<p>Salivary glands (SGs) represent a permissive site for several sialotropic viruses whose persistence is linked to the development of autoimmunity. Natural Killer (NK) cells play a key role in viral clearance but their involvement in viral infection control and in tertiary lymphoid structures (TLS) development within SGs is unknown. By using an inducible model of TLS in the SGs of wild-type C57BL/6 mice, induced by the local delivery of a replication-defective adenovirus (AdV), we demonstrated that circulating NK cells are rapidly recruited to SGs and highly enrich the early inflammatory infiltrate prior to TLS development. NK cells migrating to SGs in response to AdV infection up-regulate NKp46, undergo proliferation, acquire cytotoxic potential, produce Granzyme-B and IFN-γ, and reduce viral load in the acute phase of the infection. Nonetheless, the selective depletion of both circulating and infiltrating NK cells in AdV-infected mice neither affect the development and frequency of TLS nor the onset of autoimmunity. These data demonstrate that, upon local viral delivery of AdV, peripheral NK cells homing to SGs can exert an early control of the viral infection but are dispensable for the formation of TLS and breach of immunologic tolerance.</p> <p> </p> <p>This research used pzf and ets file form extensions, we attach pdfs information about</p>
Extracellular Vesicles from Pneumocystis carinii-infected rats impair fungal viability but are dispensable for macrophage functions
<p><em>Pneumocystis</em> spp. are host obligate fungal pathogens that can cause severe pneumonia in mammals and rely heavily on their host for essential nutrients. The lack of a sustainable <em>in vitro</em> culture system poses challenges in understanding their metabolism and the acquisition of essential nutrients from host lungs remains unexplored.</p> <p>Transmission electron micrographs show Extracellular Vesicles (EVs) are found near <em>Pneumocystis</em>spp. within the lung. We hypothesized that EVs transport essential nutrients to the fungi during infection. To investigate this, EVs from <em>P. carinii</em>- and <em>P. murina-</em>infected rodents were biochemically and functionally characterized. These EVs contained host proteins involved in cellular, metabolic, and immune processes as well as proteins with homologs found in other fungal EV proteomes, indicating <em>Pneumocystis</em> may release EVs. Notably, EV uptake by<em> P. carinii</em> indicated their potential involvement in nutrient acquisition and indicated a possibility for using engineered EVs for efficient therapeutic delivery. However, EVs added to <em>P. carinii</em> <em>in vitro</em>, did not show increased growth or viability, implying that additional nutrients or factors are necessary to support their metabolic requirements. Exposure of macrophages to EVs increased proinflammatory cytokine levels but did not affect macrophages' ability to kill or phagocytose <em>P. carinii</em>. These findings provide vital insights into <em>P. carinii</em> and host EV interactions, yet the mechanisms underlying <em>P. carinii</em>'s survival in the lung remain uncertain. These studies are the first to isolate, characterize, and functionally assess EVs from <em>Pneumocystis</em>-infected rodents, promising to enhance our understanding of host-pathogen dynamics and therapeutic potential.</p>

They could be current users of slowfeeders (SFs), former users or non-users. </p> <p>Former- and non-users were asked about their perception of slowfeeding and their former use (for former users). <br>Operators answered few general questions about their yard (number of liveries, housing ...) and their slowfeeding practices.<br>Owners answered questions about their slowfeeding practices and could then fill the questionnaire for up to 5 horses (general information, housing, feeding, training, health).</p> <p>The questionnaire can be found in Appendix. </p> <p>Four (4) datasets were created based on the answers : DataFormerUsers (n=124), DataOperators (n=229), DataOwners (n=963) and DataHorses (n=1425). <br>The three last ones were the ones used for the analysis of the paper. DataFormerUsers is a supplementary dataset.</p> <p>All headers are in english. Some variables were recoded in English, and some are in French.</p> <p>A "how to read" file describing the different variables is provided with each dataset.</p>
PARK15/FBXO7 is dispensable for PINK1/Parkin mitophagy in iNeurons and HeLa cell systems
<p>The protein kinase PINK1 and ubiquitin ligase Parkin promote removal of damaged mitochondria via a feed-forward mechanism involving ubiquitin (Ub) phosphorylation, Parkin activation, and ubiquitylation of mitochondrial outer membrane proteins to support recruitment of mitophagy receptors. The ubiquitin ligase substrate receptor FBXO7/PARK15 is mutated in an early-onset parkinsonian-pyramidal syndrome. Previous studies have proposed a role for FBXO7 in promoting Parkin-dependent mitophagy. Here, we systematically examine the involvement of FBXO7 in depolarization-dependent mitophagy in the well-established HeLa and induced-neurons cell systems. We find that FBXO7<sup>-/-</sup> cells have no demonstrable defect in: 1) kinetics of pUb accumulation, 2) pUb puncta on mitochondria by super-resolution imaging, 3) recruitment of Parkin and autophagy machinery to damaged mitochondria, 4) mitophagic flux, and 5) mitochondrial clearance as quantified by global proteomics. Moreover, global proteomics of neurogenesis in the absence of FBXO7 reveals no obvious alterations in mitochondria or other organelles. These results argue against a general role for FBXO7 in Parkin-dependent mitophagy and point to the need for additional studies to define how FBXO7 mutations promote parkinsonian-pyramidal syndrome. </p>
