Disease-driven mass mortality event leads to widespread extirpation and variable recovery potential of a marine predator across the eastern Pacific
<p>The prevalence of disease-driven mass mortality events is increasing, but our understanding of spatial variation in their magnitude, timing, and triggers are often poorly resolved. Here, we use a novel range-wide dataset comprised of 48,810 surveys to quantify how Sea Star Wasting Disease affected <i>Pycnopodia</i> <i>helianthoides</i>, the sunflower sea star, across its range from Baja California, Mexico to the Aleutian Islands, USA. We found that the outbreak occurred more rapidly, killed a greater percentage of the population, and left fewer survivors in the southern half of the species' range. <i>Pycnopodia</i> now appears to be functionally extinct (> 99.2% declines) from Baja California, Mexico to Cape Flattery, Washington, USA and exhibited severe declines (> 87.8%) from the Salish Sea to the Gulf of Alaska. The importance of temperature in predicting <i>Pycnopodia</i> distribution rose 450% after the outbreak, suggesting these latitudinal gradients may stem from an interaction between disease severity and warmer waters. We found no evidence of population recovery in the years since the outbreak. Natural recovery in the southern half of the range is unlikely over the short-term and assisted recovery will likely be required for recovery in the southern half of the range on ecologically-relevant time scales.</p>

Low toxicity crop fungicide (Fenbuconazole) impacts reproductive male quality signals leading to a reduction of mating success in a wild solitary bee
<p>Recent reports on bee health suggest that sub-lethal doses of pesticides have negative effects on wild bee reproduction and ultimately on their population growth.</p> <p>Females of the solitary horned mason bee, Osmia cornuta, evaluate thoracic vibrations and odours of males to assess male quality. When certain criteria are met, the female accepts the male and copulates. However, these signals were found to be modified by sub-lethal doses of pesticides in other hymenopterans. Here, we tested whether sub-lethal doses of a commonly used fungicide (Fenbuconazole) impact male quality signals and mating success in O. cornuta.</p> <p>Males exposed to Fenbuconazole exhibited reduced thoracic vibrations and an altered cuticular hydrocarbon profile compared to the control bees. Moreover, males exposed to the fungicide were less successful in mating than control males.</p> <p>Synthesis and applications: Our results indicate that a low toxicity fungicide can negatively affect male reproductive success by altering behavioural and chemical cues. This could explain the decreasing pollinator populations in a pesticide-polluted environment. This study highlights the need for a more comprehensive approach, including behaviour and chemical cues, when testing new pesticides and a more cautionary approach to the pesticides already used on crops.</p>

Data from: Stochasticity leads to coexistence of generalists and specialists in assembling mutualistic communities
<p>Previous models for assembling ecological networks did not include stochasticity at the level of population dynamics (e.g., demographic noise, environmental noise) and focused mainly on food webs. Here, we present a model for the assembly of mutualistic bipartite networks, such as plant-pollinator networks, and examine the influence of demographic noise on the trajectory of species and strategy diversity, i.e., the range of present strategies from specialism to generalism. We find that assembled communities show at intermediate assembly stages a maximum of species diversity and of average generalization. Our model thus provides a mechanism for non-linear, hump-shaped diversity trajectories at intermediate succession, consistent with the intermediate disturbance hypothesis. Long-term coexistence of specialists and generalists emerges only in the presence of demographic noise and is due to a persistent species turnover. These findings highlight the importance of stochasticity for maintaining long-term diversity.</p>
Intraspecific trait changes in response to drought lead to trait convergence between- but not within species
<p>Drought is expected to increase in future climate scenarios. Although responses to drought of individual functional traits are relatively well-known, simultaneous changes across multiple traits in response to water scarcity remain poorly understood despite its importance to understand alternative strategies to resist drought. We grew 52 herbaceous species in monocultures under drought and control treatments and characterized the functional space using seven measured above- and belowground traits: plant height, leaf area, specific leaf area, leaf dry matter content, specific root length, average root diameter, and root dry matter content. Then, we estimated how each species occupied this space and the amount of functional space occupied in both treatments using trait probability density functions. We also estimated intraspecific trait variability (ITV) for each species as the dissimilarity in trait values between the individuals of each treatment. We then mapped drought resistance and ITV in the functional space using generalized additive models. The response of species to drought strongly depended on their traits, with species that invested more in root tissues and conserved small size being both more resistant to drought and having higher ITV. We also observed a significant trend of trait displacement towards less conservative strategies. However, these changes depended strongly on the trait values of species in the control treatment, with species with different traits having opposing responses to drought. These contrasting responses resulted in lower trait variability in the species pool in drought compared to control conditions. Our results suggest strong trait filtering acting on conservative species as well as the existence of an optimal part in the functional space to which species converge under drought. Our results show that changes in species trait-space occupancy are key to understand plant strategies to withstand drought, highlighting the importance of individual variation in response to environmental changes, and suggest that community-wide functional diversity and biomass productivity could decrease in a drier future. Knowing these shifts will help to anticipate changes in ecosystem functioning facing climate change.</p>

