Untargeted LC–MS metabolomics reveals an adverse effect of high-fat diet on hepatic metabolism of Oreochromis niloticus

<p>Hepatic steatosis commonly occurs in intensively farmed tilapia. This disease is harmful to fish growth and health, but knowledge of the metabolic changes in tilapia with fatty liver is limited. In the present study, we compared genetically improved farmed tilapia (GIFT, <em>Oreochromis niloticus</em>) fed a high-fat diet (HFD) with those fed a normal-fat diet (NFD) for 8 weeks using LC-MS-based hepatic metabolomic assays and traditional nutritional assessments. Juvenile GIFT fed a HFD displayed higher fat disposition in the liver than did those fed a NFD. The metabolomic analyses revealed 61 differentially accumulated metabolites between the groups, and these metabolites were involved in 37 signaling pathways. Our results illustrate the development of metabolic disorders related to various hepatic biological processes, including protein metabolism, lipid metabolism, carbohydrate metabolism, and nucleotide metabolism in HFD-fed GIFT. These physiological changes may be related to the lower growth rate of GIFT. Overall, our study reveals the metabolic disorders in GIFT fed HFD, and enhance our knowledge of the mechanism of fatty liver formation in GIFT.</p>

Intracellular sodium elevation reprograms cardiac metabolism: Metabolomics data

<p><span><span><span><span><span><span><span><span><span><span><span>Intracellular Na elevation in the heart is a hallmark of pathologies where both acute and chronic metabolic remodeling occurs. We assessed whether acute (75μM ouabain 100nM blebbistatin) and chronic myocardial Na<sub>i</sub>load (PLM<sup>3SA </sup>mouse) are causally linked to metabolic remodeling and whether the hypertrophied failing heart shares a common Na-mediated metabolic 'fingerprint'. Control (PLM<sup>WT</sup>), transgenic (PLM<sup>3SA</sup>), ouabain treated and hypertrophied Langendorff-perfused mouse hearts were studied by <sup>23</sup>Na, <sup>31</sup>P, <sup>13</sup>C NMR followed by <sup>1</sup>H NMR metabolomic profiling. Elevated Na<sub>i</sub> leads to <i><u>common </u></i>adaptive metabolic alterations preceding energetic impairment: a switch from fatty acid to carbohydrate metabolism and changes in steady-state metabolite concentrations (glycolytic, anaplerotic, Krebs cycle intermediates). Inhibition of mitochondrial Na/Ca exchanger by CGP37157 ameliorated the metabolic changes. <i>In silico</i> modelling indicated altered metabolic fluxes (Krebs cycle, fatty acid, carbohydrate, amino acid metabolism). Prevention of Na<sub>i</sub> overload or inhibition of Na/Ca<sub>mito</sub>may be a new approach to ameliorate metabolic dysregulation in heart failure.</span></span></span></span></span></span></span></span></span></span></span></p>

<p>This&nbsp;dataset&nbsp;accompanies&nbsp;the publication &quot;Abundant metabolite-matrix adducts illuminate the dark metabolome of MALDI-mass-spectrometry imaging datasets&quot;. The dataset&nbsp;includes all files, scripts and results that are included in the associated publication.</p> <p>Spatial metabolomics using mass spectrometry imaging (MSI) is a powerful tool to map hundreds or thousands of metabolites across biological systems. One major challenge is the complexity of the data, which includes signals from experimental artifacts. Formation of adducts (<em>e.g.&nbsp;</em>with Na+or K+) or abundant matrix-cluster, in the case of matrix-assisted laser desorption ionization (MALDI)-MSI, strongly increase peak counts. We developed&nbsp;<em>mass2adduct</em>, a universally applicable tool for adduct abundance estimations in high-mass-resolution spatial metabolomics datasets. Our study illustrates that MALDI-MSI data density is remarkably driven by adduct formation and revealed a major influence of so far unrecognized metabolite-matrix adducts on total peak counts. Metabolomics dataset to identify plasma biomarkers in mice with metabolically heterogeneous melanoma xenografts

<p>Tumor xenografts were generated in mice using BRAF mutant A375 and WM47, NRAS mutant WM3000, and WM3311 (BRAF, NRAS, NF-1 wildtype) human melanoma cells. Xenograft tumor and plasma samples obtained from melanoma-bearing and non-tumor mice were analyzed using the MxP&reg; Quant 500 kit in combination with the inhouse UHPLC-MS/MS-based acylcarnitine assay from Biocrates Life Sciences AG, Innsbruck, Austria. The data were used to characterize the metabolomes of human melanoma cell lines with different genetic background transplanted into mice in order to identify potential new biomarkers and targets for melanoma therapy.</p>
