Dissolved organic carbon (DOC) and total dissolved nitrogen (TDN) from river, lagoon, and open ocean sites along the Alaska Beaufort Sea coast, 2018-ongoing

Multiple water types (river, lagoon, ocean) from the North Slope of Alaska and nearshore Beaufort Sea are sampled seasonally by the Beaufort Lagoon Ecosystems LTER (BLE LTER) Core Program to investigate biogeochemical linkages between terrestrial, lagoon, and open ocean ecosystems. Water samples are collected during full ice cover (April), ice break-up (mid-June to early July), and open water (late July and August) periods and analyzed for dissolved organic carbon and total dissolved nitrogen content.

Total dissolved nitrogen (TDN), dissolved organic carbon (DOC), radiocarbon (14C-DOC), and stable carbon (13C-DOC) of surface waters from the Canning River watershed, 2019 and 2021

Sites along the Canning River mainstem and contributing streams near the Kavik River Camp, Alaska, were visited to track changes in stream and river total dissolved nitrogen (TDN) concentration, dissolved organic carbon (DOC) concentration, and the stable carbon (13C) and radiocarbon (14C) isotopic composition of DOC across transitions between the Brooks Range, Brooks foothills, and Arctic Coastal Plain. The dataset also includes water samples collected from lakes, springs, groundwater, and streams and rivers outside the Canning River watershed. Water samples were collected in late April and early August 2019 and in late July and early August 2021. Data include measurements of individual samples for TDN (milligrams nitrogen per liter), DOC (milligrams carbon per liter), carbon-13 of DOC (reported as delta-13C, per mil), carbon-14 of DOC (reported as fraction modern), and analytical error in the fraction modern values. Ramped Pyrolysis Oxidation (RPO) coupled radiocarbon (14C-DOC) and stable carbon (13C-DOC), high-resolution molecular composition (FT-ICR MS), and biodegradable dissolved organic carbon (BDOC) of groundwater, river water, and lagoon water in northeast Alaska, 2017

Supra-permafrost groundwater (SPGW), river water, and lagoon water were sampled near Kaktovik, AK to assess the reactivity and origin of dissolved organic matter (DOM) across interconnected hydrologic systems during late summer. Water samples were collected on August 17th 2017 from SPGW along the beach of Jago Lagoon (Jago GW), surface water from the Jago River’s main channel above tidal influence (Jago R), and from the water column of Kaktovik Lagoon at 2–3 m depth (KA LW). Measurements were made from grab samples for river and lagoon water, and from a composite sample for SPGW gathered from 10 individual shoreline locations. Data include dissolved organic carbon concentration (DOC, mg C L-1), Ramped Pyrolysis Oxidation (RPO) derived fraction compositions of 13C-DOC (δ13C ‰), 14C-DOC (in fraction modern), and method/instrumental error in the 14C and 13C results, and Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR MS) molecular composition and summarized compound classes. Biodegradable DOC (BDOC) bottle experiments were performed using all three sample types, where DOC concentration was subsequently measured at 2, 7, 14, and 28 days. FT-ICR MS composition was subsequently measured at the 28-day timepoint to track changes in molecular formulae and compound class relative abundance following biodegradation. Data from RPO serial thermal oxidation include temperature and normalized CO2 profiles for each background sample. Thermal-oxidation profiles of CO2 were transformed into non-parametric activation energy (E) distributions using an inverse model. Model output includes C mass of oxidized CO2 (µg C), Tmax (K), Emax (kJ mol-1), Emean (kJ mol-1), Estd (kJ mol-1), and p(0,E)max of user-defined sample fractions. FT-ICR MS results include a summary table of the relative abundance of compound classes (e.g., unsaturated phenolic, polyphenolic, aliphatic, condensed aromatics, peptide-like) and elemental groupings (e.g., CHO-type, CHON-type, CHOS-type, CHON

Dissolved Organic Carbon (DOC) and Dissolved Total Nitrogen (DTN) from Northeast U.S. Shelf Long Term Ecological Research (NES-LTER) Transect cruises, ongoing since 2022

Dissolved organic carbon and dissolved total nitrogen are measured from discrete bottle samples collected during CTD rosette casts on Northeast U.S. Shelf Long-Term Ecological Research (NES-LTER) transect cruises (ongoing since 2022). Sampling frequency is approximately seasonal. Sample collection is paired with particulate organic carbon at surface, subsurface chlorophyll max, and sometimes a third depth. Samples are filtered directly from the CTD rosette and acidified in the field, then analyzed using a Shimadzu TOC-LCPH total organic carbon analyzer coupled to a TNM-L analyzer for total nitrogen. Values are reported in micromoles per liter.

Accession numbers for radiocarbon and stable carbon isotopes of dissolved organic carbon (DOC) and dissolved inorganic carbon (DIC) in soil leachates from permafrost soils collected from the North Slope of Alaska in the summers of 2018 and 2022

Leachates of dissolved organic carbon (DOC) from permafrost soils were prepared from soils collected from the North Slope of Alaska in 2018 and 2022. Soil leachates were then either kept in the dark or exposed to light from LEDs at 305 nm (UV) and 405 nm (visible), and then inoculated with native microbial communities and incubated. At the start of the biological incubations, single replicates of the DOC after dark or light treatment and inoculation were assigned accession numbers and analyzed for 14C and 13C at the National Ocean Sciences Accelerator Mass Spectrometry (NOSAMS) facility. At the end of the biological incubations, duplicates of the dissolved inorganic carbon (DIC) in those waters were assigned accession numbers and analyzed for 14C and 13C at the NOSAMS facility.

