Perry et al. (2025) Data Package: Effects of diluted bitumen and remediation methods on lower trophic levels within boreal lake enclosures. Data were collected during 2019 at the IISD Experimental Lakes Area in Northwestern Ontario.

This data package corresponds to a research study by Perry et al. (2025) titled "The effects of diluted bitumen, the shoreline cleaner Corexit EC9580A, and bio-stimulation on the lower food web of a boreal lake, with a focus on natural phytoplankton communities." The study examines the effect of controlled spills of diluted bitumen and two remediation methods on lower trophic levels (phytoplankton, periphyton, zooplankton). The study was undertaken within shoreline enclosures within Lake 260 at the IISD Experimental Lakes Area during 2019. In addition to primary oil recovery using sorbent pads, the two secondary remediation methods: 1) enhanced monitoring natural recovery (eMNR) that included the biostimulation of microbial communities via a slow release nutrient fertilizer, and 2) a shoreline washing agent (SWA or SCA; Corexit 9580) used to increase oil removal from affected shorelines. This data package includes the response of perphyton and zooplankton.

Jornada Basin and Experimental Range Mesquite Herbicide Project (JERHM) Core Methods Data, 2020-2022

This dataset includes contains line-point intercept, plant height, gap, and species inventory data collected over three years (2020-2022) as part of the Jornada Experimental Range Herbicide Mesquite Project (JERHM). Data were collected to assess plant community composition and structural change to herbicide application across a Black grama (Bouteloua eriopoda) grassland to Honey mesquite (Neltuma glandulosa [=Prosopis glandulosa]) shrubland encroachment gradient. Twenty sets of paired, 5-hectare plots (n=40 plots total) were established across a N. glandulosa encroachment gradient in 2020. One plot within each plot pair received an aerial application of herbicide in 2021, with the second plot left untreated by herbicide as a control. Data were collected annually following the following the Monitoring Manual for Grassland, Shrubland, and Savanna Ecosystems (Herrick et al. 2017) on each of three, 50m permanent transects established on each plot. These data are also available within the Landscape Data Commons (https://landscapedatacommons.org/) under ProjectKey=Jornada_JERHM. There are no immediate plans to continue data collection.

Experimental characterization of transversal-heterogeneous clay mixtures by the spectral induced polarization method

<p>In this folder you will find multiple datasets (*.txt) from SIP measurements of transversal-heterogeneous clay mixtures using spectral induced polarization acquired between April and May 2022. Additionally, we include two python codes to read and process the data.</p> <p>SIP_Plot_ReWrite.py is a python program aimed to process a .res file from a SIP Fuchs III.<br> It gives a .txt file with the frequency, the resistivity, the phase and the associated errors.<br> In order for the program to give the resistivity, you will need to enter the geometric factor of the studied sample.</p> <p><br> The six text files in the folder (excluding README.txt) were created using SIP_Plot_ReWrite.py.</p> <p>IL_1by1.txt and IL_1by1_V2.txt are from two different homogeneous mixtures of illite and water with a concentration of initially 0.01 mol/L of NaCl.<br> MtR_1by1.txt is from a homogeneous mixture of red montmorillonite and water with a concentration of initially 0.01 mol/L of NaCl.</p> <p>IL_MtR_1by2.txt, IL_MtR_1by4.txt and IL_MtR_1by8.txt are from three transversal-heterogeneous mixtures of illite and red montmorillonite with water containing a concentration of initially 0.01 mol/L of NaCl.</p> <p><br> For IL_MtR_1by2.txt, there was one portion of each clay types, occupying a half of the cylindrical container each.</p> <p><br> For IL_MtR_1by4.txt, there was two portions of each clay types, occupying a quarter of the cylindrical container each.</p> <p><br> These two samples were made using the same mixtures as for IL_1by1.txt and MtR_1by1.txt.</p> <p><br> For IL_MtR_1by8.txt, there was four portions of each clay types, occupying an eighth of the cylindrical container each.<br> This sample was made using the same mixtures as for IL_1by1_V2.txt and MtR_1by1.txt.</p> <p><br> TestDoubleColeColeFit.py is a python program which optimizes a double Cole-Cole model by multiplication on SIP data.<br> This program needs a file with the same structure as the .txt file made by SIP_Plot_ReWrite.py.</p>

This drying and warming experiment addresses the following questions: 1) Does ecosystem drying, warming and permafrost thaw cause a net release or uptake of C from the ecosystem to the atmosphere?, 2) Does the decomposition of old C that comprises the bulk of the soil C pool influence ecosystem C loss? 3) How do drying and warmign affect plant communities and ecosystem properties? We are answering these questions using a combined warming and drying experiment (DryPEHR), which is situated with the Carbon in Permafrost Experimental Heating Research (CiPEHR) project and located in an upland tundra field site near Healy, Alaska in the foothills of the Alaska Range. Warming treatment here refers to growing season air temperature warming (~1C) using open top chambers (OTC) combined with soil 'warming' using snow fences during the snow covered months. Drying is achieve using an automated pumping system that lowers the water table in the dry plots. Soil warming began in 2008; OTCs and drying in 2011. This dataset contains point measurements of winter ecosystem respiration fluxes using the on-plot method and the soil temperature, air temperature, and snow depth associated with each flux.

Data from: Head-to-head comparison of three experimental methods of quantifying competitive fitness in C. elegans

Organismal fitness is relevant in many contexts in biology. The most meaningful experimental measure of fitness is competitive fitness, when two or more entities (e.g., genotypes) are allowed to compete directly. In theory, competitive fitness is simple to measure: an experimental population is initiated with the different types in known proportions and allowed to evolve under experimental conditions to a predefined endpoint. In practice, there are several obstacles to obtaining robust estimates of competitive fitness in multicellular organisms, the most pervasive of which is simply the time it takes to count many individuals of different types from many replicate populations. Methods by which counting can be automated in high throughput are desirable, but for automated methods to be useful, the bias and technical variance associated with the method must be (a) known, and (b) sufficiently small relative to other sources of bias and variance to make the effort worthwhile. The nematode Caenorhabditis elegans is an important model organism, and the fitness effects of genotype and environmental conditions are often of interest. We report a comparison of three experimental methods of quantifying competitive fitness, in which wild-type strains are competed against GFP-marked competitors under standard laboratory conditions. Population samples were split into three replicates and counted (1) "by eye" from a saved image, (2) from the same image using CellProfiler image analysis software, and (3) with a large particle flow cytometer (a "worm sorter"). From 720 replicate samples, neither the frequency of wild-type worms nor the among-sample variance differed significantly between the three methods. CellProfiler and the worm sorter provide at least a tenfold increase in sample handling speed with little (if any) bias or increase in variance.
