GFP and JPH2 fluorescence and WGA staining in JPH2+ ARVMs at day 4 in culture: views across the z-axis.

Supplementary Video 1. Complete views across the Z-axis (from bottom to top) for GFP (left), JPH2 (middle) and WGA (right) channels for two day 4 ventricular myocytes cultured with Adv-JPH2-GFP. The above cell has greater GFP (left) and JPH2 (middle) intensity, indicating more successful viral transduction, but internal WGA staining (right) is little different for both cells. T-tubules are visible in the WGA channel towards the very top and very bottom of each cell. The punctate nature of JPH2 staining in the above cell is readily apparent throughout. Z-depth was 1 µm. The non-cell staining apparent in the WGA channel for deeper Z-slices is staining of the cover slip.
