<p>This record contains data related to article “Dose-Dependent Impairment of the Immune Response to the Moderna-1273 mRNA Vaccine by Mycophenolate Mofetil in Patients with Rheumatic and Autoimmune Liver Diseases”</p> <p>The purpose of this study was to evaluate the efficacy and safety of the Moderna-1273 mRNA vaccine for SARS-CoV-2 in patients with immune-mediated diseases under different treatments. Anti-trimeric spike protein antibodies were tested in 287 patients with rheumatic or autoimmune diseases (10% receiving mycophenolate mofetil, 15% low-dose glucocorticoids, 21% methotrexate, and 58% biologic/targeted synthetic drugs) at baseline and in 219 (76%) 4 weeks after the second Moderna-1273 mRNA vaccine dose. Family members or caretakers were enrolled as the controls. The neutralizing serum activity against SARS-CoV-2-G614, alpha, and beta variants in vitro and the cytotoxic T cell response to SARS-CoV-2 peptides were determined in a subgroup of patients and controls. Anti-SARS-CoV-2 antibody development, i.e., seroconversion, was observed in 69% of the mycophenolate-treated patients compared to 100% of both the patients taking other treatments and the controls (<em>p</em> &lt; 0.0001). A dose-dependent impairment of the humoral response was observed in the mycophenolate-treated patients. A daily dose of &gt;1 g at vaccination was a significant risk factor for non-seroconversion (ROC AUC 0.89, 95% CI 0.80-98, <em>p</em> &lt; 0.0001). Moreover, in the seroconverted patients, a daily dose of &gt;1 g of mycophenolate was associated with significantly lower anti-SARS-CoV-2 antibody titers, showing slightly reduced neutralizing serum activity but a comparable cytotoxic response compared to other immunosuppressants. In non-seroconverted patients treated with mycophenolate at a daily dose of &gt;1 g, the cytotoxic activity elicited by viral peptides was also impaired. Mycophenolate treatment affects the Moderna-1273 mRNA vaccine immunogenicity in a dose-dependent manner, independent of rheumatological disease.</p> <p>.</p>
High-resolution African HLA resource uncovers HLA-DRB1 expression effects underlying vaccine response: summary statistics
<p>How human genetic variation contributes to vaccine immunogenicity and effectiveness is unclear, particularly in infants from Africa. We undertook genome-wide association analyses of eight vaccine antibody responses in 2,499 infants from three African countries and identified significant associations across the human leukocyte antigen (HLA) locus for five antigens spanning pertussis, diphtheria and hepatitis B vaccines. Using high-resolution HLA typing in 1,706 individuals from 11 African populations we constructed a continental imputation resource to fine-map signals of association across the class II HLA observing genetic variation explaining up to 10% of the observed variance in antibody responses. Using follicular helper T-cell assays, <em>in silico</em> binding, and immune cell eQTL datasets we find evidence of <em>HLA-DRB1</em> expression correlating with serological response and inferred protection from pertussis following vaccination. This work improves our understanding of molecular mechanisms underlying HLA associations that should support vaccine design and development across Africa with wider global relevance.</p>
CD4+ and CD8+ T cell responses to peptides covering SARS-CoV-2 Spike in response to mRNA vaccination in persons recovered from SARS-CoV-2 infection
<p>These files contain intracellular cytokine staining flow cytometry data for CD4+ and CD8+ T cells after exposure of serial PBMC to SARS-CoV-2 spike peptides or control antigens. PBMC are from subjects recovered from SARS-CoV-2 infection that subsequently received mRNA vaccination. The data were analyzed and exported from FlowJo version 10 as individual gated events and related Boolean subsets for four functional markers: CD40L, IFN-g, IL-2, and TNF-a. Counts and frequencies are both included.</p> <p> </p> <p><strong>Procedure for obtaining data:</strong></p> <p>Cryopreserved peripheral blood mononuclear cells (PBMC) were thawed and rested overnight. PBMC (1 x 10<sup>6</sup> per well) were stimulated with a SARS-CoV-2 spike overlapping peptide pool (JPT, 1 μg/mL each peptide; 0.4% final DMSO concentration), 0.4% DMSO as a negative control, or PHA-P (Remel; 1.6 μg/mL final concentration) as a positive control, in the presence of anti-CD28 and anti-CD49d (BD Biosciences) antibodies at 37°C for 6 hours. Brefeldin A (Sigma) was added after 2 hours. Cells were stained with Live/Dead Near IR dye (Invitrogen), treated with FACS lyse (BD Biosciences) and frozen at -80<sup>o</sup>C. For staining, the cells were thawed, washed, and permeabilized with Permeabilizing solution 2 (BD Biosciences) then stained with fluorochrome labeled monoclonal antibodies to anti-human CD3 mAb (clone UCHT) conjugated with PE-Texas Red (ECD; Beckman Coulter), anti-human CD4 mAb (clone SK3) conjugated with BV510 (Biolegend), anti-human CD8 mAb (clone SK1) conjugated with PerCP-Cy5.5 (BD Biosciences), and the activation markers anti-human CD40L mAb (clone TRAP1) conjugated with BV421 (BD Biosciences), anti-human IFNγ mAb (clone 4S.B3) conjugated with PE (BD Biosciences), anti-human IL2 mAb (clone MQ1-17H12) conjugated with APC (BD Biosciences), and anti-human TNFα mAb (clone MAb11) conjugated with FITC (BD Biosciences). Events were recorded with BD Fortessa and analyzed with FlowJo (v10 for Mac; BD). </p>
