Background optimization of powder electron diffraction to implement e-PDF technique and study the local structure of iron oxide nanocrystals

The local structural characterization of iron oxide nanoparticles is explored using a total scattering analysis method known as Pair Distribution Function (PDF) (also known as Reduced Density Function) profiles derived from background corrected powder electron diffraction patterns. Due to the strong coulombic interaction between the electron beam and the sample, electron diffraction generally leads to multiple scattering, causing redistribution of intensities towards higher scattering angles and an increased background in the diffraction profile. In addition to this, the electron-specimen interaction gives rise to an undesirable inelastic scattering signal that contributes primarily to the background. The present work demonstrates the efficacy of a pre-treatment of the underlying complex background function, which is a combination of both incoherent multiple and inelastic scatterings that cannot be identical for different electron beam energies. Therefore, two different background subtraction approaches are proposed for the electron diffraction patterns acquired at 80 kV and 300 kV beam energies. From the least square refinement (small-box modelling), both approaches are found to be very promising, leading to a successful implementation of the e-PDF technique to study the local structure of the considered nanomaterial.

Data: In vivo fate of free and encapsulated iron oxide nanoparticles after injection of labelled stem cells

This data set is composed of magnetic resonance images (MRI) that are supporting the article entitled In vivo fate of free and encapsulated iron oxide nanoparticles after injection of labelled stem cells by the same authors. Nanoparticle contrast agents are used to label stem cells and monitor their bio-distribution in pre-clinical models of disease. Due to the impact on the interpretation of imaging results, understanding the in vivo fate of the particles is important. The bio-distribution after intra-cardiac injection of labelled cells with superparamagnetic iron oxide nanoparticles was monitored longitudinally by MRI.

Spatiotemporal study of iron oxide nanoparticle monolayer formation at liquid/liquid interfaces by using in situ small-angle x‐ray scattering

Spatial and temporal small angle x-ray scattering (SAXS) scans show that 8.6 and 11.8 nm iron oxide nanoparticles (NPs) in heptane drop-cast on top of a heptane layer atop a diethylene glycol (DEG) layer are trapped at the DEG/heptane interface to generally form a single ordered, hexagonal close packed monolayer (ML), and this occurs long before the heptane evaporates. Many NPs remain dispersed in the heptane after this NP assembly. Assembly occurs faster than expected from considering only the diffusion of NPs from the drop-cast site to this liquid/liquid interface. The formation of the ordered NP ML occurs within 100 s of drop-casting, as followed by using the (10) ordered NP SAXS peak, and on the same time scale there is a concomitant decrease in the SAXS form factor from disordered NPs that is apparently from disordered NPs at the meniscus. Usually, most of the ordered NPs are close packed, but there is evidence that some are ordered though not close packed. After the heptane evaporates, a close-packed ordered NP ML remains at the DEG/vapor interface, though with smaller NP--NP separation, as expected due to less van der Waals shielding caused by the upper medium in the interface. x-ray beam transmission at different vertical heights characterizes the heptane and DEG bulk and interfacial regions, while monitoring the time dependence of SAXS at and near the DEG/heptane interface gives a clear picture of the evolution of NP assembly at this liquid/liquid interface. These SAXS observations of self-limited NP ML formation at the DEG/heptane interface are consistent with those using the less direct method of real-time optical reflection monitoring of that interface.

The magnetic recording stability of vortex state irregularly shaped natural iron oxides: raw data, processing and micromagnetic modelling results

Magnetic minerals, especially those existing within the vortex domain state, serve as the primary natural archives of ancient magnetic fields. In this investigation, we introduce an innovative method to examine the magnetic stability of remanence-bearing minerals. This method involves integrating Synchrotron-based Ptychographic X-ray Computed Nano-tomography (PXCT) with micromagnetic modelling. PXCT, a tomographic technique, is a non-destructive resource, which enables its application to valuable (unique) samples. When applied to a microscopic sample of weakly magnetic carbonate rock, PXCT revealed numerous nanoscopic grains of magnetite/maghemite, each exhibiting diverse morphologies, alongside various non-magnetic minerals present in the rock matrix. Subsequently, micromagnetic models were employed to predict the properties of these grains and investigate the potential impacts of irregular morphologies.

Oxygen availability regulates the quality of soil dissolved organic matter by mediating microbial metabolism and iron oxidation

Dissolved organic matter (DOM) plays a vital role in biogeochemical processes and in determining the responses of soil organic matter (SOM) to global change. Although the quantity of soil DOM has been inventoried across diverse spatio-temporal scales, the underlying mechanisms accounting for variability in DOM dynamics remain unclear, especially in upland ecosystems. Here, a gradient of SOM storage across twelve croplands in northeast China was used to understand links between DOM dynamics, microbial metabolism, and abiotic conditions. We assessed the composition, biodegradability and key biodegradable components of DOM. In addition, SOM and mineral-associated organic matter (MAOM) composition, soil enzyme activities, oxygen availability, soil texture, iron (Fe), Fe-bound organic matter and nutrient concentrations were quantified to clarify the drivers of DOM quality (composition and biodegradability). The proportion of biodegradable DOM increased exponentially with decreasing initial DOM concentration due to larger fractions of depolymerized DOM that was rich in small-molecular phenols and proteinaceous components. Unexpectedly, the composition of DOM was decoupled from that of SOM or MAOM, but significantly related to enzymatic properties. These results indicate that microbial metabolism exhibited a dominant role in DOM generation. As DOM concentration declined, increased soil oxygen availability regulated DOM composition and enhanced its biodegradability mainly through mediating microbial metabolism and Fe oxidation. The oxygen-induced oxidation of Fe(II) to Fe(III) removed complex DOM compounds with large molecular weight. Moreover, increased oxygen availability stimulated oxidase-catalyzed depolymerization of aromatic substances, and promoted production of protein-like DOM components due to lower enzymatic C/N acquisition ratio. As global changes in temperature and moisture will have large impacts on soil oxygen availability, the role of oxygen in regulating DOM dynamics highlights the importance of integrating soil oxygen supply with microbial metabolism and Fe redox status to improve model predictions of soil carbon under climate change.

CO2 Protects Cells from Iron-Fenton Oxidative DNA Damage in E. coli and Humans

Whereas hydroxyl radical is commonly named as the Fenton product responsible for DNA and RNA
damage in cells, here we demonstrate that the cellular reaction generates carbonate radical
anion due to physiological levels of bicarbonate. Analysis of the metabolome, the transcriptome,
and the human genome shows a consistent buffering of H2O2-induced oxidative
stress leading to one common pathway, namely guanine oxidation. Particularly revealing are nanopore-
based studies of direct RNA sequencing of cytosolic and mitochondrial ribosomal RNA along with
glycosylase-dependent qPCR studies of oxidative DNA damage in telomeres. The focusing of oxidative
modification on one pathway is consistent with the highly evolved base excision repair suite of enzymes
and their involvement in gene regulation in response to oxidative stress.