Off-target integron activity leads to rapid plasmid compensatory evolution in response to antibiotic selection pressure
Integrons are mobile genetic elements that have played an important role in the dissemination of antibiotic resistance. As shown previously (Souque et al, 2021), the integron can generate under stress combinatorial variation in resistance cassette expression by cassette re-shuffling, accelerating the evolution of resistance. However, the flexibility of the integron integrase site recognition motif hints at potential off-target effects of the integrase on the rest of the genome that may have important evolutionary consequences. Here we test this hypothesis by selecting for increased piperacillin resistance populations of <em>P.aeruginosa</em> with a mobile integron containing a hard-to-mobilise beta-lactamase cassette to minimize the potential for adaptive cassette re-shuffling. We found that integron activity can both decrease overall survival rate but also improve the fitness of the surviving populations. Off-target inversions mediated by the integron accelerated plasmid adaptation by disrupting costly conjugative genes otherwise mutated in control populations lacking a functional integrase. Plasmids containing integron-mediated inversions were associated with lower plasmid costs and higher stability than plasmids carrying mutations, albeit at a cost of reduced conjugative ability. These findings highlight the potential for integrons to create structural variation that can drive bacterial evolution, and they provide an interesting example showing how antibiotic pressure can drive the loss of conjugative genes.
Projected climatic changes lead to biome changes in areas of previously constant biome
<p class="Indentedmaintext"><b>Aim: </b>Recent studies in southern Africa identified past biome stability as an important predictor of biodiversity. We aimed to assess the extent to which past biome stability predicts present global biodiversity patterns, and the extent to which projected climatic changes may lead to eventual biome changes in areas with constant past biome.</p> <p class="Indentedmaintext"><b>Location: </b>Global.</p> <p class="Indentedmaintext"><b>Taxon: </b>Spermatophyta; terrestrial vertebrates.</p> <p class="Indentedmaintext"><b>Methods: </b>Biome constancy was assessed and mapped using results from 89 dynamic global vegetation model simulations, driven by outputs of palaeoclimate experiments spanning the past 140 ka. We tested the hypothesis that terrestrial vertebrate diversity is predicted by biome constancy. We also simulated potential future vegetation, and hence potential future biome patterns, and quantified and mapped the extent of projected eventual future biome change in areas of past constant biome.</p> <p class="Indentedmaintext"><b>Results: </b>Approximately 11% of global ice-free land had a constant biome since 140 ka. Aside from areas of constant Desert, many areas with constant biome support high species diversity. All terrestrial vertebrate groups show a strong positive relationship between biome constancy and vertebrate diversity in areas of greater diversity, but no relationship in less diverse areas. Climatic change projected by 2100 commits 46–66% of global ice-free land, and 34–52% of areas of past constant biome (excluding areas of constant Desert) to eventual biome change.</p> <p class="Indentedmaintext"><b>Main conclusions: </b>Past biome stability strongly predicts vertebrate diversity in areas of higher diversity. Future climatic changes will lead to biome changes in many areas of past constant biome, with profound implications for biodiversity conservation. Some projected biome changes will result in substantial reductions in biospheric carbon sequestration and other ecosystem services.</p>
Data for PNASnexus article Aldehyde dehydrogenase 3A1 deficiency leads to mitochondrial dysfunction and impacts salivary gland stem cell phenotype
<p>Adult salivary stem/progenitor cells (SSPC) have an intrinsic property to self-renew in order to maintain tissue architecture and homeostasis. Adult salivary glands have been documented to harbor SSPC, which have been shown to play a vital role in the regeneration of the glandular structures post radiation damage. We have previously demonstrated that activation of aldehyde dehydrogenase 3A1 (ALDH3A1) after radiation reduced aldehyde accumulation in SSPC, leading to less apoptosis and improved salivary function. We subsequently found that sustained pharmacological ALDH3A1 activation is critical to enhance regeneration of murine submandibular gland after radiation damage. Further investigation shows that ALDH3A1 function is crucial for SSPC self-renewal and survival even in the absence of radiation stress. Salivary glands from <em>Aldh3a1</em>-null mice have fewer acinar structures than wildtype mice. ALDH3A1 deletion or pharmacological inhibition in SSPC leads to a decrease in mitochondrial DNA copy number, lower expression of mitochondrial specific genes and proteins, structural abnormalities, lower membrane potential, and reduced cellular respiration. Loss or inhibition of ALDH3A1 also elevates ROS levels and accumulation of ALDH3A1 substrate 4-hydroxynonenal (4-HNE, a lipid peroxidation product), leading to decreased survival of murine SSPC that can be rescued by treatment with 4-HNE specific carbonyl scavengers. Our data indicate that ALDH3A1 activity protects mitochondrial function and is important for the regeneration activity of SSPC. This knowledge will help to guide our translational strategy of applying ALDH3A1 activators in the clinic to prevent radiation-related hyposalivation in head and neck cancer patients.</p>

