Disease and lesion maps - part of the Scientific opinion on the evaluation of public and animal health risks in case of a delayed post-mortem inspection in ungulates
<p>EFSA was requested to assess the impact on effectiveness of <em>post-mortem</em> inspection in terms of any change in the sensitivity of detection of animal diseases of domestic and wild ungulates listed according to Article 5 of Regulation (EC) No 2016/429 and septicaemia, pyaemia, toxaemia or viraemia, when carried out after up to 24 hours or up to 72 hours after slaughter in comparison to when it is carried out immediately after slaughter. In order to identify the main lesions associated with the target diseases, a so called “disease map” was built, with the information about the clinical forms of the diseases (acute, subacute, chronic, or latent forms) and clinical signs, as well as potential lesions that could be observed on the respective diseased animals.</p> <p>The following information is indicated for each disease/condition in the disease map:</p> <ol> <li>The susceptible animal species</li> <li>Whether there is any surveillance programme in place in the EU</li> <li>The signs associated with the disease that could be detected at <em>ante mortem </em>inspection;</li> <li>The lesions associated with the disease that could be detected at <em>post mortem</em> inspection</li> <li>The probability of detecting the disease during the PMI as normally carried out.</li> <li>Whether carcass swabbing and/or laboratory tests are normally carried out.</li> </ol> <p>From the disease map, the list of organs to be considered at <em>post mortem</em> inspection and the lesions, a “lesion map” was built by connecting animal species with organs, lesions and corresponding disease. This was done to facilitate the construction of a questionnaire where, for each organ and the five types of lesions, the respondents (meat inspectors) had to provide a numerical answer about how many carcasses out of 100 with the given lesion, will be still detected after 24- or 72-h of refrigerated storage.</p>

Enhanced mTORC1 signaling and protein synthesis in pathologic alpha-synuclein cellular and animal models of Parkinson's disease
<p>Pathologic alpha-synuclein plays an important role in the pathogenesis of alpha-synucleinopathies such as Parkinson's disease (PD). Disruption of proteostasis is thought to be central to pathologic alpha-synuclein (alpha-syn) toxicity; however, the molecular mechanism of this deregulation is poorly understood. Here we report that pathologic alpha-syn activates the mechanistic target of rapamycin (mTOR) complex 1 (mTORC1) leading to enhanced mRNA translation via binding tuberous sclerosis protein (TSC) 2 and destabilizing the TSC1-TSC2 complex. Genetic and pharmacologic inhibition of mTOR and protein synthesis rescue the dopamine neuron loss, behavioral deficits, and aberrant biochemical signaling in the alpha-syn preformed fibril (PFF) and Drosophila alpha-syn transgenic models of pathologic alpha-syn induced degeneration. Our findings establish a potential molecular mechanism by which pathologic alpha-syn activates mTORC1 leading to enhanced protein synthesis and concomitant neurodegeneration in PD.</p>

The effects of exploratory behavior on physical activity in a common animal model of human disease, zebrafish (Danio rerio)
<p>Zebrafish (Danio rerio) are widely accepted as a multidisciplinary vertebrate model for neurobehavioral and clinical studies, and more recently have become established as a model for exercise physiology and behavior. Individual differences in activity level (e.g., exploration) have been characterized in zebrafish, however, how different levels of exploration correspond to differences in motivation to engage in swimming behavior has not yet been explored. We screened individual zebrafish in two tests of exploration: the open field and novel tank diving tests. The fish were then exposed to a tank in which they could choose to enter a compartment with a flow of water (as a means of testing voluntary motivation to exercise). After a 2-day habituation period, behavioral observations were conducted. We used correlative analyses to investigate the robustness of the different exploration tests. Due to the complexity of dependent behavioral variables, we used machine learning to determine the personality variables that were best at predicting swimming behavior. Our results show that contrary to our predictions, the correlation between novel tank diving test variables and open field test variables was relatively weak. Novel tank diving variables were more correlated with themselves than open field variables were to each other. Males exhibited stronger relationships between behavioral variables than did females. In terms of swimming behavior, fish that spent more time in the swimming zone spent more time actively swimming, however, swimming behavior was inconsistent across the time of the study. All relationships between swimming variables and exploration tests were relatively weak, though novel tank diving test variables had stronger correlations. Machine learning showed that three novel tank diving variables (entries top/bottom, movement rate, average top entry duration) and one open field variable (proportion of time spent frozen) were the best predictors of swimming behavior, demonstrating that the novel tank diving test is a powerful tool to investigate exploration. Increased knowledge about how individual differences in exploration may play a role in swimming behavior in zebrafish is fundamental to their utility as a model of exercise physiology and behavior.</p>
