Three-tiered authentication of herbal Traditional Chinese Medicine ingredients used in women's health provides progressive qualitative and quantitative insight - part 2/4

<p>Traditional Chinese Medicine herbal products are increasingly used in Europe but prevalent authentication methods have significant gaps in detection. In this study, three authentication methods were tested in a tiered approach to improve accuracy on a collection of 51 TCM plant ingredients obtained on the European market. We show the relative performance of conventional barcoding, metabarcoding and standardized chromatographic profiling for TCM ingredients used in one of the most diagnosed disease patterns in women, endometriosis. DNA barcoding using marker ITS2 and chromatographic profiling are methods of choice reported by regulatory authorities and relevant national pharmacopeias. HPTLC was shown to be a valuable authentication tool, combined with metabarcoding, which gives an increased resolution on species diversity, despite dealing with highly processed herbal ingredients. Conventional DNA barcoding as a recommended method was shown to be an insufficient tool for authentication of these samples, while DNA metabarcoding yields an insight into biological contaminants. We conclude that a tiered identification strategy can provide progressive qualitative and quantitative insight in an integrative approach for quality control of processed herbal ingredients.&nbsp;</p>

Leech-derived iDNA complements traditional surveying methods, enhancing species detections for rapid biodiversity sampling in the tropics

<p>Deforestation, exploitation, and other drivers of biodiversity loss in Madagascar leave its highly endangered and predominantly endemic wildlife at risk of extinction. Decreasing biodiversity threatens to compromise ecosystem functions and vital services provided to people. New, economical, and diverse methods of biodiversity monitoring can help to establish reliable baseline and long-term records of species richness. Metabarcoding with invertebrate-derived DNA (iDNA) has emerged as a promising new biosurveillance tool. An unexpected wet forest fragment tucked in the dry cliffs of Madagascar's southcentral plateau, the Ivohibory Protected Area (IPA), hosts a unique mosaic of species diversity, featuring both dry and wet forest species. Recently elevated to protected status, the IPA has been surveyed for flora and fauna with a range of inventory methods over three years and six expeditions (2016, 2017, &amp; 2019). We collected 1,451 leeches over 12 days from the IPA to supplement known species richness and to compare results against current records. With iDNA, we pooled tissues, and isolated, and amplified bloodmeal DNA with five sets of primers. We detected 20 species of which four are species of frogs previously undetected and three of which are previously unknown to exist in this region. iDNA surveys can provide complementary data to traditional surveying methods like camera traps, line transects, and bioacoustic methods.</p>

Does automated reassesment & relearning improve math performance in comparison to traditional computer adaptive practice alone?

<p>Scholarly interest in the notion on how students learn and retain learned skills is vast. Cognitive psychologists have recommended various techniques used to increase retention. One technique, spaced retrieval practice, involves extending opportunities to retrieve course content beyond a customarily short window following the initial learning process. Our preregistered counterbalanced randomised control trial investigated whether an online tutor with an Automated Reassessment and Relearning System (ARRS) improves retention for individual students across specific mathematical skills. While our findings revealed a null result, in part due to the small number of participants, we feel this experimental design could be used in future studies with more favourable results. Additionally, we feel ARRS could be used to support low prior knowledge students and Students With Interrupted or Limited Formal Education (SLIFE), a growing population in schools that is often overlooked in academic research.</p>

Data from: Fallow deer foraging alone does not preserve the vegetation of traditionally sheep-grazed calcareous grasslands

<p>The goal of this study was to evaluate to what extent wild ungulates (fallow deer) can contribute to the maintenance of semi-natural calcareous grasslands, which are a threatened habitat type (natura 2000 code *6210). In a ten-year exclosure experiment we tested the effects of ungulate foraging using three treatments: (A) control with combined foraging of herded sheep and wild fallow deer, (B) sheep exclosure with only deer foraging and (C) total exclosure with no foraging. Treatments not grazed by sheep (B, C) were characterized by significantly declining species numbers, litter accumulation and shrub encroachment. Despite high population densities, the effect of fallow deer alone (B) was weak: Succession of woody species was only partly inhibited, while annuals, short-growing and rosette-building plant species were strongly suppressed by litter accumulation. Only the combination of sheep and fallow deer foraging preserved vegetation structure and species richness and led to a promotion of target species. Synthesis and applications: We conclude that we need to continue the traditional land-use forms such as sheep grazing in order to maintain calcareous grasslands. However, we should also raise our awareness for wild animals and analyse more in depth their potential contribution to the conservation management of open habitats.</p>

