Decoupling silicon metabolism from carbon and nitrogen assimilation poises diatoms to exploit episodic nutrient pulses in a coastal upwelling system
<p>Diatoms serve as the major link between the marine carbon (C) and silicon (Si) biogeochemical cycles through their contributions to primary productivity and requirement for Si during cell wall formation. Although several culture-based studies have investigated the molecular response of diatoms to Si and nitrogen (N) starvation and replenishment, diatom silicon metabolism has been understudied in natural populations. A series of deckboard Si-amendment incubations were conducted using surface water collected in the California Upwelling Zone near Monterey Bay. Steep concentration gradients in macronutrients in the surface ocean coupled with substantial N and Si utilization led to communities with distinctly different macronutrient states: replete ('healthy'), low N ('N-stressed'), and low N and Si ('N- and Si-stressed'). Biogeochemical measurements of Si uptake combined with metatranscriptomic analysis of communities incubated with and without added Si were used to explore the underlying molecular response of diatom communities to different macronutrient availability. Metatranscriptomic analysis revealed that N-stressed communities exhibited dynamic shifts in N and C transcriptional patterns suggestive of compromised metabolism. Expression patterns in communities experiencing both N and Si stress imply that the presence of Si stress may partially ameliorate N stress and dampen the impact on organic matter metabolism. This response builds upon previous observations that the regulation of C and N metabolism is decoupled from Si limitation status, where Si stress allows the cell to optimize the metabolic machinery necessary to respond to episodic pulses of nutrients. Several well-characterized Si-metabolism associated genes were found to be poor molecular markers of Si physiological status; however, several uncharacterized Si-responsive genes were revealed to be potential indicators of Si stress or silica production.</p>
Decoupling of Spin Decoherence Paths near Zero Magnetic Field
<p>We demonstrate a method to quantify and manipulate nuclear spin decoherence mechanisms that are active in zero to ultralow magnetic fields. These include (i) nonadiabatic switching of spin quantization axis due to residual background fields and (ii) scalar pathways due to through-bond couplings between <sup>1</sup>H and heteronuclear spin species, such as <sup>2</sup>H used partially as an isotopic substitute for <sup>1</sup>H. Under conditions of free evolution, scalar relaxation due to <sup>2</sup>H can significantly limit nuclear spin polarization lifetimes and thus the scope of magnetic resonance procedures near zero field. It is shown that robust trains of pulsed dc magnetic fields that apply π flip angles to one or multiple spin species may switch the effective symmetry of the nuclear spin Hamiltonian, imposing decoupled or coupled dynamic regimes on demand. The method should broaden the spectrum of hyperpolarized biomedical contrast-agent compounds and hyperpolarization procedures that are used near zero field.</p> <p>Entry contains processed experimental data for Figures 4-5 of the main paper, and Figures 2-5 of the Supporting Information.</p>

Code and data for: Decoupling channel count from field-of-view and spatial resolution in single-sensor imaging systems for fluorescence image-guided surgery
<p><em>Significance</em></p> <p>Near-infrared fluorescence image-guided surgery is often thought of as a spectral imaging problem where the channel count is the critical parameter, but it should also be thought of as a multiscale imaging problem where the field-of-view and spatial resolution are similarly important.</p> <p><em>Aim</em></p> <p>Conventional imaging systems based on division-of-focal-plane architectures suffer from a strict relationship between the channel count on one hand and the field-of-view and spatial resolution on the other, but bioinspired imaging systems that combine stacked photodiode image sensors and long-pass/short-pass filter arrays offer a weaker tradeoff.</p> <p><em>Approach</em></p> <p>In this paper, we explore how the relevant changes to the image sensor and associated image processing routines affect image fidelity during image-guided surgeries for tumor removal in an animal model of breast cancer and nodal mapping in women with breast cancer.</p> <p><em>Results</em></p> <p>We demonstrate that a transition from a conventional imaging system to a bioinspired one, along with optimization of the image processing routines, yields improvements in multiple measures of spectral and textural rendition relevant to surgical decision-making.</p> <p><em>Conclusions</em></p> <p>These results call for a critical examination of the devices and algorithms that underpin image-guided surgery to ensure that surgeons receive high-quality guidance and patients receive high-quality outcomes as these technologies enter clinical practice.</p>

