Effects of microarthropod exclusion and water amendments on fluffgrass rhizosphere total soil nitrogen at the Jornada Basin, 1986-1987
This data package contains data on soil nitrogen content in the rhizosphere of fluff grass (Dasyochloa pulchella) plants collected from 1986-1987 at the Jornada Basin LTER site. The purpose of this study was to test how watering, and changing densities of soil microarthropods and nematodes, results in changes in fluff grass rhizosphere total nitrogen. Twenty 6 x 6 m plots were established with a 3 m buffer between plots. Five plots were randomly assigned to one of four treatments: 1) Chlordane to exclude microarthropods, 2) Chlordane and water, 3) Water, and 4) Control. Every three months from June 1986 to January 1987, randomly selected fluff grass plants were harvested from each plot and rhizosphere soil samples were collected for total nitrogen digestion analysis. This data set consists of sampling date, treatment, subsample number, and total nitrogen (mg/kg). This study was completed in January 1987.

Milked colostrum is not initially sterile and may even contain high bacterial counts. Minimizing total bacterial counts in colostrum is generally advised, however bacterial quality of colostrum comprises more than just bacterial quantities, but also depends on the specific bacteria present. While duration and temperature of colostrum storage are known to affect total plate counts (TPC), less is known about the effects of storage on the actual bacterial composition of the TPC. We speculated that, depending on the storage conditions, colostrum is a substrate in which certain bacterial species can thrive affecting the quality of colostrum.</p> <p>We therefore aimed to characterize the effects of different colostrum storage methods on the composition of the viable, aerobic, microbial community. Colostrum samples were stored at different temperatures and for different durations. Next, bacterial growth was assessed using the aerobe plate count culture method, followed by 16S rRNA gene amplicon sequencing. Differences in the TPC bacterial compositions of the stored colostrum samples, as determined by 16S rRNA gene amplicon sequencing, were mostly explained by the variation in bacterial composition of the colostrum sample directly after milking. In line with earlier studies, the results from our study show that the TPC increased when colostrum was stored for 24 hours at room temperature, but not when stored in a refrigerator for the same duration. Community structure of the TPC of colostrum stored at room temperature for 24 hours and stored in a refrigerator for a week was significantly different from the baseline samples. The 16S rRNA sequencing results indicate this is because of increased numbers of <em>Enterobacteriaceae.</em> <em>Enterobacteriaceae</em> abundance in refrigerated samples seemed to remain stable for the first 24 hours, but increased drastically after one week. The results indicate that microbial composition of stored colostrum is mostly influenced by the composition of the colostrum sample directly after milking, which is most probably the result of contamination or other environmental influences during the milking process. This study provides a deeper insight in the changes in the microbial composition of colostrum TPC during practical storage conditions and provides a primer for more detailed research into the determinants of bacterial composition of colostrum and the linked health effects.</p>

The effect of the 2017 solar eclipse in the ionospheric total electron content
<p>Using 15-second RINEX files inside the totality path, I calculated the Total Electron Content (TEC) at each station-satellite pair for each epoch recorded during the day of the 2017 solar eclipse. These calculations result in TEC observations at the ionospheric piercing points (IPP), the intersection of the station-satellite line-of-sight (LOS) and the peak electron density of the ionospheric F layer at a height of ~300 km. Using the estimated TEC at each IPP, I produced a video showing the TEC change during the passage of the umbra and penumbra of the eclipse over North America over a period of one hour. The video shows that as the moon’s shadow passes and the sunlight is blocked, the TEC drops due to the recombination of the ionized particles. Total biomass of two native and one nonnative Eugenia congeners from growth chamber experiment with site, soil microbe, and temperature treatments to test biotic interaction effects of range expansion.
To assess the potential for invasive plants to shift ranges more readily than native plant species, a growth chamber experiment was set up to monitor changes in biomass under different site, temperature, and soil biota treatments. Total biomass (above and belowground) of three Eugenia congeners was assessed by site, soil microbe, and temperature treatments. Of the three congeneric shrub species, two are native to southern and central Florida (Eugenia foetida and E. axillaris,), and one is a non-native invasive from south America (E. uniflora). We measured total biomass for these plant species in growth chambers grown under live and sterile soils from two sites within their current range, and one site in their expected range, simulating current (2010) and predicted future (2050) spring growing season temperatures in the new range. The experiments took place from 2012 - 2013 in University of Central Florida growth chambers.