The project has co-created a framework and roadmaps for a responsible transition to self-sustaining, digitalised industrial innovation ecosystems. It has worked out the challenges in the interplay between business, academia, government and society to initiate openness, democratic accountability and responsiveness in a process that will in turn promote resilience within these new, digitalised innovation ecosystems. Important attributes for this transition are also gender equality, science education, open access, public engagement, and ethics, which support both organisations and citizens in adapting the transformation that is revolutionising research, industry, economy, &amp; society. Finally, 12 actions, based on the roadmap goals and objectives in each territory have been developed and implemented. Measuring avian bill size: Comparing and evaluating 3D surface scanning with traditional size estimates in Australian birds

<p>Unidimensional measurements for estimating bill size, like length and width, are commonly used in ecology and evolution, but can be criticised due to issues with repeatability and accuracy. Furthermore, formula-based estimates of bill surface area tend to assume uniform bill shapes across species, which is rarely the case. 3D surface scanning can potentially help overcome some such issues by collecting detailed external morphology and direct measurements of surface area, rather than composite estimates of size. Here, we evaluate the use of 3D surface scanners on avian museum specimens to test the repeatability of 3D-based measurements and compare these to traditional formula-based methods of estimating bill size from unidimensional measurements. Using 28 Australian bird species, we investigate inter-observer repeatability of surface area measurements from 3D surface scans. We then compare 3D-based size estimates to formula-based size estimates to infer the accuracy and precision of formula-based measurements of bill surface area. We find that morphometric measurements from 3D surface scans are highly repeatable between observers, without the need for extensive training, demonstrating an advantage over unidimensional measuring methods, like callipers. When comparing 3D-based measurements to formula-based estimates of bill surface area, most formulae for estimating size consistently underestimate surface area, and with considerable variation between species. Where 3D scanning is not possible, we find that a commonly used cone formula for estimating bill size is most precise across diverse bill shapes, therefore supporting its use in interspecific contexts. However, we find that incorporating an additional unidimensional measure of bill curvature into formulae improves the accuracy of the calculated area. Our results reveal the high potential for 3D surface scanners in avian morphometric research, especially for studies necessitating large sample sizes collected by multiple observers, and gives suggestions for formula-based approaches to estimate bill size.</p>

Uncovering the little known impact of a millennia-old traditional use of temperate oak forests: free-ranging domestic pigs markedly change the herb layer, but barely affect the shrub layer

<p>This excel file contains data on ground, herb and shrub layer characteristics of hardwood floodplain forests under different disturbance intensity by free-ranging domestic pigs, a millennia-old practice in Eurasia. We used these dataset to create figures 3, 4, 5, 6 (a) (b), 7, 8 presented in the paper &bdquo;Uncovering the little known impact of a millennia-old traditional use of temperate oak forests: free-ranging pigs markedly change the herb layer, but barely affect the shrub layer&rdquo;. Vegetation surveys were conducted in the spring and late summer periods of 2017 and 2018. The bare soil surface and litter cover were visually estimated within each 0.25 ha sampling plot. The cover and composition of the herb layer were recorded using 30 subsamples (1 m2) at each SP (altogether 1680 subsamples). 28 subsamples were located along the four cardinal directions at 2 m distances running from the centre of sampling plots and 2 other subsamples were randomly positioned. The total cover of the upper (higher than 130 cm) and lower (50-130 cm) shrub layer, and the cover of each species by layers, were visually estimated in each 0.25 ha SP. The browsing, rooting and rubbing impacts of pigs were measured along a 56-m-long transect running through the centre of the SPs from north to south. We sampled altogether from 3138 to 3282 woody specimens depending on variables used to describe pigs impacts on specimens.</p>
