Data from: Host plant-related genomic differentiation in the European cherry fruit fly, Rhagoletis cerasi (L., 1758) (Diptera: Tephritidae)
<p>Elucidating the mechanisms and conditions facilitating the formation of biodiversity are central topics in evolutionary biology. A growing number of studies imply that divergent ecological selection may often play a critical role in speciation by counteracting the homogenising effects of gene flow. Several examples involve phytophagous insects, where divergent selection pressures associated with host plant shifts may generate reproductive isolation, promoting speciation. Here, we use ddRADseq to assess the population structure and to test for host-related genomic differentiation in the European cherry fruit fly, Rhagoletis cerasi (L., 1758) (Diptera: Tephritidae). This tephritid is distributed throughout Europe and western Asia, and has adapted to two different genera of host plants, Prunus spp. (cherries) and Lonicera spp. (honeysuckle). Our data imply that geographic distance and geomorphic barriers serve as the primary factors shaping genetic population structure across the species range. Locally, however, flies genetically cluster according to host plant, with consistent allele frequency differences displayed by a subset of loci between Prunus and Lonicera flies across four sites surveyed in Germany and Norway. These 17 loci display significantly higher FST values between host plants than others. They also showed high levels of linkage disequilibrium within and between Prunus and Lonicera flies, supporting host-related selection and reduced gene flow. Our findings support the existence of sympatric host races in R. cerasi embedded within broader patterns of geographic variation in the fly, similar to the related apple maggot, Rhagoletis pomonella, in North America.</p>

Data from: Detection of the endangered European weather loach (Misgurnus fossilis) via water and sediment samples: testing multiple eDNA workflows.
<p>The European weather loach (<i>Misgurnus fossilis</i>) is classified as highly endangered in several countries of Central Europe. Populations of <i>M. fossilis</i> are predominantly found in ditches with low water levels and thick sludge layers and are thus hard to detect using conventional fishing methods. Therefore, environmental DNA (eDNA) monitoring appears particularly relevant for this species. In previous studies, <i>M. fossilis</i> was surveyed following eDNA water sampling protocols, which were not optimized for this species. Therefore, we created two full factorial study designs to test six different eDNA workflows for sediment samples and twelve different workflows for water samples. We used qPCR to compare the Threshold cycle (Ct) values of the different workflows, which indicate the target DNA amount in the sample, and spectrophotometry to quantify and compare the total DNA amount inside the samples. We analyzed 96 water samples and 48 sediment samples from a pond with a known population of <i>M. fossilis</i>. We tested several method combinations for long-term sample preservation, DNA capture and DNA extraction. Additionally, we analyzed the DNA yield of samples from a ditch with a natural <i>M. fossilis</i> population monthly over one year to determine the optimal sampling period. Our results showed that the long-term water preservation method commonly used for eDNA surveys of <i>M. fossilis </i>did not lead to optimal DNA yields, and we present a valid long-term sample preservation alternative. A cost-efficient high salt DNA extraction led to the highest target DNA yields and can be used for sediment and water samples. Furthermore, we were able to show that in a natural habitat of <i>M. fossilis</i>, total and target eDNA were higher between June and September, which implies that this period is favorable for eDNA sampling. Our results will help to improve the reliability of future eDNA surveys of <i>M. fossilis</i>.</p>

Pistil length and width, pollen diameter, pollen tube elongations, fruitset, seedset and germination rates in european invasive populations of Ludwigia grandiflora subsp. hexapetala in the two floral morphs
<p>Mating system influences local population genetic structure, effective size, offspring fitness and functional variation. Determining the respective importance of self- and cross-fertilization in hermaphroditic flowering plants is thus important to understand their ecology and evolution. The worldwide invasive species, <em>Ludwigia </em><em>grandiflora</em> subsp. <em>hexapetala </em>(<em>Lgh</em>) presents two floral morphs: one self-compatible short-styled morph (S-morph) and one self-incompatible long-styled morph (L-morph). Most invasive populations worldwide are only composed of self-incompatible L-morphs, which questions the importance of sexual reproduction during the invasion. In this study, we identified the mating systems of western European experimental and natural populations of <em>Lgh </em>by comparing structural characteristics of pollen and style, by studying self- and cross-pollen tube elongations and the viability of the resulting seeds and seedlings in both morphs. The dataset provides measures of pistil length and width; pollen diameter; pollen tube elongations along time after pollination; fruitset, seedset and germination rate in in situ monomorphic L-morph and S-morph populations of western europe invasive <em>Lgh</em>.
