Data from: Geographic distribution of terpenoid chemotypes in Tanacetum vulgare mediates tansy aphid occurrence but not abundance

<p>Intraspecific variation of specialized metabolites in plants, such as terpenoids, are used to determine chemotypes. Tansy (<em>Tanacetum vulgare</em> L.) exhibits diverse terpenoid profiles that affect insect communities. However, it is not fully known whether patterns of their chemical composition and associated insects vary beyond the community scale. Here, we investigated the geographic distribution of mono- and sesquiterpenoid chemotypes in tansy leaves and their relationships with specific insect communities across Germany. We sampled tansy leaves from ten plants with and five plants without aphids in each of 26 sites along a north-south and west-east transect in Germany. Hexane-extracted metabolites from leaf tissues were analyzed by gas chromatography-mass spectrometry (GC-MS). Plant morphological traits, aphid occurrence and abundance, and occurrence of ants were recorded locally. The effect of plant chemotype, plant morphological parameters, and abiotic site parameters such as soil types, temperature and precipitation on insect occurrences were analyzed. Plants clustered into four monoterpenoid and four sesquiterpenoid chemotype classes. Monoterpene classes differed in their latitudinal distribution, whereas sesquiterpenes were more evenly distributed across the transect. Aphid and ant occurrence was influenced by monoterpenoids. Plants of monoterpenoid class 1 were colonized by aphids and ants significantly more often than expected by chance, whereas in other classes there were no significant differences. Aphid abundance was affected by soil type, and average annual temperature positively correlated with the occurrence of ants. We found significant geographic patterns in the distribution of tansy chemodiversity and show that monoterpenoids affect aphid and ant occurrence, while the soil type can influence aphid abundance. We show that geographic variation in plant chemistry influences insect community assembly on tansy plants.</p>

Data for isolation-by-environment and its consequences for range shifts with global change: Landscape genomics of the invasive common tansy

<p>Invasive species are a growing global economic and ecological problem. However, it is not well understood how environmental factors mediate invasive range expansion. In this study, we investigated the recent and rapid range expansion of common tansy across environmental gradients in Minnesota, U.S.A. We densely sampled individuals across the expanding range and performed reduced representation sequencing to generate a dataset of 3071 polymorphic loci for 176 individuals. The dataset includes additional samples from the native range in Finland that were not used in the downstream analysis but are contributed for completeness. The dataset includes the genotype calls for all individuals sampled and sequenced. The genotype file was generated by stacks2.59 running the denovo pipeline and then using the populations function where we kept loci that were in 70% of populations and had a minor allele frequency of at least 1%. We used non-spatial and spatially-explicit analyses to determine the relative influences of geographic distance and environmental variation on patterns of genomic variation. We found no evidence for isolation-by-distance (IBD) but strong evidence for isolation-by-environment (IBE), indicating that environmental factors may have modulated patterns of range expansion.</p>

Data from: Stay at home aphids: comparative spatial and seasonal metapopulation structure and dynamics of two specialist tansy aphid species studied using microsatellite markers

Two tansy-feeding aphids – Macrosiphoniella tanacetaria (MA) and Metopeurum fuscoviride (ME) – were studied at a small spatial scale in and around Jena (&lt; 80 km2) using polymorphic microsatellite markers. Both species were found in ~ 60% of sites formerly known to harbour the aphids, although generally when they did occur, they occurred singly (MA ~ 50%; ME ~60%) and rarely together on the same plant at the same time (~10%) and then usually only in the early part of the growing season. This difference may be due to quasi-apparent competition effects elicited to ants farming ME aphids, and preferentially actively eliminating or disturbing MA aphids. In terms of population genetics, both aphids showed extreme genetic heterogeneity within a metapopulation structure, ME more than MA, i.e. higher FST values, ~ 0.4 vs. 0.15, respectively, and limited levels of interpopulation gene flow. Subpopulations often deviated from Hardy-Weinberg equilibrium and showed linkage disequilibria, as expected in animals with extended parthenogenetic reproduction, and had positive FIS values for most large samples, suggesting inbreeding, and possibly philopatry, certainly in ME. Hierarchical analysis (allele range and number per locus, analysis of molecular variance and FST) strongly suggested that the plant rather than site governs the level of genetic variation. Bayesian clustering analysis revealed that both species had heterogeneous historical genetic patterning, with K (number of subgroups) ranging from 3-7. Evidence is also provided from isolation by distance (IBD) and private allele analyses, that in MA, the presence of winged autumn males, absent in ME where males are wingless, influences comparative population genetic structuring, such that ME subpopulations are comparatively more inbred and genetically differentiated than MA subpopulations. Lastly, additional spatial arrangement (ALLELES-IN-SPACE) analysis showed that in both species, certain subpopulations were genetically isolated from the remainder, probably due to geographical barriers, including intervening buildings and woods. As such, the biology of these tansy aphids living in semi-natural habitats is very different from many pest aphid species examined within agro-ecosystems and infesting ephemeral crops, since the former seem much more reluctant to fly and hence show contrastingly much higher levels of interpopulation divergence, even at small spatial scales as here investigated. Indeed, the number of genotypic clusters found for tansy aphids found using Bayesian approaches is similar to that for the major pest the peach-potato aphid, Myzus persicae, globally.
