Dataset for: Dopamine neurons that inform Drosophila olfactory memory have distinct, acute functions driving attraction and aversion

<p>The brain must guide immediate responses to beneficial and harmful stimuli while simultaneously writing memories for future reference. While both immediate actions and reinforcement learning are instructed by dopamine, how dopaminergic systems maintain coherence between these two reward functions is unknown. Through optogenetic activation experiments, we showed that the dopamine neurons that inform olfactory memory in Drosophila have a distinct, parallel function driving attraction and aversion (valence). Sensory neurons required for olfactory memory were dispensable to dopaminergic valence. A broadly projecting set of dopaminergic cells had valence that was dependent on dopamine, glutamate, and octopamine. Similarly, a more restricted dopaminergic cluster with attractive valence was reliant on dopamine and glutamate; flies avoided opto-inhibition of this narrow subset, indicating the role of this cluster in controlling ongoing behavior. Dopamine valence was distinct from output-neuron opto-valence in locomotor pattern, strength, and polarity. Overall our data suggest that dopamine&rsquo;s acute effect on valence provides a mechanism by which a dopaminergic system can coherently write memories to influence future responses while guiding immediate attraction and aversion.</p>

Interspecies chimerism with human embryonic stem cells generates functional human dopamine neurons at low efficiency

<p>Interspecies chimeras offer great potential for regenerative medicine and creation of human disease models. Whether human pluripotent stem cell (hPSC) derived neurons in an interspecies chimera can differentiate into functional neurons and integrate into host neural circuity is not known. Here we show, using Engrailed 1 (En1) as a development niche that human naïve-like ES cells can incorporate into embryonic and adult mouse brains. Human-derived neurons including tyrosine hydroxylase (TH) positive neurons integrate into the mouse brain at low efficiency.  These TH-positive neurons have electrophysiologic properties consistent with their human origin. Additionally, these human-derived neurons in the mouse brain accumulate pathologic phosphorylated α-synuclein in response to α-synuclein preformed fibrils.  Optimization of human/mouse chimeras could be utilized to study human neuronal differentiation and human brain disorders.</p>

Data from: Neuronal function and dopamine signaling evolve at high temperature in Drosophila

<p>Neuronal activity is temperature sensitive and affects behavioral traits important for individual fitness, such as locomotion and courtship. Yet, we do not know enough about the evolutionary response of neuronal phenotypes in new temperature environments. Here, we use long-term experimental evolution of <em>Drosophila simulans</em> populations exposed to novel temperature regimes. Here, we demonstrate a direct relationship between thermal selective pressure and the evolution of neuronally expressed molecular and behavioral phenotypes. Several essential neuronal genes evolve lower expression at high temperatures and higher expression at low temperatures, with dopaminergic neurons standing out by displaying the most consistent expression change across independent replicates. We functionally validate the link between evolved gene expression and behavioral changes by pharmacological intervention in the experimentally evolved <em>D. simulans</em> populations as well as by genetically triggered expression changes of key genes in <em>D. melanogaster</em>. As natural temperature clines confirm our results for <em>Drosophila</em> and <em>Anopheles</em>populations, we conclude that neuronal dopamine evolution is a key factor for temperature adaptation.</p>

Population and single dopamine neuron activity during classical conditioning

<p>We trained naive or trained mice to associate odor cues with outcome (water, air puff or no outcome), and recorded dopamine cell body activity in the vetral tegmental area (VTA), dopamine axon activity in the ventral striatum (VS) or dopamine release in VS with optic fiber fluorometry (photometry). In different set of mice, single dopamine neuron activiy was recorded with 2-photon microscope. In some of these mice, we reversed odor-outcome contingency so that an odor that was associated with no outcome or air puff became associated with water reward. Licking pattern was also recorded.</p>

Characterizing the diversity of enteric neurons using Dopamine Transporter (DAT)-Cre reporter mice

<p>The enteric nervous system (ENS) comprises a complex network of neurons whereby a subset appears to be dopaminergic, although the characteristics, roles, and implications in disease are less understood. Most investigations relating to enteric dopamine (DA) neurons rely on immunoreactivity to tyrosine hydroxylase (TH) - a rate-limiting enzyme in the production of DA. However, TH immunoreactivity is likely to provide an incomplete picture given previous work has showed that some DA neurons contain little if any TH and its levels tend to be decreased in response to cellular stress. The study herein provides a comprehensive characterization of DA neurons in the gut using a well-accepted reporter mouse line, expressing a fluorescent protein under control of the DA transporter (DAT) promoter. Our findings confirm a unique localization of DA neurons in the gut and unveil the discrete subtypes of DA neurons in this organ, which we characterized using both immunofluorescence and single-cell transcriptomics, validated by&nbsp;<em>in situ</em>hybridization. We observed distinct subtypes of DAT neurons expressing co-transmitters and modulators across both plexuses; some of them likely co-releasing acetylcholine, and a smaller population likely releasing nitric oxide; while others were positive for a slew of canonical DA markers. Interestingly, we uncovered a seemingly novel population of DA neurons unique to the ENS which were ChAT/DAT-positive neurons and characterised by the expression of&nbsp;<em>Th</em>,&nbsp;<em>Grp</em>,<em>Calcb</em>&nbsp;and<em>&nbsp;Sst</em>. Given the clear heterogeneity of DAergic gut neurons, further investigation is warranted to define their functional signatures&nbsp;and discover any inherent vulnerabilities in disease.&nbsp;&nbsp;</p>

Single unit electrophysiology of optogenetically identified dopamine neurons in classical conditioning with probabilistic outcome of reward and punishment

<p>We collected single-unit data (.mat file) from optogenetically identified dopamine neurons in the lateral VTA in mice. These dopamine neurons were recorded while animals performed 4 different variation of classical conditioning tasks with different ratio of probabilistic outcomes of water reward and aversive air puff. In one task, one cue was associated with both water and puff in a probabilistic manner.</p>
