Data and R code for: Survival to weaning in arid-country vervet monkeys

<p>We addressed the relative contributions of maternal rank and sociability to the survival of infant vervet monkeys (Chlorocebus pygerythrus) to nutritional independence (~210 days)&nbsp;in a resource-poor environment. Data from 153 infants across three troops and 10 birth cohorts indicated a pre-weaning mortality of 30% (Range: 9%-85%), with a median age at death of 50 days. In addition to the consequences of resource availability, increased&nbsp;infant survival was independently and equivalently associated with higher maternal rank&nbsp;and a greater number of maternal spatial partners. We use this outcome to suggest that apparent discrepancies in the relative importance of different maternal attributes in determining reproductive outcomes may be resolved by considering more closely local&nbsp;&nbsp;sources of infant mortality.</p>

Focal vs. faecal: Seasonal variation in the diet of wild vervet monkeys from observational and DNA metabarcoding data

<p>1. Assessing the diet of wild animals reveals valuable information about their ecology and trophic relationships that may help elucidate dynamic interactions in ecosystems and forecast responses to environmental changes.</p> <p>2. Advances in molecular biology provide valuable research tools in this field. However, comparative empirical research is still required to highlight strengths and potential biases of different approaches. Therefore, this study compares environmental DNA and observational methods for the same study population and sampling duration.</p> <p>3. We employed DNA metabarcoding assays targeting plant and arthropod diet items in 823 faecal samples collected over 12 months in a wild population of an omnivorous primate, the vervet monkey (<em>Chlorocebus pygerythrus</em>). DNA metabarcoding data were subsequently compared to direct observations.</p> <p>4. We observed the same seasonal patterns of plant consumption with both methods, however, DNA metabarcoding showed considerably greater taxonomic coverage and resolution compared to observations, mostly due to the construction of a local plant DNA database. We found a strong effect of season on variation in plant consumption largely shaped by the dry and wet seasons. The seasonal effect on arthropod consumption was weaker but feeding on arthropods was more frequent in spring and summer, showing overall that vervets adapt their diet according to available resources. The DNA metabarcoding assay outperformed also direct observations of arthropod consumption in both taxonomic coverage and resolution.</p> <p>5. Combining traditional techniques and DNA metabarcoding data can therefore not only provide enhanced assessments of complex diets or reveal trophic interactions to the benefit of wildlife conservationists and managers but also opens new perspectives for behavioural ecologists studying whether diet variation in social species is induced by environmental differences or might reflect selective foraging behaviours.</p>

Climate induced stress and mortality in vervet monkeys

As the effects of global climate change become more apparent, animal species will become increasingly affected by extreme climate and its effect on the environment. There is a pressing need to understand animal physiological and behavioural responses to climatic stressors. We used the reactive scope model as a framework to investigate the influence of drought conditions on vervet monkey (Chlorocebus pygerythrus) behaviour, physiological stress and survival across 2.5-years in South Africa. Data were collected on climatic, environmental and behavioural variables and physiological stress via faecal glucocorticoid metabolites (fGCMs). There was a meaningful interaction between water availability and resource abundance: when food availability was high but standing water was unavailable, fGCMs were higher compared to when food was abundant and water was available. Vervet monkeys adapted their behaviour during a drought period by spending a greater proportion of time resting at the expense of feeding, moving, and social behaviour. As food availability decreased, vervet mortality increased. Peak mortality occurred when food availability was at its lowest and there was no standing water. A survival analysis revealed that higher fGCMs were associated with an increased probability of mortality. Our results suggest that with continued climate change, the increasing prevalence of drought will negatively affect vervet abundance and distribution in our population. Our study contributes to knowledge of the limits and scope of behavioural and physiological plasticity among vervet monkeys in the face of rapid environmental change.

mRNA expression analysis of the hippocampus in a Vervet monkey model of FASD

<p>The vervet monkey (<i>Chlorocebus </i>aethiops) has proven to be an invaluable tool for researching voluntary alcohol ingestion and the sequelae that can arise from such behaviour. In this study, a vervet monkey model for fetal alcohol spectrum disorder was generated by providing a cohort of alcohol preferring, pregnant dams the option to ingest alcohol between gestational days 90-165 with a corresponding sucrose matched control group. Subsequently, gene expression analysis of the hippocampus was contrasted at 5 months and 2 years using the GeneChip Rhesus Macaque Genome Array in a 2x2 study design which interrogated two independent variables, Age and Alcohol consumption. The analysis identified a global downregulation of expression when interrogating Alcohol as a main effect with a relative balance of upregulation and downregulation using Age as a main effect. Functional annotation of both independent variables was performed with Alcohol generating broad functional annotation clusters which could implicate an epigenetic role in downregulation, while Age reliably produced functional annotation clusters predominantly related to development. Furthermore, our data reveal a novel connection between <i>EFNB1</i> and FASD which is highly plausible given its role in development as well as its central role in craniofrontal nasal syndrome (CFNS).</p>
