Data from: Telomere length declines with age, but relates to immune function independent of age in a wild passerine
<p><span>Telomere length (TL) shortens with age but telomere dynamics can relate to fitness components independent of age. Immune function often relates to such fitness components and can also interact with telomeres. Studying the link between TL and immune function may therefore help us understand telomere-fitness associations. We assessed the relationships between erythrocyte TL and four immune indices (haptoglobin, natural antibodies, complement activity, heterophil-lymphocyte ratio; n=477-589), from known-aged individuals of a wild passerine (<em>Malurus coronatus</em>). As expected, we find that TL significantly declined with age. To verify whether associations between TL and immune function were independent of parallel age-related changes (e.g. immunosenescence), we statistically controlled for sampling age, and used within-subject centring of TL to separate relationships within or between individuals. We found that TL positively predicted complement activity at the between-individual level (individuals with longer average TL had higher complement activity), but no other immune indices. In contrast, age predicted levels of natural antibodies and heterophil-lymphocyte ratio, allowing inference that respective associations between TL and age with immune indices are independent. Any links existing between TL and fitness are therefore unlikely to be strongly mediated by innate immune function, while TL and immune indices appear independent expressions of individual heterogeneity.</span></p>

Independence of mate choice components (choosiness and preference functions) in Hyla versicolor
<p>Mate choice is an important cause of natural and sexual selection, driving the evolution of ornaments and promoting diversification and speciation. Mate choice decisions arise from the interaction of several components, and knowledge of whether they interact, and how, is crucial for understanding their contributions to selection. Here we focus on the relationship between preference functions (attractiveness ranking of prospective mates) and choosiness (effort invested in obtaining the preferred mate) and test the hypothesis that they are independent components of mate choice decisions. We examine individual variation in preference functions and choosiness for call duration in female <em>Hyla versicolor </em>treefrogs, and show that measures describing preference functions and choosiness are not correlated. We also found a suggestive but inconclusive pattern that both components are influenced by different factors (body measures and hormones). Independence of preference and choosiness suggests that the joint study of variation in both components is required to gain a complete understanding of how mate choice contributes to sexual selection and speciation.</p>

<p><b>PREMISE </b></p> <p>Across taxa, vegetative and floral traits that vary along a fast-slow life-history axis are often correlated with leaf functional traits arrayed along the leaf economics spectrum, suggesting a constrained set of adaptive trait combinations. Such broad-scale convergence may arise from genetic constraints imposed by pleiotropy (or tight linkage) within species, or from natural selection alone. Understanding the genetic basis of trait syndromes and their components is key to distinguishing these alternatives and predicting evolution in novel environments.</p> <p><b>METHODS </b></p> <p>We used a line-cross approach and quantitative trait locus (QTL) mapping to characterize the genetic basis of twenty leaf functional/physiological, life history, and floral traits in hybrids between annualized and perennial populations of scarlet monkeyflower (<i>Mimulus cardinalis</i>).</p> <p><b>RESULTS </b></p> <p>We mapped both single and multi-trait QTLs for life history, leaf function and reproductive traits, but found no evidence of genetic co-ordination across categories. A major QTL for three leaf functional traits (thickness, photosynthetic rate, and stomatal resistance) suggests that a simple shift in leaf anatomy may be key to adaptation to seasonally dry habitats.</p> <p><b>CONCLUSIONS </b></p> <p>Our results suggest that the co-ordination of resource-acquisitive leaf physiological traits with a fast life history and more selfing mating system results from environmental selection rather than functional or genetic constraint. Independent assortment of distinct trait modules, as well as a simple genetic basis to leaf physiological traits associated with drought escape, may facilitate adaptation to changing climates. </p>

A histone methyltransferase-independent function of PRC2 controls small RNA dynamics during programmed DNA elimination in Paramecium
<p><span lang="EN-US">To limit transposable element (TE) mobilization, most eukaryotes have evolved small RNAs to silence TE activity via homology-dependent mechanisms. Small RNAs, 20-30 nucleotides in length, bind to PIWI proteins and guide them to nascent transcripts by sequence complementarity, triggering the recruitment of histone methyltransferase enzymes on chromatin to repress the transcriptional activity of TEs and other repeats. In<span> the ciliate <em>Paramecium tetraurelia</em>,</span> 25-nt scnRNAs corresponding to TEs recruit Polycomb Repressive Complex 2 (PRC2), and trigger their elimination during the formation of the somatic nucleus. Here, we sequenced sRNAs during the entire sexual cycle with unprecedented resolution. Our data confirmed that scnRNAs are produced from the entire germline genome, from TEs and non-TE sequences, during meiosis. Non-TE scnRNAs are selectively degraded, which results in the specific selection of TE-scnRNAs. We demonstrate that PRC2 is essential for the selective degradation of non-TE-scnRNAs, independently of its histone methyltransferase activity. We further show that the PRC2 cofactor Rf4 mediates the physical interaction between the scnRNA-binding protein Ptiwi09 and the zinc finger protein Gtsf1, pointing to an architectural role of PRC2 in scnRNA degradation.</span></p>
