<p>Raw feature tables used for metabolomic analysis of the <em>Caenorhabditis elegans</em> mutant <em>swip-10</em>. The data were generated using liquid chromatography coupled high-resolution mass spectrometry. Two different columns were used: HILIC (+ ESI) and C18 (-ESI), coupled to a Thermo Q-Exactive Orbitrap mass spectrometer. Correlation of the kinetics of viral antigen and genomic RNA with restoration of normal cell homeostasis
<p>The main objective of the studies within COCID Work Package 6 is to understand basic mechanisms by which viral replication machineries are removed from cells after pharmacological interruption of viral replication using functional as well as imaging techniques, including soft X-ray tomography.</p> <p>In this document, we highlight the progress in establishing the HCV replication models (replicons), the antiviral treatment chosen to eliminate viral structures from the host cells and the correlation between elimination of the viral replication machinery and restoration of a normal host cell homeostasis, using markers of HCV-induced stress. These data are essential to frame the experimental setup chosen for the imaging process required in subsequent stps of the project.</p>

Both impaired DNA repair mechanisms and disrupted telomere length homeostasis represent potential culprits in CRC onset, its dissemination in the body and prognosis. Above parameters are becoming critical as prognostic markers, since CRC therapy is based on compounds interacting with DNA. DNA repair capacity in CRC patients have recently been studied in order to address prediction of therapy response. Due to the substantial interindividual variations in DNA repair capacities and relative telomere length, these markers may pose a possible contribution in individualized therapeutical regimen of CRC patients. Telomere attrition, responsible for replicative senescence in healthy cells, may become a hallmark of malignant transformation of the cell due to by-passing cell cycle checkpoints. Telomerase – a key enzyme keeping homeostasis of telomere - is almost ubiquitous in advanced solid cancers, including CRC, and its expression is fundamental to cell immortalization.<br> Here we present our data based on the investigation of base excision repair capacities and relative telomere length in tumor tissues and adjacent non-malignant mucosa of sporadic CRC patients. The relative gene expression of telomerases is monitored as well. Particular attention will be dedicated to the CRC phenotypes and clinicopathological characteristics. We also addressed telomere homeostasis in peripheral blood lymphocytes of CRC patients in several consecutive samplings (at diagnosis, immediately after treatment and in additional follow-up intervals), which could provide us the insight into the treatment response. This aspect is of particular relevance, since there is currently a persistent effort to develop therapeutics, which are telomerase-specific and gentle to non-malignant tissue. However, in practice, we are at the dawn of clinical trials. Additionally, emerging crosstalks between DNA repair and telomere length homeostasis may cast some lights on a dynamic of genomic instability, a fundamental hallmark (or cause) of cancer.</p> <p>Acknowledgement: GACR 21-04607X, 19-10543S, AZV NV18/03/00199</p>
Patient-specific induced pluripotent stem cell properties implicate Ca2+-homeostasis in clinical arrhythmia associated with combined heterozygous RYR2 and SCN10A variants
<p class="MsoNormal"><span>We illustrate the use of induced pluripotent stem cells (iPSCs) as platforms for investigating cardiomyocyte phenotypes in a human family pedigree exemplified by novel heterozygous RYR2-A1855D and SCN10A-Q1362H variants occurring alone and in combination. The proband, a four-month-old boy, presented with </span><span>polymorphic</span><span> ventricular tachycardia (</span><span>P</span><span>VT). Genetic tests revealed double novel heterozygous RYR2-A1855D and SCN10A-Q1362H variants inherited from his father (F) and mother (M) respectively. His father showed ventricular premature beats (VPB); his mother was asymptomatic. Molecular biological characterisations demonstrated greater <em>TNNT2</em> mRNA expression in the iPSCs-induced cardiomyocytes (iPS-CMs) than in the iPSCs</span><span>.</span><span> </span><span>c</span><span>TNTs became progressively organised, but cytoplasmic RYR2 and SCN10A aggregations occurred in the iPS-CMs. Proband-specific iPS-CMs showed decreased <em>RYR2</em> and <em>SCN10A</em> mRNA expression. The RYR2-A1855D variant resulted in premature spontaneous sarcoplasmic reticular (SR) Ca<sup>2+</sup> transients (PCTs), Ca<sup>2+</sup> oscillations (COs), and increased action potential durations (APDs). SCN10A-Q1362H did not confer any specific phenotype. However, the </span><span>combined </span><span>heterozygous RYR2-A1855D and SCN10A-Q1362H variants in the proband iPS-CMs resulted in accentuated Ca<sup>2+</sup> homeostasis disorders, AP prolongation and susceptibility to early afterdepolarisations (EADs) at high stimulus frequencies. These findings attribute the clinical phenotype in the proband to effects of the heterozygous <em>RYR2</em> variant exacerbated by heterozygous <em>SCN10A</em> modification. </span></p>
