the cf-mRNA profile of patients with advanced fibrosis. Finally, we used an elastic net ordinal logistic model to develop a classifier that predicts clinically significant fibrosis (F2-4). In an independent cohort, the cf-mRNA classifier was able to identify 50% of patients with at least 90% probability of clinically significant fibrosis. We demonstrate<b> </b>a<b> </b>novel and robust cf-mRNA-based RNA-Seq platform for non-invasive identification of diverse hepatic molecular disruptions and for fibrosis staging with promising potential for clinical trials and clinical practice.</p>
Data from: Nut intake and non-alcoholic fatty liver disease risk
Abstract Objectives: Nut consumption has been associated with a lower risk of type 2 diabetes, metabolic syndrome, and insulin resistance. However, its effect on the risk of non-alcoholic fatty liver disease (NAFLD) is unknown. Therefore, we investigated the relationship between nut consumption and NAFLD risk. Setting and participants: We conducted a retrospective case-control study including 534 patients diagnosed with NAFLD and 534 controls matched by sex and age (± 5) from the Affiliated Nanping First Hospital of Fujian Medical University in China. Main outcome measures: Information on dietary intake was collected using a semi-quantitative food frequency questionnaires and nut consumption was calculated. Nut consumption was categorized using quartiles based on the distribution of daily nut intake of the controls. Binary logistic regression models were used to estimate odds ratio (ORs) and the 95% confidence intervals (CIs) for the association between nut consumption and NAFLD risk. Results: After adjusting for potential confounding variables, nut consumption was not associated with NAFLD risk in the overall sample. When the fully adjusted model was stratified by sex, a significant inverse association was found between high nut consumption and NAFLD only among the men in the highest quartile (OR = 0.43; 95% CI: 0.26–0.71; Ptrend = 0.01). The inverse association of nut consumption with NAFLD risk in men remained significant after controlling for other known or suspected risk factors for NAFLD. Conclusions: Diets with a higher intake of nuts may be associated with a decreased risk of NAFLD, particularly in men.
Data from: Alcohol reduces muscle fatigue through atomistic interactions with nicotinic receptors
Alcohol consumption affects many organs and tissues, including skeletal muscle. However, the molecular mechanism of ethanol action on skeletal muscle remains unclear. Here, using molecular dynamics simulations and single channel recordings, we show that ethanol interacts with a negatively charged amino acid within an extracellular region of the neuromuscular nicotinic acetylcholine receptor (nAChR), thereby altering its global conformation and reducing the single channel current amplitude. Charge reversal of the negatively charged amino acid abolishes the nAChR-ethanol interaction. Moreover, using transgenic animals harboring the charge-reversal mutation, ex vivo measurements of muscle force production show that ethanol counters fatigue in wild type but not homozygous αE83K mutant animals. In accord, in vivo studies of motor coordination following ethanol administration reveal an approximately twofold improvement for wild type compared to homozygous mutant animals. Together, the converging results from molecular to animal studies suggest that ethanol counters muscle fatigue through its interaction with neuromuscular nAChRs.
Experimental data on the physicochemical properties of non- and low-alcohol beers produced with commercial maltose- and maltotriose-negative yeasts
<p>Globally, beer consumption has been stagnating, yet the consumption and popularity of non-alcohol and low-alcoholic beers (NABLAB) is still increasing. NABLAB are mainly produced by removing ethanol from a traditional beer through distillation or nanofiltration. These processes require high investment costs, energy and/or water consumption. An interesting alternative is preventing alcohol production by using alternative yeasts. Much research has been focused on maltose- and/or maltotriose-negative yeasts, and several are commercially available. However, a comparative study between commercial yeasts has not yet been done, making it difficult for a brewer to estimate the impact of these alternative yeasts. Therefore, test beers were made on 20 L-scale with nine commercial NABLAB yeasts. These beers (and used wort samples) were chemically analytically analyzed to compare different commercial yeasts and evaluate the resulting beers. This dataset contains physicochemical properties (alcohol, degree of fermentation, density, extract, color, haze, foam stability, pH, sugars, amino acids, FAN, glycerol, viscosity, bitterness, total polyphenols, higher alcohols, esters, aldehydes, diacetyl, and DMS) of wort and the resulting beers after fermentation with commercial yeasts, including 7 maltose-negative and 2 maltotriose-negative, as well as 2 reference yeasts. All fermentations were carried out as biological duplicates, hence two batches for each yeast. Data are given as the mean of technical replicate analysis, with a distinction made between the biological replicate fermentations. </p>
Stereospecific Hydrogenolysis of Benzylic Alcohols over Pd/C
<p>Hydrogenolysis of tertiary benzylic alcohols on palladium on carbon (Pd/C) generally proceeds with inversion of configuration. However, little is known about the hydrogenolysis mechanism of primary and secondary benzylic alcohols. Literature precedents suggest that these substrates may interact differently with the catalyst. To study the mechanism, we synthesized a pair of deuterated diastereoisomers with a chiral center at the benzylic position. Chemical derivatization of the hydrogenolysis products showed that the reaction proceeds with inversion of configuration for these substrates.</p>
