Species-specific effects of thermal stress on the expression of genetic variation across a diverse group of plant and animal taxa under experimental conditions

<p>Assessing the genetic adaptive potential of populations and species is essential for better understanding evolutionary processes. However, the expression of genetic variation may depend on environmental conditions, which may speed up or slow down evolutionary responses. Thus, the same selection pressure may lead to different responses. Against this background, we here investigate the effects of thermal stress on genetic variation, mainly under controlled laboratory conditions. We estimated additive genetic variance (<i>V<sub>A</sub></i>), narrow-sense heritability (<i>h</i><sup>2</sup>), and the coefficient of genetic variation (<i>CV<sub>A</sub></i>) under both benign control and stressful thermal conditions. We included six species spanning a diverse range of plant and animal taxa and a total of 25 morphological and life-history traits. Our results show that (1) thermal stress reduced fitness components, (2) the majority of traits showed significant genetic variation, and that (3) thermal stress affected the expression of genetic variation (<i>V<sub>A</sub></i>, <i>h</i><sup>2</sup> or <i>CV<sub>A</sub></i>) in only one third of the cases (25 of 75 analyses, mostly in one clonal species). Moreover, effects were highly species-specific, with genetic variation increasing in 11 and decreasing in 14 cases under stress. Our results hence indicate that thermal stress does not generally affect the expression of genetic variation under laboratory conditions but, nevertheless, increases or decreases genetic variation in specific cases. Consequently, predicting the rate of genetic adaptation might not be generally complicated by environmental variation, but requires a careful case-by-case consideration.</p>

Data from: Increased survival of experimentally evolved antimicrobial peptide-resistant Staphylococcus aureus in an animal host

Antimicrobial peptides (AMPs) have been proposed as new class of antimicrobial drugs, following the increasing prevalence of bacteria resistant to antibiotics. Synthetic AMPs are functional analogues of highly evolutionarily conserved immune effectors in animals and plants, produced in response to microbial infection. Therefore, the proposed therapeutic use of AMPs bears the risk of 'arming the enemy': bacteria that evolve resistance to AMPs may be cross-resistant to immune effectors (AMPs) in their hosts. We used a panel of populations of Staphylococcus aureus that were experimentally selected for resistance to a suite of individual AMPs and antibiotics to investigate the 'arming the enemy' hypothesis. We tested whether the selected strains showed higher survival in an insect model (Tenebrio molitor) and cross-resistance against other antimicrobials in vitro. A population selected for resistance to the antimicrobial peptide iseganan showed increased in vivo survival, but was not more virulent. We suggest that increased survival of AMP-resistant bacteria almost certainly poses problems to immune-compromised hosts.

Data from: Revisiting Brownian motion as a description of animal movement: a comparison to experimental movement data

Characterization of patterns of animal movement is a major challenge in ecology with applications to conservation, biological invasions and pest monitoring. Brownian random walks, and diffusive flux as their mean field counterpart, provide one framework in which to consider this problem. However, it remains subject to debate and controversy. This study presents a test of the diffusion framework using movement data obtained from controlled experiments. Walking beetles (Tenebrio molitor) were released in an open circular arena with a central hole and the number of individuals falling from the arena edges was monitored over time. These boundary counts were compared, using curve fitting, to the predictions of a diffusion model. The diffusion model is solved precisely, without using numerical simulations. We find that the shape of the curves derived from the diffusion model is a close match to those found experimentally. Furthermore, in general, estimates of the total population obtained from the relevant solution of the diffusion equation are in excellent agreement with the experimental population. Estimates of the dispersal rate of individuals depend on how accurately the initial release distribution is incorporated into the model. We therefore show that diffusive flux is a very good approximation to the movement of a population of Tenebrio molitor beetles. As such, we suggest that it is an adequate theoretical/modelling framework for ecological studies that account for insect movement, although it can be context specific. An immediate practical application of this can be found in the interpretation of trap counts, in particular for the purpose of pest monitoring.
