BEAT AML 2.0

Acute myeloid leukemia (AML) is a cancer of myeloid-lineage cells with limited therapeutic options. We previously combined ex vivo drug sensitivity with genomic, transcriptomic, and clinical annotations for a large cohort of AML patients, which facilitated discovery of functional genomic correlates. Here, we present a dataset that has been harmonized with our initial report to yield a cumulative cohort of 805 patients (942 specimens). We show strong cross-cohort concordance and identify features of drug response. Further, deconvoluting transcriptomic data shows that drug sensitivity is governed broadly by AML cell differentiation state, sometimes conditionally affecting other correlates of response. Finally, modeling of clinical outcome reveals a single gene, PEAR1, to be among the strongest predictors of patient survival, especially for young patients. Collectively, this report expands a large functional genomic resource, offers avenues for mechanistic exploration and drug development, and reveals tools for predicting outcome in AML. The dataset presented in CCDC only includes pediatric and young adult cases.

Beat AML

The Beat AML program involved 11 academic medical centers who worked collectively to accrue a cohort of ~950 AML patient specimens and 11 pharmaceutical and biotechnology companies, who supplied drugs for testing. These specimens were subjected to whole exome sequencing, RNA-sequencing, and ex vivo drug sensitivity analyses.

BINAURAL BEATS ON PARKINSON DISEASE: A LONGITUDINAL STUDY

File structure description:

Patient data: Excel file with the most relevant patient data.

RAW: Contains the raw data in the appropriate format to be processed in BRAINSTORM. These are the records of all patients (ID 5,14, 16, 23, 30, 33, 33, 35, 38, 39, 40, 64, 65) in the pre-stimulation (PRE) and post-stimulation (POST) condition in each of the sessions (Session 1, 2, 3).

Theta power: This folder contains the Excel files with the results of the relative power of the theta band obtained in Brainstorm, conveniently ordered to later generate the .mat files with the patients to be analyzed. The files are sorted by session (S1, S2, S3) and in turn by condition (PRE, POST).

Matlab scripts: Folder containing the .txt scripts that generate the heads of the figures in the paper.

Matlab Functions: Contains the necessary functions to obtain the heads of the figures in the paper, including the .mat files with the relative powers of the theta band of the 8 patients analyzed. It includes the FieldTrip software, since the scripts call some functions of this program, some of them have been modified for this particular study. FieldTrip is the MATLAB software toolbox for MEG, EEG and iEEG analysis, which is released free of charge as open source software under the GNU general public license. Fieldtrip version 20160222 was used for this study, although the updated version can be downloaded at https://www.fieldtriptoolbox.org/download/.

Beating Versus Arrested Heart Isolated Tricuspid Valve Surgery: Long-term Outcomes

Russo M, Di Mauro M, Saitto G, Lio A, Berretta P, Taramasso M, Scrofani R, Della Corte A, Sponga S, Greco E, Saccocci M, Calafiore A, Bianchi G, Leviner DB, Biondi A, Della Ratta E, Livi U, Sharoni E, Werner P, De Vincentiis C, Di Eusanio M, Kocher A, Antona C, Miraldi F, Troise G, Solinas M, Maisano F, Laufer G, Musumeci F, Andreas M. Beating Versus Arrested Heart Isolated Tricuspid Valve Surgery: Long-term Outcomes. Ann Thorac Surg. 2022 Feb;113(2):585-592. doi: 10.1016/j.athoracsur.2021.03.070. Epub 2021 Apr 5. PMID: 33831392.

Abstract

Background: Isolated tricuspid valve (TV) surgery is a rare procedure generally considered at high risk for perioperative mortality and poor long-term outcomes. Surgical treatment can be performed with either an arrested heart (AH) or beating heart (BH) technique. The aim of this study was to compare the outcomes of isolated tricuspid surgery with 2 different approaches.

Methods: The Surgical-Tricuspid Study is a multicenter international retrospective study enrolling adult patients who underwent isolated TV procedures (n = 406; age 56 ± 16 years; 56% female) at 13 international sites. The AH and BH strategies were performed in 253 and 153 patients, respectively. Propensity score-matched analysis was used to compare groups.

Results: After matching, 129 pairs were obtained and analyzed. The 30-day mortality rate was 6.2% versus 5.0% in the AH and BH groups, respectively (P = .9). The rates of acute renal failure requiring replacement therapy (10% versus 3%; P = .02) and stroke (1.6% versus 0%; P = .08) were numerically higher in the AH group. The 6-year survival rate was 67% ± 6% versus 78% ± 5% in the AH and BH groups, respectively (P = .18), whereas freedom from cardiac death was 75% ± 5% versus 84% ± 4% (P = .21). The 6-year composite cardiac end point of cardiac death and reoperation rate was 60% ± 9% versus 86% ± 5% (P = .024) comparing AH-TV replacement and BH-TV repair groups.

Conclusions: Isolated TV surgery performed with a BH strategy is a safe option and resulted in a trend of increased long-term survival and freedom from reoperation compared with the standard AH technique. Patients undergoing BH valve repair had the best long-term outcome.