Stable isotope and conservative tracer data used to estimate uptake of stream water dissolved organic carbon (DOC) through a whole-stream addition of a ¹³C-DOC tracer coupled with laboratory measurements of bioavailability of the tracer and stream water DOC using lability profiling with bioreactors

We performed a whole-stream addition of a ¹³C-DOC tracer and made laboratory measurements of the biological availability of the tracer as well as stream water DOC. The study was performed in October 2002 in a 1.27 km stretch of the third-order White Clay Creek in southeastern Pennsylvania. The tracer was prepared as a cold-water leachate of ¹³C-labeled tulip poplar saplings and it was added to the stream along with sodium bromide, a conservative tracer, over a 2-h period. Stream water samples were collected at 8 downstream stations over an 8-h period, filtered, and analyzed for concentrations of bromide and DOC. DOC was measured by Pt-catalyzed, persulfate oxidation, Br- was analyzed by ion chromatography, and C isotope samples were rotary evaporated, acidified, lyophilized, combusted, and the CO₂ analyzed with an elemental analyzer interfaced with an isotope ratio mass spectrometer. Lability profiling of the ¹³C-DOC tracer and stream water DOC were performed with a series of plug-flow bioreactors of increasing empty-bed contact times with the concentration of biodegradable DOC operationally defined as the difference between the DOC concentrations in the influent and effluent waters of the bioreactors. The bioreactor measurements were performed 2 days after the whole-stream release. Data were analyzed to estimate the uptake of stream water DOC associated with labile and semi-labile fraction of biodegradable DOC. The dynamics and stoichiometry of dissolved organic carbon release by kelp: Carbon fixation, DOC release and nutrient uptake data

<p>Canopy-forming kelps are foundational species in coastal ecosystems, fixing tremendous amounts of carbon, yet we know little about the ecological and physiological determinants of dissolved organic carbon (DOC) release by kelps. We examined DOC release by the bull kelp, <i>Nereocystis luetkeana</i>, in relation to carbon fixation, nutrient uptake, tissue nitrogen content, and light availability. DOC release was approximately 3.5 times greater during the day than at night. During the day, <i>N. luetkeana</i> blades released an average of 16.2% of fixed carbon as DOC. Carbon fixation increased with light availability but DOC release did not, leading to a lower proportion of fixed carbon released as DOC at high light levels. We found no relationship between carbon fixation and DOC release rates measured concurrently. Rather, DOC release by <i>N. luetkeana</i> blades declined with marginal significance as blade tissue nitrogen content increased and with experimental nitrate addition, supporting the role of stoichiometric relationships in DOC release. Using a stable isotope (<sup>13</sup>C) tracer method, we demonstrated that inorganic carbon is rapidly fixed and released by <i>N. luetkeana</i> blades as <sup>13</sup>DOC, within hours. However, recently fixed carbon (<sup>13</sup>DOC) comprised less than 20% of the total DOC released, indicating that isotope studies that rely on tracer production alone may underestimate total DOC release, as it is decoupled from recent kelp productivity. Comparing carbon and nitrogen assimilation dynamics of the annual kelp <i>N. luetkeana</i> with the perennial kelp <i>Macrocystis pyrifera</i> revealed that <i>N. luetkeana </i>had significantly higher carbon fixation, DOC production and nitrogen uptake rates per unit dry mass. Both kelp species were able to perform light-independent carbon fixation at night. Carbon fixation by the annual kelp <i>N. luetkeana </i>is as high as 2.35 kg C m<sup>-2</sup> yr<sup>-1</sup>, but an average of 16% of this carbon (376 g C m<sup>-2</sup> yr<sup>-1</sup>) is released as DOC. As kelp forests are increasingly viewed as vehicles for carbon sequestration, it is important to consider the fate of this substantial quantity of DOC released by canopy-forming kelps.</p>

Dissolved organic carbon (DOC) is a poorly-characterized but large and dynamic pool of actively-cycling carbon in the oceans, and one of the largest organic carbon pools on the planet. The total DOC pool consists of three major fractions: refractory DOC resistant to microbial oxidation with a turnover time of millennia; semi-labile DOC, produced and decomposed on seasonal timescales, and labile DOC, consisting of simple, recently-produced compounds with nanomolar concentrations, and turnover times of minutes-days. The background concentration of refractory DOC in the deep ocean is 35-45 micromolar. DOC concentration in the upper 100-200 meters is enhanced by 10-50 micromolar with the addition of semilabile DOC. In subtropical and temperate oceans, semilabile DOC can form an important part of the carbon export by deep vertical mixing into the oceanic mid-depths. Concentrations of semilabile DOC are lower in the polar Southern Ocean than in most other regions.
