Kupffer Cell Plasticity

<p>The study by Huang et al. delineates the phenotypic and functional transformation of Kupffer cells (KCs) within metastatic liver microenvironments, characterized by an apparent reprogramming towards an inflammatory macrophage niche prevalent in liver metastasis-associated macrophages (LMAMs). Standard lineage tracing and proliferation recording systems revealed that LMAMs are primarily derived from circulating monocytes, but paradoxically, monocyte ablation only marginally reduced LMAM numbers. Instead, KC infiltration compensated for the monocyte deficit, with KCs displaying significant phenotypic and functional shifts attributable to both increased local macrophage proliferation and epigenetic remodeling. Notably, selective co-culturing with apoptotic cells demonstrated that KC-derived LMAMs retained some original KC functions despite their reprogramming. The study has profound implications for therapeutic interventions, suggesting that targeting both peripheral monocytes and differentiated macrophages in situ may prove more efficacious in reversing immunosuppressive myelopoiesis within liver metastases.</p>
