Theoretical work suggests that sexual conflict should promote the maintenance of genetic diversity by the opposing directions of selection on males and females. If such conflict is pervasive, it could potentially lead to genomic heterogeneity in levels of genetic diversity an idea that so far has not been empirically tested on a genome-wide scale. We used large-scale population genomic and transcriptomic data from the collared flycatcher (Ficedula albicollis) to analyse how sexual conflict, for which we use sex-biased gene expression as a proxy, relates to genetic variability. Here, we demonstrate that the extent of sex-biased gene expression of both male-biased and female-biased genes is significantly correlated with levels of nucleotide diversity in gene sequences and that this correlation extends to diversity levels also in intergenic DNA and introns. We find signatures of balancing selection in sex-biased genes but also note that relaxed purifying selection could potentially explain part of the observed patterns. The finding of significant genetic differentiation between males and females for male-biased (and gonad-specific) genes indicates ongoing sexual conflict and sex-specific viability selection, potentially driven by sexual selection. Our results thus indicate that sexual antagonism could potentially be considered as one viable explanation to the long-standing question in evolutionary biology of how genomes can remain so genetically variable in face of strong natural and sexual selection.

Natural variation at a single gene generates sexual antagonism across fitness components in Drosophila

<p class="MsoNormal"><span>Mutations with conflicting fitness effects in males and females accumulate in sexual populations, reducing their adaptive capa</span><span>city</span><span>. Although quantitative genetic studies indicate that sexually antagonistic polymorphisms are common</span><span>, their molecular basis and population genetic properties remain poorly understood</span><span>. Here, we show in fruit flies how natural variation at a single gene generates sexual antagonism through phenotypic effects on cuticular hydrocarbon (CHC) traits that function as both mate signals and protectors against abiotic stress</span><span></span><span> across a latitudinal gradient. Tropical populations of <em>Drosophila serrata </em><span>have </span>polymorphic CHCs producing sexual antagonism through opposing but sex-limited effects on these two fitness-related functions. We dissected this polymorphism to a single fatty-acyl CoA reductase gene, <em>DsFAR2-B</em>, that is expressed in oenocyte cells where CHCs are synthesised. RNAi mediated disruption of the <em>DsFAR2-B</em> ortholog in <em>D. melanogaster</em> oenocytes affected CHCs in a similar way to that seen in <em>D. serrata</em>. Population genomic analysis revealed that balancing selection likely operates at the <em>DsFAR2-B </em>locus in the wild. Our study provides insights into the genetic basis of sexual antagonism in nature and connects sexually varying antagonistic selection on phenotypes with balancing selection on genotypes that maintains molecular variation. </span></p>

Data from: Sexual antagonism in the pistil varies among populations of a hermaphroditic mixed-mating plant

Sexual conflicts and their evolutionary outcomes may be influenced by population-specific features such as mating system and ecological context; however, very few studies have investigated the link between sexual conflict and mating system. The self-compatible, mixed-mating hermaphrodite Collinsia heterophylla (Plantaginaceae) is thought to exhibit a sexual conflict over timing of stigma receptivity. This conflict involves 1) delayed stigma receptivity, which intensifies pollen competition, and 2) early fertilization forced by pollen, which reduces seed set. We investigated the potential for the conflict to occur under field conditions and performed greenhouse crosses within eight populations to assess its consistency across populations. Flowers were visited, and produced seeds after pollination, at all developmental stages, suggesting that the conflict can be of significance under natural conditions. In the greenhouse, early pollination imposed costs in all populations. Overall, the timing of first seed set was most strongly affected by the maternal parent, denoting stronger female than male ability to influence onset of stigma receptivity. Crosses also revealed a negative relationship between donor- and recipient-related onset of receptivity within individuals, a novel result hinting at trade-offs in sex-allocation or a history of antagonistic selection. Neither timing of stigma receptivity, timing of first seed set, nor pollen competitive ability covaried with population outcrossing rate. In conclusion, these results indicate that sexually antagonistic selection may be present in varying degrees in different populations of C. heterophylla, but this variation does not appear to be directly related to mating system variation.

A critical task in evolutionary genetics is to explain the persistence of heritable variation in fitness-related traits such as immunity. Ecological factors can maintain genetic variation in immunity, but less is known about the role of other factors, such as antagonistic pleiotropy, on immunity. Sexually dimorphic immunity—with females often being more immune-competent—may maintain variation in immunity in dioecious populations. Most eco-immunological studies assess host resistance to parasites rather than the host's ability to maintain fitness during infection (tolerance). Distinguishing between resistance and tolerance is important as they are thought to have markedly different evolutionary and epidemiological outcomes. Few studies have investigated tolerance in animals, and the extent of sexual dimorphism in tolerance is unknown. Using males and females from 50 Drosophila melanogaster genotypes, we investigated possible sources of genetic variation for immunity by assessing both resistance and tolerance to the common bacterial pathogen Pseudomonas aeruginosa. We found evidence of sexual dimorphism and sexual antagonism for resistance and tolerance, and a trade-off between the two traits. Our findings suggest that antagonistic pleiotropy may be a major contributor to variation in immunity, with implications for host–parasite coevolution.

Data from: Protein deprivation decreases male survival and the intensity of sexual antagonism in southern field crickets Gryllus bimaculatus

Recent theory predicts that the magnitude of sexual antagonism should depend on how well populations are adapted to their environment. We tested this idea experimentally by comparing intersexual genetic correlations for adult survival in pedigreed populations of southern field crickets (Gryllus bimaculatus) raised on naturally balanced (free-choice) vs. imbalanced (protein-deprived) diets. We tested for (1) sex differences in nutritional intake and preference, (2) sex-specific effects of protein deprivation on survival and (3) diet dependence of the level of sexual antagonism. Adult males and females consumed a similar amount of protein, but protein deprivation decreased male survival but not female survival. Protein deprivation appeared to decrease the degree of sexual antagonism as intersexual genetic correlations were significantly lower than 1 only for the complementary free-choice diet group but close to 1 for the protein-deficient diet group. Our findings thereby implied that variation in nutritional environments can alter the magnitude of sexual antagonism. This research represents an important step towards understanding the relationship between sexual antagonism and adaptation in heterogeneous environments.
