Molecular Probes for Tracking Lipid Droplet Membrane Dynamics
<p><strong><span>Abstract</span></strong></p> <p><span>Lipid droplets (LDs) and their membrane proteins play crucial roles in lipid metabolism, signaling, and information transport within cells. LDs feature a unique monolayer lipid membrane that has not been extensively studied due to the lack of suitable molecular probes that are able to distinguish this membrane from the LD lipid core. In this work, we present a three-pronged molecular probe design strategy that combines lipophilicity-based organelle targeting with microenvironment-dependent activation. As a proof-of-concept, we designed an <u>LD</u> <u>m</u>embrane labeling pro-probe called<strong> LDM</strong>, which selectively localizes around LD membranes. Upon activation by the HClO/ClO</span><sup><span>−</span></sup><span> microenvironment that surrounds LDs, <strong>LDM</strong> pro-probe undergoes a color change and releases<strong> LDM-OH</strong> probe that binds to LD membrane proteins. This localizes the probe to the LD-as</span><span>sociated protein space which is restricted to the membrane thus enabling visualization of the ring-like LD membrane. By utilizing<strong> LDM</strong>, we identified the dynamic mechanism of LD membrane contacts and their protein accumulation parameters. Furthermore, using <strong>LDM</strong> in liver cancer cells allowed us to examine the changes in LD/mitochondrial protein accumulation caused by the state of starvation these cells encounter. This led to the discovery that liver cancer cells respond to energy stress during hunger by enhancing LD-mitochondria interactions. <p>This file contains the basic profiles and time series of cloud-field properties, cloud organization metrics, and cold-pool properties associated with the project titled "Cold pools mediate mesoscale adjustments of trade-cumulus fields to changes in cloud-droplet number concentration".</p> <p>The numbers of simulations 104, 105, 1, 106, 107, and 108 are associated with the cloud-droplet number concentrations Nc of 20, 50, 70, 100, 200, and 1000 /cm3, respectively.</p> <p>The character "-cnstsun" at the end of a file name is related to simulations without the diurnal cycle of solar radiation.</p> <p>The movie (Nc20_Nc1000.mp4) shows the evolution of cloud albedo (1st column), mixed-layer height hmix (2nd column), total moisture anomaly qt' at the 200-m level (3rd column), and the vertical velocity w at the 200-m level (4th column) for simulations with Nc of 20 (1st row) and 1000 (2nd row) /cm3 and without the the diurnal cycle. </p>

<p>Contains the design files and source data linked to the following publication in Cell Reports Methods (2023, https://doi.org/10.1016/j.crmeth.2023.100478)</p> <p><strong><em>iSort </em></strong><strong>enables automated complex microfluidic droplet sorting in an effort to democratize technology.</strong></p> <p><strong>Abstract: </strong><br> Fluorescence activated droplet sorting (FADS) is a widely used microfluidic technique for high throughput screening. However, it requires highly trained specialists to determine optimal sorting parameters, resulting in a large combinatorial space that is challenging to optimize systematically. Additionally, there is currently no way to track every single droplet within a screen, leading to compromised sorting and “hidden” false positive events. To overcome all these limitations, we have developed a setup in which the droplet frequency, spacing and trajectory at the sorting junction are monitored in real time using impedance analysis. The resulting data is used to continuously optimize all parameters automatically and to counteract perturbations, resulting in higher throughput, higher reproducibility, increased robustness and a beginner-friendly character. We believe this provides a missing piece for the spreading of phenotypic single-cell analysis methods, similar to what we have seen for single-cell genomics platforms.</p> <p><strong>To cite:</strong> https://doi.org/10.1016/j.crmeth.2023.100478</p>

<p>Super-droplet simulations of a one-dimensional rain shaft model, generated by using the Python-based code PySDM v2.15. The updraft speed and the temperature profile are prescribed, as described in<br> "Shipway, B. J., & Hill, A. A. (2012). Diagnosis of systematic differences between multiple parametrizations of warm rain microphysics using a kinematic framework. Quarterly Journal of the Royal Meteorological Society, 138(669), 2196-2211".<br> The data were generated for five surface pressure levels, ranging from 988 hPa to 1012 hPa, with updraft amplitudes ranging from 1 to 4 m/s and an initial aerosol number density varying from 10 to 1000 per cubic centimeter.</p>
