Disentangling relationships between physiology, morphology, diet, and gut microbial diversity in American Kestrel nestlings

<p>Gut microbiota are increasingly recognized as important drivers of host health and fitness across vertebrate taxa. Given that gut microbial composition is directly influenced by the environment, gut microbiota may also serve as an eco-physiological mechanism connecting host ecology, such as diet, and physiology. Although gut microbiota have been well-studied in mammalian systems, little is known about how gut microbial diversity and composition impact morphological and physiological development in wild birds. Here, we characterized both diet and gut microbial diversity of free-living American kestrel (<em>Falco sparverius</em>) nestlings throughout development to test whether gut microbial diversity predicts host morphological and physiological traits in either contemporary or time-lagged manners. Gut microbial alpha diversity on day 21 of nestling development was positively correlated with diet alpha diversity representative of the majority of nestling development (days 5–20). Gut microbial alpha diversity early in development was negatively correlated with body mass in both contemporary and time-lagged manners. Gut microbial alpha diversity early in development was positively correlated with blood glucose later in development. As nestlings experience rapid growth demands in preparation to fledge, these time-lagged associations may indicate that gut microbial diversity at early critical developmental windows may determine the future trajectory of morphological and physiological traits underlying metabolism that ultimately impact fitness.</p>

Impacts of gut microbial depletion on tadpole thermal physiology

<p>The goal of our study was to determine if depletion of gut microbiome diversity would alter host thermal tolerance in a vertebrate ectotherm. We raised laboratory-reared tadpoles of the green frog in two microbial colonization treatments: colonized and depleted. Colonized tadpoles were raised in natural pond water to provide a rich source of microbiota for colonization of the gut, and depleted tadpoles were raised in autoclaved pond water to reduce this source of microbiota. We then performed a suite of experiments to test for differences in thermal physiology between groups. In our first experiment we tested tadpole acute thermal tolerance (critical thermal minimum and maximum). We verified our results in a second set of tadpoles and then tested tadpole survival under heat stress and thermal sensitivity of locomotion performance. Lastly, we tested for differences in physiological traits at lower levels of biological organization to identify putative mechanisms behind our results including mitochondrial enzyme activities, phospholipid membrane composition, and whole-organism metabolic rate.</p>

Data on physico-chemical quality and microbial community level physiological profiles of drinking water and freshwater resources in Mega Manila Philippines

<p><strong>Samples of drinking water and freshwater resources in Mega Manila, Philippines were analyzed for physical and chemical quality as well as physiological profiles of microbial communities. Temperature, pH, oxidation-reduction potential, absolute conductivity, resistivity, total dissolved solids, salinity, pressure, and dissolved oxygen were measured using a multi-parameter probe and reported as averages from three biological replicates per sample. Detection and quantification of arsenic were carried out via 3114 B: Hydride Generation AAS method, while analysis for lead and cadmium was carried out via 3111 B: Direct Air-Acetylene Flame method. Detection and quantification of mercury was performed via Inductively Coupled Plasma-Optical Emission Spectroscopy (ICP-OES). Detection and quantification of polyaromatic hydrocarbons (PAHs) and organochlorine pesticides (OCPs) were carried out via Gas Chromatography-Mass Spectrometry (GC-MS) and Gas Chromatography-Electron Capture Detection (GC-ECD). Assessment of the physiological activities of microbial communities in the water samples was done using the Biolog EcoPlateTM system.&nbsp; Eight samples returned less than the minimum detection limit for analysis of heavy metals and no samples contained polyaromatic hydrocarbons and organochlorine pesticides. In terms of microbial community physiological profiles, 66% of the samples (19 out 29) are found to demonstrate consistent increases in AMR and CMD over time. These data can be reused as comparators for retrospective and prospective quality assessments of drinking&nbsp; water and freshwater sources.</strong></p>