Data for: Decoupling of sexual signals and their underlying morphology facilitates rapid phenotypic diversification
<p>How novel phenotypes evolve is challenging to imagine because traits are often underlain by numerous integrated phenotypic components, and changes to any one form can disrupt the function of the entire module. Yet novel phenotypes do emerge, and research on adaptive phenotypic evolution suggests that complex traits can diverge while either maintaining existing form-function relationships or through innovations that alter form-function relationships. How these alternate routes contribute to sexual signal evolution is poorly understood, despite the role of sexual signals in generating biodiversity. In Hawaiian populations of the Pacific field cricket, male song attracts both female crickets and a deadly acoustically orienting parasitoid fly. In response to this conflict between natural and sexual selection, male crickets have evolved altered wing morphologies multiple times, resulting in loss and dramatic alteration of sexual signals. More recently, we and others have observed a radical increase in sexual signal variation and the underlying morphological structures that produce song. We conducted the first combined analysis of form (wing morphology), function (emergent signal), and receiver responses to characterize novel variation, test alternative hypotheses about form-function relationships (Form-Function Continuity vs. Form-Function Decoupling) and investigate underlying mechanistic changes and fitness consequences of novel signals. We identified three sound-producing male morphs (one previously undescribed, named "rattling") and found that relationships between morphology and signals have been rewired (Form-Function Decoupling), rapidly and repeatedly, through the gain, loss, and alteration of morphological structures, facilitating the production of signals that exist in novel phenotypic space. By integrating across a hierarchy of phenotypes, we uncovered divergent morphs with unique solutions to the challenge of attracting mates while evading fatal parasitism.</p>

Growth Chamber mesocosm experiment to assess the effects of the OSS decoupled from the presence of G. senegalensis (PRJNA930014)
<p>The Sahel region of West Africa is a vulnerable eco-region where a growing population and climate-change induced drought threaten food security. The subsistence farmers here grow pearl millet (Pennisetum glaucum) without fertilizers or irrigation. Local and biologically-based means of maintaining yields are needed, and an agroforestry system in Senegal - the Optimized Shrub-intercropping System (OSS) - provides a solution. In the OSS, the shrub Gueira senegalensis performs hydraulic lift, distributing deep subsurface water to neighboring millet plants. The shrub also supports a distinct microbial community and significantly improves carbon storage and nutrient dynamics. Here, we aimed to test whether shrub-impacted soils differed in microbiome and millet outcomes under simulated early-season drought in a growth chamber. Shrub impact was separated into residual impacts on microbiome and soil, versus ongoing shrub-derived organic matter (OM) input. decoupled from the effects of the growing shrub. Methods: We characterized the microbiota through dry-down and rewetting periods, with particular attention to lineages with known plant growth promoting (PGP) properties, via amplicon sequencing of the 16S rRNA gene V3-V4 region and the ITS2, . Results: Both bacterial and fungal communities were significantly altered by imposed drought, OM amendment, and original soil type (+/-OSS). The largest significant bacterial community impact under dry down occurred for +shrub/-OM treatments, and under rewetting for -OM treatment regardless of +/- OSS. Known bacterial PGP lineages were only enriched under drought in +OSS/-OM treatments. The fungal community behaved differently with a significant dry-down impact only in +OSS/+OM treatments, while rewetting enriched for fungal pathogens but only in -OSS/+OM soils. Decoupled from ongoing shrub growth, both residual shrub impacts and shrub OM inputs altered microbiota and increased millet biomass under drought. These results are part of a growing body of work aimed at understanding microbiome roles in increasing ecological resilience and combating food insecurity.