Data from: Climate warming leads to advanced fruit development period of temperate woody species but divergent changes in its length
<p><span>Climate warming has significantly altered the phenology of plants</span><span> in recent decades. However, in contrast to the widely reported warming-induced extension of vegetative growing season, the response of fruit development period (FDP) from flowering to fruiting remains largely unexplored, particularly for woody plants. Analyzing >560,000 <em>in situ</em> observations of both flowering and fruiting dates for six temperate woody species across 2958 European phenological observations sites during </span><span>1980 – 2013</span><span>, we found that in all species both flowering and fruiting phenology, i.e., the FDP, advanced with climate warming. However, the advancing rates of the two events were not necessarily equal for any given species, resulting in divergent changes in the length of FDP among species with climate warming. During </span><span>1980 – 2013</span><span>, not only the temperature during FDP but also the forcing requirement for fruit development increased, both affecting the length of FDP. The shortened FDP was mainly due to elevated temperature, thus accelerating the accumulation of forcing, whereas the prolonged FDP was primarily caused by the substantial increase of the forcing requirement of fruiting, which could be fulfilled only in a longer time and thus slowed down the advance of fruiting. This study provides large-scale empirical evidence of warming-induced </span><span>advances of FDP but divergent changes in its length in temperate woody species. Our findings demonstrate the contrasting reproductive phenological strategies among temperate woody species under the pressure of warming climate, contrary to the lengthening of vegetative growing season, which is by and largely similar with different woody species.</span></p>

<p>Aim: Test for genetic signatures of island biogeographic patterns, using a slowly evolving, recent colonist with a low dispersal capability in an island chain without linear chronosequence; evaluate the multiple-endemic status of the species; and quantify genetic diversity of extant island populations.</p> <p>Location: The Lesser Antilles (Caribbean).</p> <p>Taxon: Magnolia dodecapetala (Magnoliaceae).</p> <p>Methods: Genetic diversity was characterised using Sanger sequencing of 21 individuals amplified for 11 DNA markers, plus microsatellite data of 195 individuals genotyped with 19 simple sequence repeat (SSR) markers. Sanger sequencing data were used to construct a Bayesian phylogenetic hypothesis, while SSR markers were used to run approximate Bayesian computation (ABC) demographic analyses and calculate population statistics.</p> <p>Results: Both types of molecular data support stepwise colonization, decoupled from known island ages. The ABC analyses support a north to south migration while the Sanger sequencing data indicates a mixture of island progression rule and stepping stone dispersal. The SSR data show strong genetic structuring per island and significant inbreeding in all populations except in Saint Lucia. The lowest genetic diversity is found in the population from Saint Vincent. A high amount of genetic linkage occurs in a subpopulation from Dominica.</p> <p>Main conclusions: Biogeographic patterns for the complex geological setting of the Lesser Antilles are uncovered using a slowly evolving study species. All genetic data support treating each island as distinct Management Units for conservation and call for a re-evaluation of the species limits. Inbreeding threatens the survival of island populations and the populations of Saint Vincent and Dominica represent conservation priorities.</p>

Working from home (WFH) has been a popular trend among organizations during the coronavirus (COVID-19) pandemic. The COVID-19 saw a breakthrough in work cultures and environments where working from home was a remarkable success in remote working environments, despite being a rare phenomenon in Sri Lanka. Yet, it is argued that the deployment of work from home has not been effective among Sri Lankan business organizations due to a lack of IT infrastructure, facilities, and knowledge. The purpose of the study is to investigate the impact of cloud computing, embracing the service models (Infrastructure as a Service, Platform as a Service, and Software as a Service) as theoretical lenses and testing the COVID-19 as the moderator. The study has been conducted based on a deductive approach and adopted a stratified random sampling method. The sample consisted of 384 IT employees among those who had experienced working from home. The study utilized multiple regression and found that cloud computing service models significantly impact work from home with the moderating effect of COVID-19.
