Dataset related to article "Perioperative corticosteroid treatment impairs tumor-infiltrating dendritic cells in patients with newly diagnosed adult-type diffuse gliomas"

This record contains raw data related to article "Perioperative corticosteroid treatment impairs tumor-infiltrating dendritic cells in patients with newly diagnosed adult-type diffuse gliomas"

Introduction: Adult-type diffuse gliomas are malignant primary brain tumors characterized by very poor prognosis. Dendritic cells (DCs) are key in priming antitumor effector functions in cancer, but their role in gliomas remains poorly understood.

Methods: In this study, we characterized tumor-infiltrating DCs (TIDCs) in adult patients with newly diagnosed diffuse gliomas by using multi-parametric flow cytometry and single-cell RNA sequencing.

Results: We demonstrated that different subsets of DCs are present in the glioma microenvironment, whereas they are absent in cancer-free brain parenchyma. The largest cluster of TIDCs was characterized by a transcriptomic profile suggestive of severe functional impairment. Patients undergoing perioperative corticosteroid treatment showed a significant reduction of conventional DC1s, the DC subset with key functions in antitumor immunity. They also showed phenotypic and transcriptional evidence of a more severe functional impairment of TIDCs.

Discussion: Overall, the results of this study indicate that functionally impaired DCs are recruited in the glioma microenvironment. They are severely affected by dexamethasone administration, suggesting that the detrimental effects of corticosteroids on DCs may represent one of the mechanisms contributing to the already reported negative prognostic impact of steroids on glioma patient survival

Dataset related to article: "Diffuse idiopathic skeletal hyperostosis in subjects with congestive heart failure undergoing cardiac rehabilitation: A decision tree analysis"

We provide the raw data used for the following article:

Ambrosino P, Scrutinio D, De Campi M, Miniero E, Formisano R, Spedicato GA, Iannuzzi GL, Pappone N.
Diffuse idiopathic skeletal hyperostosis in subjects with congestive heart failure undergoing cardiac rehabilitation: A decision tree analysis.
"J Rehabil Med". 2020 Mar 18;52(3):jrm00030. doi: 10.2340/16501977-2658. PMID: 32104899.

Abstract

Objective: To assess the prevalence of diffuse idiopathic skeletal hyperostosis and its relationship with vascular risk factors among patients with congestive heart failure.

Design: Population-based cross-sectional study.

Participants: A total of 584 consecutive patients admitted to a Rehabilitative Cardiology Unit.

Methods: Chi-square Automatic Interaction Detector (CHAID) decision tree analysis was used to build a predictive model.

Results: The mean age (standard deviation) of the study population was 68.1 years (standard deviation 12.3), and 77.7% of the subjects were men. The overall prevalence of diffuse idiopathic skeletal hyperostosis in the cohort was 49.8%. Logistic regression analysis showed that age was a predictor of diffuse idiopathic skeletal hyperostosis (odds ratio: 1.034; 95% confidence interval 1.021-1.047, p < 0.001), with increasing odds ratios for increasing age tertiles. The CHAID prediction model identified 2 age "buckets": < 69 and ≥ 69 years. Patients ≥ 69 years had a diffuse idiopathic skeletal hyperostosis prevalence of 60.1%, compared with 39.2% among those < 69 years. Notably, body mass index was a predictor of diffuse idiopathic skeletal hyperostosis in this younger subset of patients (p = 0.028), with 2 body mass index "buckets", ≤ 23.3 and > 23.3 kg/m2, the latter showing more than twice the prevalence of diffuse idiopathic skeletal hyperostosis (43.2% vs 20%).

Conclusion: Diffuse idiopathic skeletal hyperostosis is extremely frequent among patients with congestive heart failure, with age and body mass index being the strongest predictors.

Muscle Diffusion Tensor Imaginig in Facioscapulohumeral muscular dystrophy

Introduction/Aims

Muscle diffusion tensor imaging (mDTI) has not yet been explored in facioscapulohumeral muscular dystrophy (FSHD). We aimed to assess diffusivity parameters in FSHD subjects compared to healthy controls (HCs), with regard to their ability to precede any fat replacement process in the muscle.

Methods

Fat Fraction (FF), water T2 (wT2), Mean, Radial and Axial Diffusivity (MD, RD, AD) and Fractional Anisotropy (FA) of thigh muscles were calculated in a cohort of ten FSHD subjects and fifteen age-matched HCs. All parameters were compared between FSHD subjects and controls, exploring also their values along the main axis of the muscle. Diffusivity parameters were tested as predictors of disease involvement in muscle compartments with no significant fat substitution and edema. Whole-thigh mDTI values were correlated to clinical severity scores.

Results

FF and wT2 were significantly higher in FSHD than controls whereas MD, RD and AD were significantly lower than controls (p < .05). No difference with controls was shown for FA. FF positively correlated with FA and negatively with MD, RD and AD. FF and FA showed significantly higher values distally than proximally. whereas wT2, MD, RD, AD showed lower values distally than proximally (p<.05). Muscles with no significant fat replacement or edema showed a significantly lower AD and FA than controls. FA was the only parameter to positively correlate with the 6-Minute Walking Test.

Discussion

Our results suggest that mDTI parameters appear to predominantly reflect fat replacement in FSHD and might be able to show disease involvement in muscles even before significant fat replacement.
